    Notes: α-Synuclein co-exists with lipids in the Lewy bodies, a pathological hallmark of Parkinson's disease. Molecular interaction between α-synuclein and lipids has been examined by observing lipid-induced protein self-oligomerization in the presence of a chemical coupling reagent of N-(ethoxycarbonyl)-2-ethoxy-1,2-dihydroquinoline. Lipids such as phosphatidic acid, phosphatidylinositol, phosphatidylserine, phosphatidylethanolamine, and even arachidonic acid induced the self-oligomerization whereas phosphatidylcholine did not affect the protein. Because the oligomerizations occurred from critical micelle concentrations of the lipids, the self interaction of α-synuclein was shown to be a lipid-surface dependent phenomenon with head group specificity. By employing β-synuclein and a C-terminally truncated α-synuclein (α-syn97), the head-group dependent self-oligomerization was demonstrated to occur preferentially at the N-terminal region while the fatty acid interaction leading to the protein self-association required the presence of the acidic C-terminus of α-synuclein. In the presence of Cu2+ and H2O2, phosphatidylinositol (PI), along with other acidic lipids, actually enhanced the metal-catalyzed oxidative self-oligomerization of α-synuclein. The dityrosine crosslink formation responsible for the PI-enhanced covalent self-oligomerization was more sensitive to variation of copper concentrations than that of H2O2 during the metal-catalyzed oxidation. The enhancement by PI was shown to be due to facilitation of copper localization to the protein because actual binding affinity between copper and α-synuclein increased from Kd of 44.7 μm to 5.9 μm in the presence of the lipid. Taken together, PI not only affects α-synuclein to be more self-interactive by providing the lipid surface, but also enhances the metal-catalyzed oxidative protein self-oligomerization by facilitating copper localization to the protein when the metal and H2O2 are provided. This observation therefore could be implicated in the formation of Lewy bodies as lipids and metal-catalyzed oxidative stress have been considered to be a part of pathological causes leading to the neurodegeneration.

    Notes: Pb0.98(La1−xLix)0.02(Zr0.55Ti0.45)O3(PLLZT with 0.1 ≤x≤ 0.7) thin films were sol-gel-grown on Pt(111)/Ti/SiO2/Si substrates, employing a thin lead zirconate titanate (PZT) template layer. Films annealed at 〉550°C showed a highly (111)-oriented preferential growth. Typical values of the switchable remanent polarization (2Pr) and the coercive field (Ec) of the PLLZT/PZT/Pt film capacitor for x= 0.3 were 50 μC/cm2 and 39 kV/cm, respectively, at 5 V. All the PLLZT/PZT/Pt capacitors (for 0.1 ≤x≤ 0.7) exhibited fatigue-free behavior up to 6.5 × 1010 switching cycles, a quite stable charge retention profile with time, and high 2Prvalues, all which assure their suitability for nonvolatile ferroelectric memories.

    Notes: α-Synuclein, a pathological component of Parkinson's disease by constituting the Lewy bodies, has been suggested to be involved in membrane biogenesis via induction of amphipathic α-helices. Since the amphipathic α-helix is also known as a recognition signal of calmodulin for its target proteins, molecular interaction between α-synuclein and calmodulin has been investigated. By employing a chemical coupling reagent of N-(ethoxycarbonyl)-2-ethoxy-1,2-dihydroquinoline, α-synuclein has been shown to yield a heterodimeric 1 : 1 complex with calmodulin on sodium dodecyl sulfate–polyacrylamide gel electrophoresis in the presence and even absence of calcium, whereas β-synuclein was more dependent upon calcium for its calmodulin interaction. The selective calmodulin interaction of α-synuclein in the absence of calcium was also demonstrated with the aggregation kinetics of the synucleins in which only the α-synuclein aggregation was affected by calmodulin. A reversible binding assay confirmed that α-synuclein interacted with the Ca2+-free as well as the Ca2+-bound calmodulins with almost identical Kds of 0.35 µm and 0.31 µm, respectively, while β-synuclein preferentially recognized the Ca2+-bound form with a Kd of 0.68 µm. By using a C-terminally truncated α-synuclein of α-syn97, the calmodulin binding site(s) on α-synuclein was(were) shown to be located on the N-terminal region where the amphipathic α-helices have been suggested to be induced upon membrane interaction. By employing liposome and calmodulin in a state of being either soluble or immobilized on agarose, actual competition of α-synuclein between membranes and calmodulin was demonstrated with the observation that α-synuclein previously bound to the liposome was released upon specific interaction with the calmodulins. Taken together, these data may suggest that α-synuclein could act not only as a negative regulator for calmodulin in the presence and even absence of calcium, but it could also exert its activity at the interface between calmodulin and membranes.

    Notes: Endoplasmic reticulum (ER) stress has increasingly come into focus as a factor contributing to neuronal injury. Although caspase-dependent mechanisms have been implicated in ER stress, the signaling pathways involved remain unclear. In this study, we examined the role of the extracellular signal-regulated kinase (ERK), a mitogen-activated protein (MAP) kinase pathway that is highly conserved in many systems for balancing cell survival and death. Prolonged treatment of the human neuroblastoma cell line SH-SY5Y with thapsigargin, an inducer of ER stress, increased cell death over 24–48 h, as measured by LDH release. Caspases were involved; increased levels of active caspase-3 and cleaved caspase substrate PARP were detected, and treatment with Z-VAD-FMK reduced thapsigargin-induced cytotoxicity. In contrast, inhibition of calpain was not protective, although calpain was activated following thapsigargin treatment. An early and transient phosphorylation of ERK1/2 occurred after thapsigargin-induced ER stress, and targeting this pathway with the MEK inhibitors U0126 or PD98059 significantly reduced cell death. Similar cytoprotection was obtained against brefeldin A, another ER stress agent. However, protection against ER stress via ERK inhibition was not accompanied by amelioration of caspase-3 activation, PARP cleavage, or DNA laddering. These data indicate that ERK may contribute to non-caspase-dependent pathways of injury after ER stress.

    Notes: Self-construal is thought to mediate and explain the effects of culture on a wide variety of outcome variables. A meta-analysis of published cross-cultural self-construal research is reported in this article, and the results across studies suggests that the evidence for the predicted cultural differences is weak, inconsistent, or nonexistent. The results of 3 priming experiments (N = 121, N = 99, and N = 361) suggest that (a) priming does not account for the inconsistent results observed in the meta-analysis, (b) that scores on a self-construal scale appear to be measuring trait-like constructs that are not sensitive to priming, and (c) that measures of self-construals lack convergent validity. The results of several measurement studies (N = 121, 223, 230, 323, 214, 206, 126, 204, 148, 141, and 150) were inconsistent with the a priori two-factor measurement model in every case. Self-construal scales were found to be radically multidimensional and highly unstable within and across cultures. These results lead us to conclude that catastrophic validity problems exist in research involving the use of self-construal scales in cross-cultural research.

    Notes: A large number of previous studies have used self-construal to predict communication outcomes. Recent evidence, however, suggests that validity problems may exist in self-construal measurement. The current study conducted a multimethod multitrait (Campbell & Fiske, 1959) validation study of self-construal measures with data (total N= 578) collected in Korea (N= 200), Japan (N= 212), and the U.S. (N= 166). The data showed that the Singelis (1994) Self-Construal Scale, the Cross, Bacon, and Morris (2000) Relational Interdependent Self-Construal Scale (RISC), and the Kuhn and McPartland (1954) Twenty Statements Test (TST) lacked convergent and discriminant validity, both pan-culturally and within each of the three countries included in the study. Scores on the TST were not significantly related to scores on the self-construal scales, and the various self-construal measures correlated more highly with measures of communication directness than with alternative measures of the same type of self-construal. Substantial method effects were also observed. The results were tested for both 2- and 3-dimensional models of self-construal and for refined scales and scales with all items retained. The results of all analyses were inconsistent with the claim that self-construal measures are construct valid.

    Notes: Effects of the orientation of ferroelectric domains on the characteristics of polarized Raman spectra were studied using an epitaxially grown PbTiO3 thin film in which the c-axis oriented domains coexist with the a-axis oriented domains on MgO (001). To obtain polarized spectra for both two distinctive c and a domains, we have employed scattering configurations in which the relevant phonon wave vector, k, is perpendicular to the c axis of the tetragonal unit cell. Compared with the mode frequencies of single-crystal PbTiO3, a softening of the E(TO) phonons was evident for both c and a domains, suggesting the presence of a strong tensile film stress. In addition to this, we observed a splitting of the degenerate "silent" mode into two distinctive B1 and E modes in an epitaxially grown film on MgO (001). © 2002 American Institute of Physics.
