    Notes: A new type of real-time optical microscope has been developed that operates on the same principle as a liquid immersion microscope, with the liquid replaced by a solid lens of high refractive index material. Using a lens with an index n=2 and 436 nm illumination, this microscope has resolved 100 nm lines and spaces and has demonstrated a factor of two improvement in the edge response over a confocal microscope.

    Notes: We have isolated a monoclonal antibody (150) that recognizes a phosphorylation epitope on the microtubule-associated protein (MAP) 1B. Immunoblot analysis of the developing rat central nervous system shows that monoclonal antibody 150 is directed against a protein of ∼325 kDa (MAP 1B) that copolymerizes with microtubules through successive cycles of temperature-dependent assembly and disassembly. Furthermore, immunoprecipitated MAP 1B contains the epitope recognized by monoclonal antibody 150. Removal of phosphate from blotted proteins using alkaline phosphatase abolishes the binding of monoclonal antibody 150 to MAP 1B, indicating that the epitiope is phosphorylated. In the developing rat nervous system, immunohistochemistry with monoclonal antibody 150 shows that the phosphorylation epitope on MAP 1B is transiently expressed in growing axons but not in dendrites. For instance, in the neonatal rat cerebellum, the parallel fibres of granule cells are stained only during elongation and not after synaptogenesis. The monoclonal antibody 150 epitope is also transiently expressed in radial glial fibres and in certain cell nuclei. All immunostaining of sections with monoclonal antibody 150 was completely abolished by alkaline phosphatase treatment. These observations and previous ones made by us in cell culture (Mansfield et al., J. Neurocytol., 20, 654 – 666, 1991) suggest that the phosphorylation epitope on MAP 1B recognized by monoclonal antibody 150, which has not been previously detected in vivo, may be important in axonogenesis.

    Notes: Summary The release of gonadotrophin-releasing hormone (Gn-RH), in response to electrical stimulation of the mediobasal part of the hypothalamus incubated in vitro, was studied in both male and female rats. In male rats significant release of Gn-RH occurred during the 10-min experimental period only when the incubated tissue was stimulated at frequencies of 10 Hz or greater. There was no release when stimulated at 5 Hz. There was also no release of hormone when the mediobasal hypothalami were incubated in a Ca2+ free medium. The amount of Gn-RH released during a 10-min incubation period increased progressively as the frequency of stimulation was raised from 10–100 Hz. During short (4-min) incubation periods the effectiveness of each stimulus pulse for Gn-RH release also increased with the frequency of stimulation. However, when stimulated for 10 min there was no increase in hormone released per stimulus pulse when frequency of stimulation was raised above 10 Hz. The amount of Gn-RH released in response to stimulation at 50 Hz was greater in male rats than in females. For the females, there was no significant difference between the amounts of Gn-RH released at dioestrus and pro-oestrus. In both male and female rats gonadectomised 4 weeks prior to hypothalamic incubation, the response to electrical stimulation at 50 Hz was reduced when compared with intact controls. Indeed, for the females there was no longer a statistically significant increase in the amount of Gn-RH in the incubation medium after 50 Hz stimulation. Ovariectomised female rats, injected twice daily for 3.5 days with 20 μg of oestradiol benzoate released Gn-RH in response to 50 Hz stimulation in the same amounts as intact control animals. By contrast, there was no recovery of Gn-RH release to normal levels in castrated male rats similarly treated with 1.25 mg testosterone propionate.

    Notes: Abstract A commercial xylanase from Trichoderma longibrachiatum was used to treat the fractions of Douglas-fir kraft pulp of different fibre length. Enzymatic prebleaching was followed by chelation and peroxide bleaching. An evaluation of both optical and physical properties of the distinct fractions was conducted. A difference in susceptibility of the fractions of different fibre length to xylanase prebleaching was observed. The bleach-boosting effect observed with all fractions appeared to be related to the high-molecular-mass UV-absorbing material solubilized during enzyme treatments. Xylanase treatments resulted in beneficial effects to handsheet density and roughness as well as to some of the strength properties. However, the response to the xylanase treatments exhibited by all fractions of different fibre length was not uniform, indicating that fiber composition played a key role in the effectiveness of xylanase treatments.
