    Notes: Initial results from direct-drive spherical cryogenic target implosions on the 60-beam OMEGA laser system [T. R. Boehly, D. L. Brown, R. S. Craxton et al., Opt. Commun. 133, 495 (1997)] are presented. These experiments are part of the scientific base leading to direct-drive ignition implosions planned for the National Ignition Facility (NIF) [W. J. Hogan, E. I. Moses, B. E. Warner et al., Nucl. Fusion 41, 567 (2001)]. Polymer shells (1-mm diam with walls 〈3 μm) are filled with up to 1000 atm of D2 to provide 100-μm-thick ice layers. The ice layers are smoothed by IR heating with 3.16-μm laser light and are characterized using shadowgraphy. The targets are imploded by a 1-ns square pulse with up to ∼24 kJ of 351-nm laser light at a beam-to-beam rms energy balance of 〈3% and full-beam smoothing. Results shown include neutron yield, secondary neutron and proton yields, the time of peak neutron emission, and both time-integrated and time-resolved x-ray images of the imploding core. The experimental values are compared with 1-D numerical simulations. The target with an ice-layer nonuniformity of σrms=9 μm showed 30% of the 1-D predicted neutron yield. These initial results are encouraging for future cryogenic implosions on OMEGA and the NIF. © 2002 American Institute of Physics.

    Notes: The upgrade to the OMEGA laser system, a 30-kJ, 351-nm, 60-beam direct-drive laser-fusion system is reported here. The system configuration is presented along with the status of its construction and the plans for system activation. The system utilizes rod and disk amplifiers and frequency tripling to produce UV pulses which are smoothed using phase conversion and smoothing by spectral dispersion. Dual driver lines will feed the propagation of two co-axial beams, which have different pulse widths and occupy different portions of the laser aperture. The laser system will be completed in November 1994 and the target area in March 1995. © 1995 American Institute of Physics.

    Notes: Abstract. The role of extracellular water in ameliorating drought stress was examined in Hemizonia luzulifola, an annual composite that comprises two subspecies differing significantly in the amount of extracellular polysaccharide within basal leaves. Rosette leaves of the high polysaccharide (HP) ssp were more than 30% pectin on a dry weight basis in contrast to only 4% in the low polysaccharide (LP) ssp. Concomitant with this difference in polysaccharide content was a significant difference in the dehydration response of leaves. Near full hydration, relative capacitances of HP leaves (0.7 MPa-1) were an order of magnitude greater than the LP leaves (0.08 MPa-1). Relative capacitance of the polysaccharide alone was 1.5 MPa-1. The weight of water per unit leaf area was not significantly different in the two taxa. Nevertheless, the pectin-like polysaccharides significantly altered the distribution of metabolically available water from primarily cell-stored water in the LP ssp to apoplasmic and symplasmic capacitors in leaves of the HP ssp. The consequence of this apoplasmic water store with colloidal properties is that the Ψ-dependence of leaf capacitance ceases to be linked directly to cell water relations. Transfer resistances for water movement between capacitors and the xylem near full hydration (0 to -0.5 MPa) were significantly larger in the HP leaf. This difference in transfer resistance was interpreted to be the result of a large resistance to water movement between the polysaccharide and the xylem. Because of these large transfer resistances, the apoplasmic capacitor probably buffers cells at lower water potentials under transient water fluxes than expected from laboratory measurements made during slow desiccation. Field measurements support this conclusions; HP leaves were better buffered than LP leaves at midday water potentials.

    Notes: Many trees in tropical dry forests flower during the dry season when evaporative demand is high and soil water levels are low. In this study the factors influencing the water balance of flowers from three species of dry forest trees were examined. Flowers had greater mucilage contents than leaves, high intrinsic and absolute capacitances, long time constants for water exchange and high transfer resistances. Flower water potentials were higher than in leaves and did not fluctuate over the lifespan of the flower. Flower water content also remained constant even though evaporation rates were high, suggesting that water was being supplied from the stem. In two of the species, the water potential gradient between flowers and leaves was opposite to that necessary for water transport from stem to flowers through the xylem, and it was therefore hypothesized that water may enter the flower through the phloem. Calculations showed that nectar production in these flowers could drive a sink of sufficient magnitude to allow water input via the phloem equal to water lost from the flower to the atmosphere.

    Notes: The difficulty in obtaining mutants in pathogenic Neisseria has limited the ability to genetically define determinants responsible for virulence as well as the ability to generate a genetic map. We show that the 16.5kb conjugative transposon Tn916 can be introduced into Neisseria meningitidis on the suicide vectors pAM120 and pAM170. After introduction, Tn916 transposed to different sites in the chromosome of recipient meningococci, apparently at random, and was stably incorporated. Following its integration into the meningococcal chromosome, Tn916 did not appear to readily express its conjugative and transpositional functions. However, chromosomal DNA from Tn916-carrying meningococci could be used to transform other meningococcal strains to tetracycline resistance. These studies indicate that Tn916 may be an important tool for genetic analysis of N. meningitidis.
