    Notes: Monoclonal antibodies (MoAb) to defined allergens of P. pratense were raised. Five of them were selected for detailed studies by means of immunoblotting after SDS–PAGE and IEF of extracts from P. pratense and L. perenne. Three antibodies (1D11, 3B2, 2D1) recognize structures with mol. wt of 29 and 34 kD and pI of 4.4-7.7, corresponding to group V allergens. Two other MoAbs (2D8, 3C4) are directed against strong basic structures with a mol. wt of 50 kD and pI of 7.7-9.9 according to group IV allergens. The specificity of antibodies was supported by direct ELISA with purified group V and IV allergens. The isolated allergens were characterized before by SDS-PAGE and CIE and that allergenicity was detected with sera of patients with allergic rhinitis. Using our selected MoAbs crossreactive epitopes on group V and IV allergens have been excluded. Our antibodies have been used to detect crossreactivity in 14 grass pollen extracts. The evaluation of the pollen extracts has been performed by enzyme immunoassay (EIA) inhibition. One MoAb (3C4) is able to recognize group IV allergens in all grass species analysed whereas the MoAb 2D8 seems to identify group IV structures in selected grasses only. Binding to conserved structures of group V has been proved for MoAb 1D11. Other group V specific MoAbs (2D1, 3B2) identify similar, however incomplete, spectra. These results have been confirmed also by the dot immunobinding assay.

    Notes: A two-site solid-phase enzymimmunoassay was used for the quantification of group V allergen in grass pollen extracts in mass units. The assay is based on the monoclonal antibodies (MoAbs), 1D11 and 3B2 which recognize different epitopes on the standard Phl p V. The MoAb-ELISA is very sensitive (15 100ng ml Phl p V)and highly specific for group V allergens. Six pollen extracts of different grasses demonstrated parallel binding curves. The group V content ranged between 73 and 673 μg/ml in the extracts. The International Standard of Phleum pratense (IS 82 520) contains 400 μg/nil Phl p V. A good correlation was observed between the group V content and RAST inhibition, with the exception of Poa pratensis.

    Notes: A two-site monoclonal antibody (MoAb) ELISA has been developed for the quantification of the Phleun pratense major allergen, Phl p V. The assay is based on two MoAbs which recognize different non-overlapping epitopes on the Phi p V molecule; one antibody (1D11) was immobilized on the solid phase and the other (3B2) was biotinylated. An affinity-purified Phi p V preparation (purity of 95%) was used as standard. The assay has a sensitivity of 10 ng/ml of allergen and is suitable for the detection of group V allergen in aqueous grass extracts. The specificity of the assay was investigated with 14 grass pollen and five non-grass pollen extracts. Different levels of group V allergen were delected in extracts of grasses, but not in non-grasses. The assay gives a good correspondence with allergenic activity of extracts as determined by ELISA inhibition using serum pool of allergic patients. The results indicate that the two-site MoAb ELISA could be very useful in the standardization of allergenic extracts from grass pollen.

    Description / Table of Contents: Summary Replacement of the tracheal conduit remains an unresolved problem. The microporous material expanded polytetrafluorethylene (ePTFE) is suitable for tracheal reconstruction. The aim of our study was to improve the limited mechanical properties of this material by incorporating reinforcement elements and to examine the influence of these elements on host incorporation and epithelialization. In so dours ePTFE prostheses were reinforced with Ionomer cement and porous high density polyethylene (pHDPE) rings and implanted into the neck muscle of miniature pigs. One of these prostheses was epithelialized by a cell seeding technique and was thus placed into a tracheal defect. Results were examined grossly by endoscopy and than by light and scanning electron microscopy. The shapes of both types of prostheses showed a high stability. The reinforcement elements did not impair bioincorporation or the ability to epithelialize. In vivo interposition of an incorporated and epithelialized prosthesis to a host led to cell differentiation. The improved biomechanical properties of the prostheses waid and the reproducible formation of epitheliums are important advances in the solution of effectively correcting tracheal defects.
    Notes: Zusammenfassung Das Problem der Herstellung eines funktionierenden alloplastischen Tracheaersatzes ist bisher nicht gelöst. Mikroporöse Biomaterialien aus ePTFE (expanded Polytetrafluorethylen) sind für die Wandstruktur einer Tracheaprothese geeignet. Ziel unserer Untersuchungen war, die ungenügende Kompressionsstabilität dieses Materials durch zusätzliche Verstärkungselemente zu verbessern und den Einfluß dieser Elemente auf Inkorporation und Epithelisierung zu untersuchen. Hierfür wurden ePTFE-Prothesen mit Spangen aus Glasionomerzement und porösem „High-density-Polyethylen” (pHDPE) verstärkt und in den Halswendermuskel von Minischweinen implantiert. In einem Pilotversuch wurde eine solche Prothese mit der Technik des Zellseedings epithelisiert und in einen Tracheadefekt interponiert. Die Ergebnisse werden durch licht-, rasterelektronenmikroskopische und endoskopische Befunde demonstriert. Bei beiden Prothesenformen konnte eine verbesserte Kompressionsstabilität erzielt werden. Inkorporation und Epithelisierung werden durch die Verstärkungselemente nicht beeinträchtigt. Die Interposition einer bereits im Muskel inkorporierten und epithelisierten Prothese in das physiologische Milieu führt zu einer Epitheldifferenzierung. Die Verbesserung der biomechanischen Eigenschaften und die reproduzierbare Epithelisierung der Prothese sind wichtige Teilschritte auf dem Weg zur Lösung des Gesamtproblems.

    Notes: Abstract The advantages of laser welding are high process velocity, the possibility to joint alloys, which are not solderable, and the high bonding strength. During the laser welding process a high energy input in a very short time and a small area is performed, resulting in possible changes of structure and composition. Direct information of the change of the electrochemical behaviour of dental alloys after Laser welding procedure does not exist. The aim of this work was, therefore, to assess the electrochemical behaviour of laser welded joints in comparison with the bulk material using the mini-cell system. The logI vs. E curves performed on the bulk material show a complete change concerning the characteristic data, the shape and the position in comparison to such curves measured on the laser track. The characterisation of the electrochemical behaviour of laser welding tracks seems to be very easy by using the mini-cell system.

    Notes: Abstract No detailed information is available about the timing and correlation of objectively gauged ventricular width (VW) and the rate of growth of the occipitofrontal circumference in premature infants with intraventricular haemorrhage preceding progressive ventricular dilation. For this study, two groups were selected according to clinical course: group A(n = 6) had ventriculomegaly with no signs of raised intracranial pressure, while patients in group B (n = 7) developed progressive ventricular dilation after a period of latency. A VW between 0.9 and 1.4 cm (group A) did not affect normal head growth, whereas a VW greater than 1.5 cm was always associated with the development of hydrocephalus. Our data further suggest that the widely used criterion of a head growth rate of more than 2 cm/week is a relatively poor criterion for the definition of post-haemorrhagic hydrocephatus.

    Notes: The application of a Mini-TEA-CO2 laser for the modification of titanium surfaces to improve the bonding stability to a PMMA polymer is described. Cast and forged samples of titanium were treated with laser irradiation under different conditions. Depending on the kind of atmosphere employed during the laser treatment a differential change of bonding strength could be estimated. The compound stability was improved by a factor of 40 compared with the reference system. SEM pictures of the surface have shown that no cracks are formed but a modification of the laser-treated Ti surface topography similar to a freeze melting was observed. The results, found for a titanium dental polymer compound, should also be interesting for other medical applications.

    Notes: Abstract A monoclonal antibody with a high affinity for digitoxin (KA = 0.50 nM) and digoxin (KA = 0.55 nM) was produced by somatic cell fusion. This antibody, designated 2A3(47), displayed little cross reactivity with other glycosides. In cultured rat heart myocytes, 2A3(47), antagonized the positive chronotropic effect exerted by digitoxin but did not alter that of ouabain. Our results suggest that this monoclonal antibody may prove to be useful in treating digoxin and digitoxin intoxication.
