    Notes: Early development of the central nervous system is influenced by several hormones including thyroid hormone. This study was designed to clone the gene whose expression is changed in association with perinatal hypothyroidism in the rat cerebellum. Rats were sacrificed at 15 day-old postnatal age (P15) and their cerebella were removed. Poly (A)+ RNA was extracted to construct a cDNA library using λgt 10 cloning vector. Differential plaque screening was then performed using 32P-labeled antisense cDNA synthesized from poly (A)+ RNA of the methimazole-treated (hypothyroid) P15 rat cerebellum (hypothyroid probe), and of the euthyroid P15 rat cerebellum (euthyroid probe). The clones, which hybridized strongly to the euthyroid probe and weakly or not at all to the hypothyroid probe, were isolated. Sequence analysis of these clones revealed that all isolated clones encode cytochrome c oxidase subunit I (COX I), which is located in the mitochondria1 DNA. The decrease in COX I gene expression was not seen in the animals, which received methimazole treatment and daily replacement of thyroid hormone. In situ hybridization detection showed not only overall decrease in COX I gene expression but also change in distribution of hybridization signal in the cerebellar cortex of hypothyroid rat. Such change was not observed in the T4-replaced animals. Based on the evidence that thyroid hormone greatly influences brain development, the results of the present study indicate that the terminal enzyme of mitochondria1 respiratory chain, COX I is one of the important target molecules regulated by thyroid hormone in the newborn rat cerebellum.

    Notes: Abstract: A circadian rhythm in cellular polyamine levels was detected in the liver, pineal gland, anterior pituitary gland, Harderian gland, and testicular seminiferous tubules of male rats fed ad libitum and maintained in a light:dark cycle of LD 12:12 (lights on at 07:00). Liver putrescine content was highest at 24:00, showing a sixfold increase over 12:00 levels. Pineal sperrnidine and spermine contents reached a maximum at 06:00, late in the dark phase. A similar pattern was also detected in the Harderian gland. In the anterior pituitary, the polyamines putrescine, sperrnidine and spermine were highest at 18:00, late in the light phase. However, the increase in putrescine was not statistically significant. The three polyamine contents decreased late in the dark phase. In testicular seminiferous tubules putrescine, on the contrary, was highest (about a twofold increase) late in the dark phase.

    Notes: Abstract Eighteen strains of Rhizobium including four species, R. leguminosarum, R. meliloti, R. loti and R. fredii, nine strains of Bradyrhizobium japonicum and three strains of Azorhizobium caulinodans contained putrescine and honospermidine as major polyamines. All these nodulating N2-fixing rhizobia lack spermidine. Spermidine and cadaverine were present only in a limited number of R. meliloti and B. japonicum. Polymanine-synthetic activity was not affected by the differences in ability to produce phytoxine (rhizobitoxine and dihydrorhizobitoxine) H2-uptake-hydrogenation in the organisms. Putrescine and homospermidine were major polyamined in a strain of Agrobacterium rhizogenes. All the eight strains of Agrobacterium tumefaciens as well as A. rubi, A. radiobacter and two other strains of A. rhizogenes contained putrescine and spermidine as major polyamines and homospermidine and spermine (and thermospermine) as minor polyamines.

    Notes: Polyamines in 12 species (19 strains) of sulfuroxidizing eubacteria belonging to Thiobacillus were analyzed. Their polyamine distribution patterns were separated into 5 types. T. neapolitanus contained putrescine alone (first type), T. intermedius, T. perometabolis, T. thioparus and a strain of T. denitrificans putrescine and 2-hydroxyputrescine (second type), T. acidophilus, T. organoparus, T. versutus, T. tepidarius, T. thiooxidans and T. ferrooxidans putrescine and spermidine (third type), T. novellus putrescine and homospermidine (fourth type), and a strain of T. denitrificans diaminopropane, putrescine and homospermidine (fifth type). Thus, thiobacilli could be rearranged into different taxonomic positions within Proteobacteria on the basis of polyamine distribution patterns.

    Notes: A slightly thermophilic, CO-utilizing pseudomonad, Pseudomonas thermocarboxydovorans grown under both autotrophic and heterotrophic growth conditions was found to contain 2-hydroxyspermidine and 2-hydroxyputrescine in addition to putrescine, diaminopropane and spermidine. A mesophilic CO — utilzing hydrogen bacterium, Pseudomonas carboxydohydrogena contained putrescine and homospermidine under both autotrophic and heterotrophic growth conditions. Although these two carboxydobacteria are classified to the same genus Pseudomonas, the difference in their polyamine distribution patterns suggests that they may belong to different subclasses of Proteobacteria.

    Notes: Abstract The Gram-negative facultative chemolithotroph, Paracoccus denitrificans contains putrescine, cadaverine, agmatine, spermidine, aminopropylcadaverine, spermine, thermospermine and aminopentylnorspermidine. This bacterium has the ability to produce norspermidine from supplemented diaminopropane. The halophile, Paracoccus halodenitrificans is devoid of any polyamines. Neither decarboxylation of ornithine, lysine or arginine, nor triamine synthetic activity from diamines was detected in this halophile. Two Gram-negative facultative photoautotrophs, Rhodobacter sphaeroides and Rhodobacter capsulatus contain putrescine, cadaverine, agmatine and spermidine and can produce norspermidine from supplemented diaminopropane. A Gram-negative eubacterium, Micrococcus cryophilus, contains histamine and homospermidine in addition to putrescine, cadaverine and spermidine. Hence, polyamine distribution patterns and polyamine biosynthetic activities were very different among the four groups of Gram-negative eubacteria examined.

    Notes: Abstract An unknown highly basic polyamine peak was observed in Thermus thermophilus and it was identified to be a pentaamine [NH2(CH2)3NH (CH2)3NH(CH2)4NH(CH2)NH2] by gas chromotography-mass spectrometry and named as thermopentamine. The new pentaamine coexisted with another isomer, homocaldopentamine, at a molar ratio of 1:1, as a major polyamine of the organism. This is the first report to show the occurrence of thermopentamine in nature.

    Notes: Abstract Four unusual polyamines were detected in the Gram-negative, aerobic, nonsporulating eubacterium, Thermomicrobium roseum (ATCC 27502), an obligate for thermophily, in which no peptidoglycan dianmino acid occurs. They were identified as caldopentamine [NH2(CH2)3NH(CH2)3 NH(CH2)3NH(CH2)3NH2], homocaldopentamine [NH2(CH2)3NH(CH2)3NH(CH2)3 NH(CH2)4NH2], thermopentamine [NH2(CH2)3 NH(CH2)3NH(CH2)4NH(CH2)3NH2] and caldohexamine [NH2(CH2)3NH(CH2)3NH (CH2)3NH(CH2)3NH(CH2)3NH2] by high-performance liquid chromatography and gas chromatography-mass spectrometry. These unusual pentaamines and hexaamine were major polyamines in the organism grown at 60–70°C.

    Notes: Abstract Polyamines were analyzed in 4 species of genus Agrobacterium. Not only putrescine, spermidine and spermine, but also homospermidine and thermospermine were found in A. tumefaciens, A. radiobacter, A. rubi and A. rhizogenes. Trace amounts of aminopropylhomospermidine were also observed. Norspermidine and norspermine were formed from diamonorpropane added to the medium. Aminopropylcadaverine and its aminopropyl derivative(s) (aminopentylnorspermidine and N,N′-bis(3-aminopropyl) cadaverine) were produced from the supplemented cadaverine. A strain of A. rhizogenes normally contains only putrescine and homospermidine; no other diamines, triamines and tetraamines were synthesized.
