    Notes: The human meniscus is subject to injury that necessitates repair or removal. Many aspects of the cellular response to injury have not been well characterized. The purpose of this study was to describe the cellular distributions within the torn human meniscus. In addition to evaluating the cell density in selected regions, we investigated the cellular expression of a contractile actin isoform that has recently been found in the intact human meniscus. Included as a contemporaneous comparative group were torn human meniscal allografts. We hypothesized that a hypercellular surface zone would be found in the torn menisci, with a higher percentage of cells in this peripheral region expressing α-smooth muscle actin compared with other locations in the interior of the remnant. The rationale for this hypothesis was based on prior immunohistochemical investigations of the distribution of α-smooth muscle actin–containing cells in the torn human anterior cruciate ligament. Eighteen torn meniscal specimens were obtained from 17 patients, 0.5 to 84 months after injury, and four torn allograft meniscal samples were retrieved from three patients, 11 to 49 months after implantation. Microtomed sections of paraffin-embedded tissue were stained with hematoxylin and eosin and a monoclonal antibody to α-smooth muscle actin. The cell density and percentage of cells containing α-smooth muscle actin were determined in the following zones: synovial, vascular, hypercellular with loose collagen, hypocellular with dense collagen, and organized collagen. A cellular layer that resembled synovium was present on the surface of all but two of the specimens. Vascular regions were often continuous with the synovium abutting the more interior loose collagen zones. The total cell density was greatest in each of the zones closest to the periphery (synovium, vascular, and loose collagen; p 〈 0.001), when compared to the interior of the tissue. The synovium-like layer was found to have the highest percentage of α-smooth muscle actin–expressing cells and the highest α-smooth muscle actin–containing cell density (p 〈 0.05). Similar results were found for the torn allograft menisci. These findings confirm the working hypothesis and suggest that the torn human meniscus is capable of mounting a reparative response, including the proliferation of cells capable of contributing to wound closure. This underscores the importance of providing a bridging scaffold into which such cells can migrate. (WOUND REP REG 2002;10:259–266)

    Notes: When 4 mg of testosterone (T) per kg food was given to 1-year-old protandrous male black porgy Acanthopagrus schlegeli for 7 months, gonadosomatic index was significantly higher than when the dose was 0.5 mg kg−1 food. Both doses of T prolonged the spawning season, and increased the number of spermiating fish and milt volume. Sperm concentrations were similar in spermiating black porgy from the treated and control groups. Low levels of oestradiol-17β were observed during the experimental period while elevated levels of plasma T were observed only in March in both control and T-treated groups. Significantly higher levels of plasma 11-ketotestosterone (11-KT) were observed in the 0.5- and 4.0-mg T-treated groups during the spawning season as compared to the control group. The present data suggest that both 0.5- and 4·0-mg T doses stimulate testicular weight, increase numbers of spermiating males and milt volume without affecting the sperm concentrations. Plasma 11-KT concentrations were elevated during T treatment and closely correlated with testicular development and spermiation.

    Notes: Summary The nucleotide sequences of the RNAs 1, 2, and 3 of the cucumber mosaic virus (CMV) Taiwan isolate NT9 were determined and compared at both the nucleotide and amino acid levels with those of CMV strains Fny, Y, O from subgroup I and strain Q from subgroup II. NT9-CMV has an unique feature at the C-terminus of the 3a protein which contains four extra-amino acids. All three RNAs and their encoded proteins, except 2b, of NT9-CMV share more than 90% identity with those of strains in subgroup I, and 72%–85% identity with Q-CMV. The results indicated the conservation of sequences of CMV derived from different geographical locations.

    Notes: Abstract A striped-leaf mutant has been identified in a Taiwanese maize line, Tainan White. The striped area is pale green at the seedling stage, and turns yellow and white at maturity, extending from the leaf blade to the sheath and the internode below the sheath. The mutant does not breed true and following self-pollination, produces three different types of progeny: green, striped and completely affected. The green type permanently loses the mutant character, while the completely affected type is lethal, without any green tissue. Only the striped type transmits the mutant phenotype to progeny, and it has a clonal distribution on the ear. Its phenotype is inherited maternally and its expression is unaffected by the paternal genotype. It cannot be inherited through the paternal parent. This pattern of inheritance, which persists after three cycles of backcrossing with the affected types as the recurrent pistillate parent, suggests that the responsible gene is located in the cytoplamic genome.

    Notes: Abstract  Four DNA fragments were amplified specifically from the B chromosome by PCR using random 10-base oligonucleotides as primers. The location of the fragments in the B chromosome was determined based on whether or not they were amplified from the hypo- ploid DNA generated by four B-A translocations, three of which break in the proximal euchromatic region and the fourth in the distal one-third of the heterochromatic region on the B long arm. Since the hypoploid DNA carries the portion of the B chromosome distal to the breakpoint of a translocation, the presence of a fragment in the hypoploid DNA, but not in the control (which is devoid of any B chromatin), indicates that the fragments is located in the B region distal to the breakpoint in the B long arm. Two fragments were mapped to the euchromatic region and two others to either the distal portion of the euchromatic region or the proximal two-thirds of the heterochromatic region. These fragments in turn mapped three B-A translocations whose breakpoints were located in the euchromatic region.

    Notes: Summary We have used denaturing gradient gel electrophoresis (DGGE) to identify genomic DNA polymorphism in maize (Zea mays L.). DGGE probes detect polymorphism in maize at a frequency comparable to the incidence of restriction fragment length polymorphism (RFLP). Probes identifying polymorphism were mapped to maize chromosome arms by utilizing DGGE and maize lines carrying B-A chromosomal translocations. The methods for library construction, probe screening, and genome analysis, described here for maize, can also be applied to the genomic analysis of other organisms.

    Notes: Abstract Terminal deficiencies (TDs) generated by the r-XI deletion system in maize were used to physically map RFLP markers on the short arm of chromosome 2 (2S) and the long arm of chromosome 6 (6L), chromosome 8 (8L), and chromosome 10 (10L). Five TDs on 2S, 8 on 6L, 10 on 8L, and 20 on 10L were isolated using the recessive morphological markers lg1, py1, j1(gl18), and sr2, respectively, for selection. Two exceptional TDs on 2S and 8L also have a second breakpoint on the long arm of chromosome 2 (2L) and 8L, respectively. The physical mapping of RFLP probes in relation to TD breakpoints was done by Southern hybridization. The five TDs on 2S divide chromosome 2 into four regions, all of which are distinguishable by RFLP markers. Likewise, three remaining chromosome arms are divided by TDs into RFLP-marked regions: 8 TDs divide 6L into five regions, 10 TDs divided 8L into seven regions, and 20 TDs divide 10L into three regions. The linear order of the physical map of 6L and 8L is consistent with that of the genetic maps, but that of 2L and 10L is not. Four groups of markers on 2S as well as 2L, and two on 10L are in reverse order in the physical map compared with the genetic maps. Other intriguing results are that breakpoints of TDs on 6L and 8L are distributed throughout the selected region, but most of those on 2L and 10L cluster in a region near the centromere; a single TD arose after fertilization.
