    Notes: To investigate the enzymatic properties of Vibrio mimicus metalloprotease, the mature metalloprotease gene (vmc) was overexpressed in Escherichia coli and the recombinant protein (rVMC61) was purified by metal affinity chromatography. rVMC61 showed maximum activity at about 37°C, pH 8. The purified rVMC61 was very specific toward collagen substrates, such as gelatin, type I, II, and III collagens and synthetic peptides (Cbz-GPLGP and Cbz-GPGGPA). But it did not show degrading activity toward other biological proteins including lysozyme, lactoferrin and bovine serum albumin. rVMC61 also showed cytotoxicity against CHSE-214 fish cells. To examine the role of the C-terminal region of rVMC61, the 3′ end of the metalloprotease gene (vmc) was digested serially with exonuclease III. The truncated vmc derivatives encoding 57–42 kDa of the protease were isolated and overexpressed in E. coli. The collagenase activities of truncated proteins were investigated using gelatin as substrate. Deletion of 100 amino acids from the C-terminus resulted in loss of gelatin degrading activity. However, deletion of 67 amino acids from the C-terminus did not affect its gelatin degrading activity.

    Notes: Oligopeptides play important roles in bacterial nutrition and signaling. The oligopeptide permease (opp) gene cluster was cloned from Vibrio fluvialis. The V. fluvialis opp operon encodes five proteins: OppA, B, C, D and F. The deduced amino acid sequence of these proteins showed high similarity with those from other Gram-negative bacteria. To investigate whether OppA is involved in biofilm production, an oppA knockout mutant was constructed by homologous recombination. The oppA mutant produced more abundant biofilm than the wild type in BHI medium. When both strains were grown in minimal medium, we could not detect biofilm formation. However, it was found that the biofilm productivity of the oppA mutant was two folds greater than that of the wild type in minimal medium containing peptone or tryptone. This variation in biofilm production was demonstrated by scanning electron microscopy (SEM). In minimal medium containing C-sources, both strains produced some biofilm without significant difference in the biofilm productivity. Complementation of oppA gene with the plasmid pOAC2, which contains oppA ORF plus promoter regions, was sufficient to restore growth rate and biofilm to the wild type. These results suggest that the OppA protein is involved in uptake of peptides and affects biofilm productivity.

    Notes: Crosslinkable fluorinated poly(arylene ethers) (FPAE-Fn-PEP) with high transparency and high thermal stability have been investigated for low-loss optical waveguide materials. FPAE-Fn-PEP bearing phenyl ethynyl moiety at the polymer chain end were synthesized by the reaction of 4,4′-(hexafluoro-isopropylidene)diphenol with an excess decafluorobiphenyl, followed by the reaction of 4-phenyl ethynyl phenol. The Mns and Mw/Mns of the polymers determined by GPC with polystyrene standard were in the range of 6200 to 19,400 and 1.4 to 2.04, respectively. The resulting polymers were thermally crosslinked at 320°C for 2 h. The cured polymers show good chemical resistance and high thermal stability up to 510°C under nitrogen. The refractive indices of their films were controlled between 1.495 and 1.530 at 1.55 μm by adjusting molecular weight. A single-mode channel waveguide made of FPAE-F20-PEP was fabricated by conventional photolithography and O2 reactive ion etching (RIE). The propagation loss of the channel waveguide was measured and found to be less than 0.2 dB/cm at 1.55 μm. © 1998 John Wiley & Sons, Inc. J Polym Sci A: Polym Chem 36: 2881-2887, 1998

    Notes: Aromatic polyimides with side chain nonlinear optical chromophores have been investigated through a facile two-step synthetic route. First, various poly(hydroxy imide)s have been synthesized by direct thermal imidization of diaminophenol dihydrochloride salt and aromatic dianhydride monomers. The resulting polyimides bearing phenolic hydroxy groups were found to react easily with the terminal hydroxy group on the chromophores via the Mitsunobu condensation to give corresponding polyimides with high optical nonlinearities and good solubility in common organic solvents. Detailed physical properties showed that these polyimides have a molecular weight (Mw) of 31,000 and high glass transition temperature above 220°C, ensuring a long-term alignment stability at elevated temperature. The electrooptic coefficients, r33, of the electrically poled polymer films were in the range 1.8-7.6 pm/V at 1.3 μm. © 1998 John Wiley & Sons, Inc. J Polym Sci A: Polym Chem 36: 301-307, 1998
