    Notes: Abstract Shifts were induced into the microbial community of activated sludge by the pulse addition of soluble starch. The subsequent changes of amylolytic and proteolytic microbial populations were recorded. Four amylolytic strains were isolated and characterized with regard to carrying capacity, specific surface and growth kinetics. The competitive ability of these strains was studied by means of two-member competition experiments. These experiments were analysed according to the Lotka-Volterra model and the de Wit method. The different results obtained suggest that the dominance of the amylolytic Pseudomonas sp. (code 01) is based on a combined occurrence of high amylolytic activity, large relative cell surface, high maximum specific growth rate and reduced sensitivity towards associated proteolytic populations.

    Description / Table of Contents: Summary A colorimetric assay for routine enzymic determination of free sialic acid in milk is proposed. Sialic acid is cleaved intoN-acetyl-d-mannosamine and pyruvate byN-acetylneuraminic acid aldolase; the pyruvate content is measured by lactate dehydrogenase in the presence of NADH, as a change of absorbance at 340 nm. The proposed method showed good correlation with the chemical method (r=0.988), high recovery (98.0%) and repeatability (S r =0.5–0.6 mg/L) as well as high accuracy (S y,x =0.28 mg/L). The assay may be useful for routine analysis of milk, as an indicator for the quality of the raw material to be used for production of UHT milk. The normal value range is 9.0±4.0 mg/L (mean±standard deviation;n=41). Conclusions about the initial concentration of the free sialic acid in the raw material, in order to obtain an idea of the quality, cannot readily be drawn from determinations in UHT milk without knowledge of the severity of the heating process.
    Notes: Zusammenfassung Es wird ein colorimetrischer Test für die routinemäßige enzymatische Bestimmung der freien Sialsäure in Milch vorgeschlagen. Dabei wird Sialsäure inN-Acetyl-d-Mannosamin und Pyruvat durchN-Acetylneuramin-Säurealdolase getrennt; der Pyruvatgehalt wird mit der Lactatdehydrogenase in Anwesenheit von NADH durch die Änderung der Absorption bei 340 nm gemessen. Die Methode zeigte eine gute Korrelation mit der chemischen Methode (r=0,988), eine hohe Rückgewinnung (98,0%) und Wiederholbarkeit (S r : 0,5–0,6 mg/L) sowie eine hohe Genauigkeit (S y,x =0,28 mg/L). Dieser Test kann bei der routinemäßigen Analyse von Milch als Indikator der Qualität des Rohstoffes bei der UHT-Milchproduktion nützlich sein. Der normale Bereich ist 9,0±4,0 mg/L (mittlerer quadratischer Fehler;n=41). Schlußfolgerungen aus der Ausgangskonzentration der freien Sialsäure im Rohstoff zur Bestimmung der Qualität sind nicht ohne weiteres möglich, wenn die Intensität des Erhitzungsprozesses nicht bekannt ist.
