    Notes: Abstract Employing a mineral salts medium containing Tween 80 as the primary carbon source, a strain of Bacillus stearothermophilus was isolated which was able to hydrolyse selected second and third-generation pyrethroids to non-insecticidal products. Of a range of pyrethroid insecticides the trans-isomer of permethrin was the most readily transformed by this microbial isolate, whilst flumethrin was the least. 3-Phenoxybenzoic acid and the respective halovinyl or haloacid moieties were detected as the major hydrolytic products of the pyrethroids. It is believed that 3-phenoxybenzoic acid was formed from 3-phenoxybenzyl alcohol which was not however detected as an intermediate in these systems. 3-Phenoxybenzoic acid was further transformed to 4-hydroxy-3-phenoxybenzoic acid. A potential metabolic pathway has been described.

    Notes: Abstract A large number of FMRFamide-related peptides (FaRPs) are found in nematodes, and some of these are known to influence tension and contractility of neuromuscular strips isolated fromAscaris suum body wall. Relaxation of these strips has been noted with five nematode FaRPs. The inhibitory actions of SDPNFLRFamide (PF1) and SADPNFLRFamide (PF2) appear to be mediated by nitric oxide, as previously demonstrated with inhibitors of nitric oxide synthase (NOS). This present study showed that the effects of PF1 were also dependent on external Ca++ and were reduced by the Ca++-channel blocker verapamil, observations consistent wirh the finding that nematode NOS is Ca++-dependent. KSAYMRFamide (PF3), KPNFIRFamide (PF4) and KNAFIRFamide (an alanine substituted analog of KNEFIRFamide, AF1, termed A3AF1) also relaxed A.suum muscle strips, but these responses were not affected by NOS inhibitors. PF3 inhibited the activity of strips prepared from the dorsal side of the worm, but contracted ventral strips. Both effects were dependent on the presence of ventral/dorsal nerve cords (unlike PF1/PF2) and were attenuated in medium which contained high K+ or low Ca++. PF4-induced muscle relaxation and hyperpolarization were independent of nerve cords, but were reversed in Cl-free medium, unlike PF1 or PF3. The PF4 effect physiologically desensitized muscle strips to subsequent treatment with PF4 and/or GABA. However, PF4 and GABA were not synergistic in this preparation. The effects of GABA, but not PF4, were reduced in muscle strips treated with the GABA antagonist, NCS 281-93. Following PF4 (or GABA) relaxation, subsequent treatment with higher doses of PF4 caused muscle strip contraction. A3AF1 was found to relax muscle strips and hyperpolarize muscle cells independently of the ventral and dorsal nerve cords, K+, Ca++, and Cl-, and mimicked the inhibitory phase associated with the exposure of these strips to AF1. On the basis of anatomical and ionic dependence, these data have delineated at least four distinct inhibitory activities attributable to nematode FaRPs. Clearly, a remarkably complex set of inhibitory mechanisms operate in the nematode neuromuscular system.

    Notes: Summary Plasmodesmata are complex channels within the plant cell wall, which create plasma membrane and symplastic continuity between neighbouring cells. To detect plasmodesmata in cell wall preparations fromNicotiana cle elandii, we have used 3,3′-dihexyl-oxacarbocyanine iodide (DiOC6), a cationic amphiphilic fluorescent probe, widely employed for general studies of membrane structure and dynamics. Punctate fluorescent staining was readily seen in pit fields, small depressions within the cell wall known to be rich in plasmodesmata. Scanning electron microscopy was used to demonstrate that the punctate staining corresponded to plasmodesmata. Treatment of cell wall fragments with chloroform-methanol to remove lipids did not alter the staining of plasmodesmata. In contrast, pronase E-sodium dodecyl sulfate treatment completely abolished staining, indicating that the DiOC6 labelling of plasmodesmata may be protein rather than lipid specific. Although not membrane mediated, DiOC6 staining of plasmodesmata is a simple, rapid, and specific tool for the detection of plasmodesmata in isolated cell walls and will prove useful for studies of plasmodesmal location, structure, and composition.

    Notes: Abstract Two years ago the first platyhelminth regulatory peptide, neuropeptide F (NPF), was isolated from the tapeworm Moniezia expansa by Maule et al. (1991). NPF is a 39 amino acid peptide with a C terminal phenylalaninamide. NPF is the first platyhelminth neuropeptide to be sequenced fully. Preabsorption with NPF quenches the immunostaining with anti-FMRF-amide and anti-bovine PP (Halton et al. 1992). As the first authentic flatworm neuropeptide, the occurence and distribution of NPF along the whole flatworm line are under investigation. Both free-living and parasitic flatworms are being studied. So far NPF-immunoreactivity has been reported from three free- living flatworms (see Grahn et al., 1995) and from four parasitic flatworms (Marks et al., 1993). FMRF- and RF-amide immunoreactive (IR) nerve cells and fibres are common in the gull-tapeworm Diphyllobothrium dendriticum. In order to test whether the patterns for NPF- and RF-immunoreactivity co- localize in the gull-tapeworm, immunostaining with anti-NPF and anti-RF were performed. To broaden the study, adult Proteochepalus exiguus from the intestine of whitefish were included in the experiment. The study was performed on whole mounts of skinned worms (Gustafsson, 1991). Anti-NPF was used in concentrations 1:500 and 1:1000. Controls included liquid phase absorption with the homologous antigen (1000 ng ml−1). In D. dendriticum NPF-immunoreactivity occurs in nerve cells and varicose nerve fibres of larval and adult worms. The NPF-IR cell bodies are more common in the peripheral nerve cords than along the main nerve cords, which contain nerve fibres with large varicosities. The cell bodies in the PNS are often triangular in shape. Immediately beneath the tegumental surface a thin NPF-IR nerve fibre is observed. As to the co-localization of NPF and RF nothing definite can be said but the general pattern seems tobe the same. In the brain commissure of D. dendriticum one large ganglion cell stains with both antisera, indicating coexistence. In P. exiguus NPF- and RF-immunoreactivity was observed in the two main nerve cords situated laterally and in the pairs of thin dorsal and ventral longitudinal nerve cords. Numerous transverse commissures connect the longitudinal cords forming an orthogonal pattern. The cell bodies along the nerve cords are multipolar. Thin projections extend from the main nerve cords to the surface of the worm. The main nerve cords are lined with NPF-and RF-IR cell bodies. The general staining patterns of NPF and RF are very similar.
