    Notes: This report discusses the results of a collaborative European Group on Fracture exercise to predict the pressure-crack size relationships of an experimental cracked pressure vessel which was undergoing tests for the CEGB. The details of the test and background mechanical properties data had been circulated to interested parties with a request for predictions to be returned prior to the vessel test results being published. The results of this exercise are described here, analysed in detail and compared with the test results.Collectively, eighteen individuals provided thirty eight different estimates of the vessel behaviour. The procedures used included R6 and variants of it, crack tip opening displacement approaches, engineering J design curve approaches, simple plastic collapse criteria and the GE J estimation scheme.Analysis of the predictions showed that:(1) in the prediction of maximum pressures, the plastic limit pressure was important;(2) in the prediction of initiation pressures the initiation toughness dominated;(3) the detailed form of the resistance curve had little influence on either the crack growth predictions or the maximum pressure predictions.(4) all forms of failure assessment diagram based upon the R6 axes were capable of producing accurate predictions.(5) where a poor model was used to represent the cracked section the results were unsatisfactory.

    Notes: The results of a round robin exercise in predicting ductile instability in two compact tension test specimens are presented and analysed. Many techniques were used, both to analyse the given raw data and to analyse the specimen behaviour, and large differences were obtained especially in defining J-resistance parameters. Despite these differences there was little scatter in the final result principally because the specimens failed at plastic collapse. It was concluded that all the techniques were broadly satisfactory provided a plastic collapse analysis is performed, either as part of the analysis or independently.

    Notes: Summary Using a variety of techniques, we have investigated the possible interrelationship between tumour-associated immunoglobulins and infiltrating host cells. Initial cytological and histological studies revealed that tumour models which normally have a high immunoglobulin content also contained a relatively high proportion of host cells which could bind immunoglobulin by cytophilic processes. Rosette assays on tumour cell suspensions from mice also challenged with SRBC suggested that some of the immunoglobulin present may be non-specifically bound to host cells or synthesized in situ. Evidence that the local production of immunoglobulin could occur was obtained by protein A-PFC analysis of tumour cell suspensions. These studies also revealed that there were many more IgA-secreting cells within tumours than in spleen or lymph nodes. Finally, immunoglobulin analysis on gradient-separated tumour cell suspensions indicated that immunoglobulin was associated with both tumour and host cell-enriched (i.e., Fc-positive) fractions. These observations emphasize that much of the immunoglobulin previously noted in tumour eluates may not be associated with tumour cells as such, but may be bound to or synthesized by infiltrating host lymphoreticular cells.

    Notes: Summary The effects of a variety of adjuvant protocols on immunoglobulin levels in normal and tumor bearing CBA mice have been investigated together with their ability to elicit immunoglobulin which bind to tumor cells in vitro and inhibit the growth of a transplanted syngeneic MC-induced fibrosarcoma. A marked increase in serum levels of certain immunoglobulins (especially IgG 2a , IgG 2b and IgM) and immunoglobulin interacting with tumor cells in vitro was noted in normal and tumor bearing mice following the administration of C. parvum (strain No. CN6134 and 10387), P. freudenreichii (strain No. 10470) and B. pertussis while a modest increase in some of these accompanied BCG injection. The Freund's complete and incomplete adjuvant protocols adopted had little effect on any of these parameters. The C. parvum protocols alone inhibited tumor growth. The immunoglobulins evoked by C. parvum strain No. 6134 which bound to tumor cells in vitro were extremely heterogeneous, activity being detected in all Ig classes and subclasses. This organism also evoked immunoglobulins which interacted with syngeneic embryonic fibroblasts, and adult syngeneic kidney and spleen cells. Tumor cells which had been preincubated in sera rich in immunoglobulins binding to tumor cells in vitro (i.e. from C. parvum-treated mice) did not exhibit reduced growth following i.v. or s.c. transplantation to syngeneic recipients.

    Notes: Summary The administration of C. parvum to mice bearing a transplanted syngeneic MC-induced fibrosarcoma resulted in a significant increase in tumor-associated immunoglobulin levels. The increase was most apparent in the IgA class, though significant increases were noted on occasion in the IgM class and certain IgG subclasses. In contrast, the C. parvum treatment used failed to induce antibody to AKR-type murine leukemia virus envelope components in either normal or tumor-bearing mice.
