    Notes: Protein phosphatase 5 (PP5) is a 58-kDa novel phosphoseryl/phosphothreonyl protein phosphatase. It is ubiquitously expressed in all mammalian tissues examined, with a high level in the brain, but little is known about its physiological substrates. We found that this phosphatase dephosphorylated recombinant tau phosphorylated with cAMP-dependent protein kinase and glycogen synthase kinase-3β, as well as abnormally hyperphosphorylated tau isolated from brains of patients with Alzheimer's disease. The specific activity of PP5 toward tau was comparable to those reported with other protein substrates examined to date. The PP5 activity toward tau was stimulated by arachidonic acid by 30- to 45-fold. Immunostaining demonstrated that PP5 was primarily cytoplasmic in PC12 cells and in neurons of postmortem human brain tissue. A small pool of PP5 associated with microtubules. Expression of active PP5 in PC12 cells resulted in reduced phosphorylation of tau, suggesting that PP5 can also dephosphorylate tau in cells. These results suggest that PP5 plays a role in the dephosphorylation of tau and might be involved in the molecular pathogenesis of Alzheimer's disease.

    Notes: The switching of health insurance plans and health care utilization are potentially correlated with both observable and unobservable information. This paper presents a two-period model of health care utilization, and attempts to account for unobserved heterogeneity that simultaneously affects utilization and the decision to switch plans. Data used in this paper are drawn from the Medical Expenditure Panel Survey. Results indicate that non-HMO enrollees increase their utilization of non-emergency related care prior to switching to HMOs, and they decrease utilization after switching. Conversely, individuals enrolled in HMOs report lower levels of utilization before and higher utilization after they switch to non-HMOs.

    Notes: Protein glycosylation modifies the processing of several key proteins involved in the molecular pathogenesis of Alzheimer's disease (AD). Aberrant glycosylation of tau and down-regulation of sialyltransferase in AD brain suggest a possible dysregulation of protein glycosylation that may play a role in AD. We therefore isolated major glycoproteins from AD brain by using lectin-affinity chromatographies and ion-exchange chromatography and further separated them using SDS-polyacylamide gel electrophoresis. Mass spectrometry analysis of 11 isolated glycoproteins led to their identification as: neuronal cell adhesion molecule, β-globin, IgM heavy chain VH1 region precursor, contactin precursor, dipeptidylpeptidase VI, CD81 partner 3, prenylcysteine lyase, adipocyte plasma-associated protein, acid ceramidase and two novel proteins. We found that the level and activity of acid ceramidase (AC), one of the major identified human brain glycoproteins, were significantly elevated in AD brain. Immunohistochemical staining indicated that AC was located mainly in the cell bodies of neurons and colocalized with neurofibrillary tangles. Our findings suggest that AC might play a role in controlling neuronal apoptosis and that AC-mediated signalling pathways might be involved in the molecular mechanism of AD.

    Notes: Abnormal hyperphosphorylation of tau is believed to lead to neurofibrillary degeneration in Alzheimer's disease (AD) and other tauopathies. Recent studies have shown that protein phosphatases (PPs) PP1, PP2A, PP2B and PP5 dephosphorylate tau in vitro, but the exact role of each of these phosphatases in the regulation of site-specific phosphorylation of tau in the human brain was unknown. Hence, we investigated the contributions of these PPs to the regulation of tau phosphorylation quantitatively. We found that these four phosphatases all dephosphorylated tau at Ser199, Ser202, Thr205, Thr212, Ser214, Ser235, Ser262, Ser396, Ser404 and Ser409, but with different efficiencies toward different sites. The Km values of tau dephosphorylation catalysed by PP1, PP2A and PP5 were 8–12 µm, similar to the intraneuronal tau concentration of human brain, whereas the Km of PP2B was fivefold higher. PP2A, PP1, PP5 and PP2B accounted for ∼ 71%, ∼ 11%, ∼ 10% and ∼ 7%, respectively, of the total tau phosphatase activity of human brain. The total phosphatase activity and the activities of PP2A and PP5 toward tau were significantly decreased, whereas that of PP2B was increased in AD brain. PP2A activity negatively correlated to the level of tau phosphorylation at the most phosphorylation sites in human brains. Our findings indicate that PP2A is the major tau phosphatase that regulates its phosphorylation at multiple sites in human brain. The abnormal hyperphosphorylation of tau is partially due to a downregulation of PP2A activity in AD brain.

    Notes: The nano-CeO2 was synthesized by two-step solid-phase reaction. The image of TEMshowed that nano-CeO2 with an average size of about 70 nm. The series of polyaniline/nano-CeO2composites with different PANi: CeO2 ratios were prepared by in-situ polymerization in the presenceof hydrochloric acid (HCl) as dopant by adding nano-CeO2 into the polymerization reaction mixtureof aniline. The composites obtained were characterized by FT-IR and UV-vis spectroscopy analysis.The FT-IR spectra of nanocomposites indicate different blue-shifts, attributed to C–N stretchingmode for benzenoid unit. The UV-vis spectra of nanocomposites display einstein-shifts comparedwith PANi at 620nm. The conductivity properties of the composites are also changed compare to thepure PANi. These results suggest that the interactions between the polymer matrix and nanoparticlestake place in polyaniline/nano- CeO2 composites

    Notes: Iron oxide nanometer powders were prepared by using sol-gel method and the supercriticalfluid drying technology (SCFD) from inorganic salts. The characteristics of the iron oxide nanometerpowders were characterized systematically. The results showed that the drying and crystallization of theproduction could be achieved in one step by SCFD method; furthermore, a highly dispersed, sizeuniformed and with better crystallization magnetite (Fe3O4) nanoparticles can be obtained. Phasetransformation of the as-synthesized powders occurs at about 80°C, and only hematite (α-Fe2O3) can beobtained when being heated at temperatures higher than 400°C. The production synthesized by SCFDexhibits significant magnetic properties. As the heat-treated temperature increase, the magneticproperties of the sample decrease gradually

    Notes: Abstract The effects of all-trans retinoic acid (ATRA) and epidermal growth factor (EGF) on the expression of nm23-H1, a metastasis suppressor gene, were studied in a human 7721 hepatocarcinoma cell line. It was discovered that the expression of nm23-H1 mRNA was up-regulated by ATRA. This was compatible with the observation that the metastasis-associated phenotypes, such as chemotaxic cell migration and invasion, were both reduced in the ATRA-treated and nm23-H1-cDNA-transferred 7721 cells. However, ability of cells to adhere to fibronectin and laminin was not altered identically in the ATRA-treated and nm23-H1-cDNA-transfected 7721 cells. In contrast, the expression of nm23-H1 mRNA in 7721 cells was down-regulated both by the treatment with EGF and by the transfection of c-erbB-2/neu cDNA, which codes a protein homologous to the EGF receptor. EGF is a compound with biological effects opposite to those of ATRA, and c-erbB-2/neu is known to be a metastasis-promoting gene. These results reveal that the metastasis-preventing effect of ATRA may partly result from the up-regulation of nm23-H1, and the metastasis-promoting effects of EGF and c-erbB-2/neu were probably mediated in part by the down-regulation of nm23-H1.

    Notes: Abstract Purpose: Both platelet-derived endothelial cell growth factor (PD-ECGF) and vascular endothelial growth factor (VEGF) are known to promote the development of new blood vessels, which are fundamental to tumor growth and metastasis. We aimed at evaluating the gene expression of PD-ECGF and VEGF in hepatocellular carcinoma (HCC) and portal vein tumor thrombus (PVTT). Patients and methods: Surgical specimens (28 HCC, 28 nontumorous liver tissues and 18 PVTT) were studied by Northern blot analysis. The levels of PD-ECGF mRNA and VEGF mRNA expression were measured by densitometric scanning of the autoradiographs, and they were normalized to the level of expression of an internal control (glyceraldehyde-phosphate dehydrogenase) mRNA. Results: The expression rates of PD-ECGF mRNA in PVTT, HCC and nontumorous liver tissues were 77.8% (14/18), 67.9% (19/28) and 35.7% (10/28), being 88.9% (16/18), 75.0% (21/28) and 17.9% (5/28) respectively for VEGF mRNA. The expressions of PD-ECGF mRNA and VEGF mRNA were higher in HCC with PVTT than when PVTT was absent (P 〈 0.05). The PVTT was more often seen in patients with positive expression of both PD-ECGF mRNA and VEGF mRNA in HCC than in patients who were positive for only one of these factors or negative for both (P 〈 0.05). Conclusion: Both PD-ECGF and VEGF correlated well with the formation of PVTT of HCC.
