    Description / Table of Contents: Summary Phosphate metabolism of phospholipids has been investigated in four cases of hereditary spherocytosis (HS) and compared with that of normal, healthy controls. At variance to other findings incorporation of inorganic phosphate into phospatidyl-ethanolamine has been found to be by one order greater with spherocytes than with normal erythrocytes. Moreover, a strong enhancement of phosphate metabolism of lecithin and phosphatidylserine was observed in not splenectomized HS-patients. Splenectomized HS-patients, however do not show increased metabolism of lecithin or phosphatidylserine.
    Notes: Zusammenfassung Bei vier Fällen von Sphärocytose wurde der Phosphatstoffwechsel der Phospholipide untersucht und mit dem von gesunden Kontrollpersonen verglichen. Im Gegensatz zu allen bisher bekannten Befunden fanden wir den Phosphateinbau in Phosphatidyläthanolamin bei Sphärocyten um eine Größenordnung höher als in normalen Erythrocyten. Außerdem läßt sich bei Patienten mit hereditärer Sphärocytose, denen die Milz noch nicht entfernt wurde, eine starke Erhöhung des Phosphatstoffwechsels von Lecithin und Phosphatidylserin feststellen. Splenektomierte Patienten zeigen demgegenüber in ihrem Lecithin- und Phosphatidylserinstoffwechsel nach unseren Untersuchungen keine Erhöhung gegenüber der Norm.

    Notes: Summary Degradation of etoposide is rapid under in vitro culture conditions. At pH 7.4 and 37°C, the isomerisation of trans-etoposide to the inactive compound cis-etoposide has a half-life of 2 days in Dulbecco's modified Eagle's medium and results in the loss of 90% of the active drug within 1 week. As a consequence, prolonged incubations with etoposide in in vitro assays may lead to erroneous interpretations ignoring the real in vitro situation. The degradation is not influenced by organic compounds such as bovine serum albumin or amino acids but depends strongly on the pH value and, to a lesser degree, on the ionic strength of the medium. Therefore, we propose a mathematical correction based on the pH value so as to obtain the real exposure of cells to trans-etoposide during in vitro assays.

    Description / Table of Contents: Summary Column chromatographic enrichment of DNA-polymerase activities revealed at least three different enzymes which were characterized by pH-, Mg++-, temperature-, template-dependency and end product of the enzyme reaction. Those confinements became necessary to rule out some confusion in the literature concerning the identification of the singular DNA-polymerase. The template characteristics pointed out many of previous investigations lacking specificity.
    Notes: Zusammenfassung Säulenchromatographische Anreicherung von DNA-Polymeraseaktivitäten leukämischer Zellen ergaben, daß in diesen Leukozyten zumindest 3 verschiedene Enzyme vorhanden sein müssen. Die einzelnen Enzymaktivitäten wurden durch Bestimmung von pH-Optimum, Mg-Ionenabhängigkeit, Temperaturabhängigkeit, Templatecharakteristika und Charakterisierung des Endproduktes der Enzymreaktion definiert und konnten eindeutig gegeneinander abgegrenzt werden. Eine solche Abgrenzung wurde notwendig, da viele bisherige Befunde mit unspezifischen Nachweismethoden Fehlinterpretationen verursacht haben.

    Description / Table of Contents: Summary Human transfer factor was prepared from “buffy coat” of human donors by ultra filtration. These batches of transfer factor were tested for their ability to increase DNA-synthesis of antigen(mitogen) incubated lymphocytes. An increase of DNA-synthesis was measured by the ratio of appr. 2. Column Chromatographic separation of transfer factor on sephadex G-10 or DEAE cellulose columns showed an elution pattern with many peaks. In the lymphoblastogenese assay some peaks were found to be stimulatory, others in contrast inhibitory. It seems likely from our results that transfer factor represents a mixture of different chemical compounds with activating or inhibitory action on lymphoblastogenesis.
    Notes: Zusammenfassung Menschlicher Transferfaktor wurde aus Speckhaut menschlicher Leukozyten durch Ultrafiltration gewonnen und die Wirkung auf die Lymphoblastogenese in vitro untersucht. Dabei fand sich eine Steigerung der Antigen(Mitogen)-induzierten Blastentransformation um das 2fache. Fraktionierung der erhaltenen Transferfaktorpräparation auf Sephadex G-10 oder DEAE-Zellulose ergab durchwegs eine mehrgipfelige Kurve. Die Zugabe der einzelnen Subfraktionen von Transferfaktor zu Lymphozyten in vitro zusammen mit Antigen (Mitogen) führte bei einzelnen Fraktionen zu einer Hemmung, bei anderen zu einer Aktivierung der Lymphoblastogenese. Es kann daher angenommen werden, daß bei der Substanz Transferfaktor heterogene chemische Moleküle vorliegen, wobei einerseits aktivierende, andererseits auch hemmende Substanzen der Lymphoblastogenese als Gemisch vorliegen.

Nach verschieden langer Dauer von Asphyxie und anschließender Wiederbeatmung wurde im Meerschweinchenherzen ATP, ADP, AMP, Kreatinphosphat und anorganisches Phosphat bestimmt. 2. Bereits 1 Minute nach Asphyxie kam es zu einem deutlichen Abfall von Kreatinphosphat und Anstieg von anorganischem Phosphat; 5 Minuten nach Asphyxie war Kreatinphosphat nicht mehr nachweisbar. 3. ATP fiel 2 Minuten nach Asphyxie ab und zeigte einen weiteren Abfall bis 15 Minuten nach Asphyxie, während AMP zuerst einen Anstieg, später einen Abfall zeigte. 4. Bei Asphyxiedauer von 1 Minute regenerierten sich die energiereichen Phosphorverbindungen im Meerschweinchenherzen nach Widerbeatmung von 1/2 Minute und bei 5 Minuten Asphyxiedauer nach 9 Minuten Wiederbeatmung. 5. Dauert die Asphyxie 7 Minuten, so ist keine vollständige Regeneration der energiereichen Phosphorverbindungen mehr erreichbar. 6. War bei der Wiederbeatmung eine 50- bis 60prozentige Regeneration des Kreatinphosphatanteiles eingetreten, so verschwanden im EKG regelmäßig die Rhythmusstörungen.
    Notes: Abstract In the myocardium of guinea pigs the levels of ATP, ADP, AMP, creatine phosphate and inorganic phosphate, respectively, were determined following different periods of asphyxia and subsequent reanimation. Depending upon the duration of asphyxia, all energy-rich phosphate compounds decreased, creatine phosphate demonstrating this effect most markedly. Following reanimation, the energy-rich phosphate compounds underwent partial or complete regeneration, unless asphyxia lasted longer than 5 to 7 minutes.

    Notes: Summary This study tried to evaluate the impact of adjuvant chemotherapy on the induction of chemoresistance in radically operated upon breast cancer patients. Remission rate, remission duration and survival of a group of women (n=22) treated with combination chemotherapy (adriamycin and cyclophosphamide, AC) for recurrent breast cancer after failed adjuvant therapy (cyclophosphamide, methotrexate, fluorouracil, vinblastine) were retrospectively compared with the clinical data of non-pre-treated patients (n=28) receiving the same regimen (AC). The two groups of patients were comparable with regard to their risk factors. In the group of women with prior adjuvant chemotherapy only 3 out of 22 had a partial response, lasting 3, 8, and 16 months; the median survival was 50 months. In the group without prior adjuvant therapy 3 complete and 7 partial remissions with a median remission duration of 15.5 months (range 2–54 months) were found; the median survival was 104 months. The percentage of objective responses among the non-pre-treated patients at 36% was almost significantly higher than that of the pretreated women with 14% (p〈0.1). Responders to chemotherapy after relapse profited in terms of survival within the first 3 years after radical mastectomy, although no statistically significant difference was observed. The survival data shown assume a “shifting” of women from a group with better prognosis to a group with unfavourable prognosis following failed adjuvant chemotherapy.
