    Notes: 1 Pig nasal mucosal strips were incubated with α-adrenoceptor antagonists followed by α2-adrenoceptor agonist concentration–response curves. 2 Contractions elicited by the α2-adrenoceptor agonists BHT-920 (pD2 = 6.16 ± 0.07), UK 14,304 (pD2 = 6.89 ± 0.13) and PGE-6201204 (pD2 = 7.12 ± 0.21) were blocked by the α2-adrenoceptor antagonist yohimbine (0.1 μm). In contrast, the α1-adrenoceptor antagonist prazosin (0.03 μm) had no effect on the BHT-920-, UK 14,304- and PGE-6201204-induced contractions, but blocked the contractile response to the α1-adrenoceptor agonist phenylephrine (pD2 = 5.38 ± 0.04) and the mixed α1- and α2-adrenoceptor agonist oxymetazoline (pD2 = 6.30 ± 0.22). 3 The α2-adrenoceptor antagonist yohimbine (0.01–0.1 μm, pA2 = 8.04), α2B/C-adrenoceptor antagonist ARC 239 (10 μm, pKb = 6.33 ± 0.21), α2A/C-adrenoceptor antagonist WB 4101 (0.3 μm, pKb = 8.01 ± 0.24), α2A-adrenoceptor antagonists BRL44408 (0.1 μm, pKb = 6.82 ± 0.34) and RX 821002 (0.1 μm, pKb = 8.31 ± 0.35), α2C-adrenoceptor antagonists spiroxatrine (1 μm, pKb = 7.32 ± 0.32), rauwolscine (0.1 μm, pKb = 8.16 ± 0.14) and HV 723 (0.3 μm, pKb = 7.68 ± 0.14) inhibited BHT-920-induced contractions in pig nasal mucosa. 4 The present antagonist potencies showed correlations with binding affinity estimates (pKi) obtained for these antagonists at the human recombinant α2A- and α2C-adrenoceptors (r = 0.78 and 0.83, respectively) and with binding affinity estimates (pKd) obtained in pig native α2A- and α2C-monoreceptor assays (r = 0.85 and 0.78, respectively). No correlation was observed for the α2B-subtype. 5 In conclusion, contractile responses to phenylephrine, BHT-920, UK 14,304, PGE-6201204 and oxymetazoline indicate that α1- and α2-adrenoceptors are present and mediate vasoconstriction in pig nasal mucosa. Furthermore, correlation analysis comparing antagonist potency in pig nasal mucosa with affinities for human recombinant α2-adrenoceptors and native pig α2-adrenoceptors suggest that α2A- and α2C-adrenoceptor subtypes constrict pig nasal mucosa vasculature.

    Notes: By observation of the intensity of the collisionally excited Li resonance line along an energetic beam injected into a low-density plasma, it is possible to infer the density profile.1 On ASDEX this technique is employed to continuously measure the density profile at the plasma edge with sub-ms time resolution. The Li-beam diagnostic system is described and results presented, for a beam energy of 60 keV, which show the method to produce profiles with low statistical noise, enabling small changes in density to be followed. The inferred relative density profiles are found to be in agreement with those determined by Thomson scattering and a Langmuir probe.

    Notes: Plasma diagnostics employing energetic lithium beams are in routine use on several present-day tokamak experiments. These measurements are constrained by the availability of a suitable lithium ion source. Necessary and/or desirable parameters include narrow energy spread, high-emission current density, simplicity, and long lifetime. The existing ion gun on the TEXT experiment uses the thermionic emitter β-eucryptite and meets the above requirements, except for being limited to a useful emission of about 4 mA. We will present design criteria and accelerator results for a new ion gun capable of 15-mA emission based on an increased emitter area. Important design considerations are the thermal and mechanical properties of the large (∼20 cm2) emitter area and the ion optics required to focus the emitted flux into a beam of ∼1-cm2 cross section. The potential improvement in the performance of both the TEXT and ASDEX diagnostic systems will be discussed.

    Notes: Abstract A 6-month-old female is described who presented with severe idiopathic macromastia. The breast enlargement began at 2 months of age and progressed such that subtotal mastectomies were necessary at 23 months. Extensive hormonal evaluation prior to surgery revealed no evidence of estrogenization or precocious puberty. There was no galactorrhea. A breast biopsy showed immature mammary tissue. In vitro analysis of the patient's serum using a mouse mammary thymidine incorporation assay revealed similar mitogenic activity in the patient's serum compared to adult controls. Post surgical follow up of this patient, 3.5 years later, has revealed no breast enlargement, precocious sexual development, or growth acceleration. Of interest, however, she has manifested an idiopathic degenerative neurologic condition characterized by psychomotor delay, ataxia, and seizures. Remarkably, hormone studies at age 5.5 years showed an exaggerated gonadotrophin response to intravenous gonadotrophin releasing hormone and prepubertal estrogen levels. While this case may represent an extraordinary example of idiopathic premature thelarche, the severe nature of this infant's macromastia in association with neurologic dysfunction and increased gonadotrophins suggests that central nervous system factors were etiologic.

    Notes: Summary Six healthy men walked 37 km (23 miles) per day over a 3-lap course for each of 4 consecutive days. Subjects were allowed breakfast and an unrestricted diet was consumed after completion of the walk, but no food was consumed during or between laps. At a later date the same subjects walked over the same course after an overnight fast and without breakfast. Completion time for each lap was 139±1 min (mean ±SE) and exercise intensity was equivalent to 17±1% $$\dot V_{{\text{O}}_{{\text{2 max}}} } $$ . Mean 24h energy intake was 14.5±0.8 MJ during the fed walk. Estimated daily energy expenditure was 12.0 MJ. Blood glucose concentration fell significantly on the first, third and fourth days of the fed walk, but no subject became hypoglycaemic. Glucose concentration did not fall during the fasted walk and was significantly higher pre-exercise and at the end of laps one and three when compared to the first day of the fed walk. Blood alanine concentration fell significantly after the end of the first lap of each day of the fed walk but not during the fasted walk. Blood lactate levels did not change during the course of either walk. Plasma free fatty acid, glycerol and blood 3-hydroxybutyrate concentrations were unchanged during the passage of the first lap on each day of the fed walk, but all three had increased significantly by the end of the first lap of the fasted walk. The elevations from rest to the end of each day's walking for these three metabolites were of similar magnitude when comparing each day of the fed walk and the fed and fasted walks. The present experiment indicates that feeding a mixed diet can affect the pattern of substrate mobilization in a similar manner to that seen during more strenuous exercise. It also appears that the pattern of fuel substrate mobilization is very similar from one day to the next providing dietary intake is similar.

    Notes: This study examined whether satellite cells express an embryonic isoform of myosin upon fusion with hypertrophying muscle fibers. Anterior latissimus dorsi (ALD) muscle hypertrophy was induced in adult chickens by weighting one wing. One and 7 days of wing-weighting produced significant increases in ALD muscle wet weight and in the number of mature fibers expressing ventricular-like embryonic (V-EMB) myosin. V-EMB myosin expression could be an event during regeneration of fibers injured by overload or part of the hypertrophy process itself. Although there was an increase in both the number of damaged fibers and the number of mature fibers expressing embryonic myosin after wing-weighting, results from this study suggest that these two events were not necessarily related. The apparent health of fibers expressing V-EMB myosin and the lack of correlation between the numbers of damaged and V-EMB myosin positive fibers (r = 0.20) suggest that embryonic myosin expression in mature fibers was likely a feature of the hypertrophy process itself. The appearance of V-EMB myosin in mature fibers 1 day after wing-weighting suggests that the change in myosin expression did not involve satellite cells since 24 hr is too short a time to permit more than limited satellite cell fusion. The relationship between satellite cells and embryonic myosin expression was examined more closely by labeling dividing satellite cells and their progeny with 5-bromo-2-deoxyuridine, and then colocalizing labeled myofiber nuclei and embryonic myosin in consecutive transverse sections of hypertrophied ALD muscle. One week of wing-weighting resulted in marked increases in myofiber nuclear labeling index and myofiber nuclear density compared to contralateral control. V-EMB myosin was not expressed uniformly throughout individual fibers, but rather in discrete regions of varying length. Many V-EMB myosin positive regions had a higher labeled nuclear density than V-EMB myosin negative regions indicating that V-EMB myosin expression was associated with an accumulation of satellite cell progeny in a restricted area. However, it was also clear that satellite cell progeny were not the sole source of V-EMB myosin since labeled nuclei were completely absent from 41% of the V-EMB positive regions. Futhermore, the presence of new nuclei did not result in obligatory expression of embryonic myosin because many V-EMB negative regions had a high labeled nuclear density. Thus, recently incorporated nuclei arising by satellite cell division are implicated as one, but not the sole source of embryonic myosin in hypertrophying muscle. These observations suggest that the non-uniform pattern of embryonic myosin expression in hypertrophying muscle resulted from differences in nuclear activity, not necessarily the addition of new nuclei. © 1994 Wiley-Liss, Inc.
