    Notes: A 26-year-old African female presented with primary amenorrhoea and genital abnormalities. She had the physical stigmata of Turner's syndrome and a 45, X karyotype. Severe masculinization of the external genitalia, apparently present from birth, was present and a laparotomy revealed a hilus-cell tumour of the right streak gonad. Clitoridectomy and vaginoplasty were performed but subsequent oestrogen therapy failed to produce breast development.

    Notes: Agglutination of Staphylococcus aureus bacteria particles, conjugated with specific virus antibodies, has been used to identify a wide range of plant viruses in crude sap extracts. The test distinguishes between seven different potyviruses in homologous and heterologous reactions, but does not distinguish different strains of bean yellow mosaic virus. The sensitivity of the virobacterial agglutination (VBA) test compares favourably with virus detection in ISEM particle trapping and local lesion assay and is only slightly less sensitive than direct ELISA tests. The test is more sensitive and simpler to user than latex particle agglutination tests.

    Notes: A rapid slide agglutination test using polyclonal antisera conjugated to protein A-rich whole-cell Staphylococcus aureus was developed for the detection and identification of bacteria from plants. The specificity and sensitivity of the technique was evaluated in 18 antibody/antigen combinations, representing six bacterial genera (Erwinia, Lactobacillus, Pseudomonas, Rhizobium, Rhodococcus and Xanthomonas). For two pathovars of Pseudomonassyringae the specificity of the technique was increased by the use of antisera prepared to somatic extracts.The advantages of the Staphylococcus aureus agglutination technique include speed, simplicity and the ability to identify organisms directly from infected plant tissues. It was applied to the detection of Pseudomonas syringae pv. phaseolicola and pv. pisi in lesions on bean and pea, respectively, to P. gladioli pv. alliicola and Lactobacillus sp. from rotted onion bulbs and specific strains of Rhizobium phaseoli in bean root nodules.

    Notes: Summary Two women with primary amenorrhoea and few other stigmata of Turner's syndrome were found to be chromosome mosaics: 45,X/46,X,idic(Y). In Case 1, the dicentric isochromosome Y was found to have a long-arm breakpoint of formation. This structure was interpreted as containing two Y short arms and centromeres separated by a region derived from the proximal Y long arm. One of the centromeres in the Case 1 —idic(Y) was suppressed in 80% of cells in blood, and in these cells it appeared as a regular Y-shaped chromosome. In Case 2 the idic(Y) was derived by a short-arm breakpoint of formation. In all the dicentrics of this case with one primary constriction (functional monocentrics) there was a single Cd band. In the 10% of dicentrics with two primary constrictions, there were two Cd bands. It is argued that the instability of sex isochromosomes is due to this functional dicentricity in some cells. These cases are compared with 42 other Y isochromosomes with various short- and long-arm breakpoints of formation. It is suggested that some of the nonheterochromatic, nonfluorescent Y chromosomes previously reported may be explained as dicentric i(Y) with proximal long-arm breakpoints of formation and one suppressed centromere.
