    Notes: Patterns for free and protein amino acids and related substances were determined for Tetrahymena limacis and 7 strains of T. pyriformis from axenic stationary phase cultures grown at 25° C by means of 2-dimensional chromatography in a butanol-acetic acid and phenol solvent system with ninhydrin and other polychromatic indicators. A uniform protein amino acid (PAA) pattern was observed in all strains. There were 14 color spots indicating 19 amino acids (including cysteic acid), identified as follows: alanine, arginine, aspartic acid, cysteine/cystine, cysteic acid, glutamic acid, glycine, leucine/isoleucine/phenylalanine, lysine/histidine, proline, serine, threonine, tyrosine, valine/methionine.The following free amino acids and related substances (FAAs) were identified with 14 spots (several different from these for PAA patterns) found in all strains: alanine, arginine, asparagine, aspartic acid, glutamic acid, glutamine, glycine, leucine/isoleucine/phenylalanine, lysine histidine. proline, serine, threonine, tyrosine, valine/methionine. T. limacis and strains LI and Gf-J of T. pyriformis exhibited these only. Chromatograms of 5 strains of T. pyriformis (PR. F. L3 WH52, HS), however, also contained 1 to 4 spots representing certain of the following substances: Cysteic acid, cysteine/cystine, taurine, and the unknowns X1, X2, and X3, having Rf's of 0.33, 0.79, and 0.72 respectively in 4:1 phenol-H2O system Excepting for F and L3, which were similar, the T. pyriformis strains showed quite different distributional patterns of these substances at 25°C. Other deviations in the distribution of the 6 compounds were noted in the chromatograms of 10° and 35° cultures of WH52 and HS. These findings on FAAs and PAAs are tabulated, along with those of previous investigators, to furnish comparisons on 13 strains of Tetrahymena pyriformis, T. limacis and 9 other species of protozoa.

    Notes: SYNOPSIS. Studies cf 3 conjugating strains of Tetrahymena pyriformis grown in a bacterized medium indicated the presence of a system of alternative immobilizing surface antigens. Each strain was found to have the potential for the expression of 3 serologically unrelated antigens. These consisted of a “high temperature” antigen (expressed in the range of 20–35d̀C), a “low temperature” antigen (exhibited in cultures at 10d̀), and an antigen induced by growth in the presence of the above-mentioned “high temperature” antiserum. Normally, by the immobilization reaction, only one of these antigens could be detected as present on any one organism at any given time. The “high temperature” antigens of two of the strains were serclogically related while that of the 3rd strain did not cross-react with antisera to the other two. The “low temperature” antigens of the 3 strains were serologically related as were the antigens induced by growth in antiserum.Studies of inbred hybrids of two of the strains indicated a potential for still more alternative antigens as well as for the spontaneous expression of the antigen which in the parental strains could only be induced to appear by the presence of the specific antiserum.Study of the 3 strains in axenic medium indicated the existence of a more complex system and the possibility that two or more immobilizing antigens may be present simultaneously.

Unabsorbed, complement-inactivated antisera produced in rabbits were used in an immobilization system to study the serology of 31 strains of Tetrahymena pyriformis grown in axenic cultures at 26°C. Fourteen serological “groups” were found: 1. “H”: H, E, S, GC, L3, W, T, GHH, G1-R and sublines W-P and T-P; 2. “PR”: PR; 3. “GL”: GL; 4. “LR”: LR; 5. “L1”: L1 and L2; 6. “GP”: GP and Ch-S; 7. “WH”: WH6, WH14 and WH52; 8. “N”: N and HS; 9. “Y”: Y and TC; 10. “AA”: AA1, AA2 and AA4; 11. “F”: F and BF; 12. “Gf-J”: Gf-J; 13. “EZ”: EZ; 14. “Lava”: Lava. Not all of these “groups” were completely distinct; weak or variable cross-reactions occurring for particular cultures and antisera during the course of several years of observation suggested a relationship in terms of serotype potentialities, for strains in groups 1 and 2, and in groups 2, 3, 4, 5, and 6. Non-reciprocal cross-reactions are tentatively explained in terms of population fluctuations of serotype within particular cultures; several other suggested explanations remain possible. On the whole, however, the strains remained remarkably stable in dominant serotype.Observations on Tetrahymena in paralyzing antisera, with respect to the extrusion of a gelatinous exudate, in general confirmed previous observations by others. “Chain” formation was noted upon recovery, under conditions suggesting that the chains do not always derive entirely from incomplete cytokinesis of dividing organisms but that at least sometimes an aggregation of separate individuals is also involved. Synchronized division was observed for recovering organisms. A gelatinous material was obtained by centrifugation of a large number of individuals; this material provided a degree of protection to the organisms against antiserum effects while other colloidal materials did not, but the protective effects proved nonspecific in the sense that they were not confined to particular strain-antiserum combinations.

    Notes: A survey of antigenic types, as revealed by immobilization reactions, among 78 strains of variety 1 of T. pyriformis grown in monoxenic cultures at 25°C discloses a minimum of six distinct classes. These are called the H serotypes, and are designated as types Ha, Hb, Hc, Hd, He and Hf. The terminal members of highly inbred series (Families A, A1, B, D, etc.) are of a single immunological class, but different inbred series show different serotypes. Early representatives of the inbred series show in contrast considerable serotypic variation. The distribution of serotypes is generally consistent with the genetic hypothesis developed in a parallel study, but new serotypes arise occasionally through some mechanism not yet adequately studied.

Meridian ranges and contractile vacuole pore patterns were determined for 36 strains of Tetrahymena pyriformis, including most of the “classic” amicronucleate and micronucleate strains used by many investigators, plus a dozen other strains not previously characterized with respect to these two morphological features. Uniform axenic culturing and staining techniques were employed.Meridian ranges and modes are analyzed with strain-by-strain comparisons to the reports of previous investigators. The overall inclusive range of the 36 strains was 15–25 meridians. Modes for individual strains varied from 16 to 23. Broader ranges and, especially, higher modes and higher limits are recorded than those reported by Corliss (1953) for strains grown in bacterized hay cultures. Two postoral meridians are characteristic, although a single individual having only one and a few having three postorals were observed. Three postorals were found only in those strains having the higher number of total meridians. A few individuals, showing an anterior half or a posterior half of a bipolar meridian missing, indicate a possible explanation of how a single dividing individual could produce a proter and an opisthe differing in the total number of meridians.Meridian ranges of individual strains do not overlap each other in all cases, but they do constitute a progressively overlapping series which does not warrant taxonomic separation of strains at either extreme from each other.Contractile vacuole pores varied from 1 to 3, with 2 pores (1 each in meridians 5 and 6) being characteristic of 22 of the strains. The 2-pore 6-7 pattern was modal for 8 of the strains. Presence of 3 pores (in 5, 6 and 7) was typical of only 2 strains, but 3 pores were found occasionally. Another pattern of 3 pores (in 6, 7 and 8) was observed occasionally in 10 strains. Double pores (in 6 and 7 or 7 and 8) and the triple pore patterns were observed only in the strains having the highest total number of meridians. These variations are not considered adequate justification for taxonomic separation.Strains Ki and Aq are affirmed to be T. pyriformis. Strains V1 PP and V, formerly regarded as strains of T. vorax, are considered to be T. pyriformis, supporting the view of Shaw and Williams (1963).

    Notes: SYNOPSIS. Free amino acids in extracts of a number of tetrahymenid genera, species and strains of Tetrahymena pyriformis were separated by a combination of paper electrophoresis at pH 1.9 and descending paper chromatography with lutidine as solvent. Ethanol extracts of ciliates cultured at temperatures of either 15°, 25° or 35°C were used. Relative evaluation of ninhydrin spot intensity was made for T. pyriformis strains WH52, UM226, hybrids No. 25, No. 76, PR, HS and N. Extracts from 15° and 25°C cultures of T. limacis, T. setifera HZ1, T. vorax V2, Glaucoma chattoni A and Colpidium campylum C were analyzed in the same way.Seventeen spots were usually found on all chromatograms indicating the presence of the substances alanine, arginine, aspartic acid/asparagine, cysteine/cystine, glutamic acid/glutamine, glycine, histidine, leucine, lysine, methionine, phenylalanine, proline, serine, taurine, threonine, tyrosine and valine. No cysteic acid was demonstrable. Cysteine/cystine, histidine, phenylalanine and taurine generally occur in relatively low concentrations or appear to be absent. Those ranked as generally abundant include aspartic acid/asparagine, arginine, serine and threonine. Spot intensities for the remainder of those listed above indicate they were very abundant in most samples.FAA analyses have now been made by us, including those in a previous report, on tetrahymenid ciliates belonging to 3 genera (Colpidium, Glaucoma and Tetrahymena), on 4 species of Tetrahymena (limacis, setifera, vorax and pyriformis) and on 11 strains of T. pyriformis. Although extracts have been compared from ciliates grown at temperatures of 10°, 15°, 25° and 35°C, no striking differences in the qualitative or quantitative composition of the known amino acids in these patterns were observed. It was not possible to establish correlations with serotypes, and no features that might serve as useful taxonomic criteria were observed.
