    Notes: Abstract Some morphological features (degree of nuclear segmentation in neutrophils and eosinophils), histochemical patterns (DHFR, LDH, G6PDH) and, in addition, the nucleic acid distribution (DNA, RNA with acridine orange) in the peripheral blood cells of Rana esculenta, during the hibernation phase were investigated. All the observed parameters varied significantly in the frog during hibernation in comparison with the active period. The most evident changes were an increase in the nuclear segmentation of the neutrophils and a lower activity of the histochemically demonstrable DHFR and LDH, probably due to cell ageing. These findings suggest that, in hibernating frogs, there is an increase in the life span of the peripheral blood cells as a consequence of a reduced metabolic activity and a slowing of haematopoiesis.

    Notes: Abstract Morphocytochemical correlation between proliferation (BrdU uptake) of the haemopoietic liver subcapsular tissue and peripheral blood cell characteristics was performed during active (summer) and hibernating (winter) periods in two Urodele species: Triturus carnifex and Triturus alpestris. The significant decrease of the haemopoietic cell proliferation during the hibernation was accompanied by some modifications both at erythrocyte and leucocyte level. This situation was primarily characterised by a hypofunctionality and ageing of these cells with the disappearance of immature elements from the circulation and cell loss by apoptotic mechanisms. The results indicate that the condition of low metabolism during the winter period in these Urodeles involved deep changes in the physiology of haemopoietic tissue and blood circulating cells.

    Notes: Abstract Morphometrical (image analysis of cell and nuclear shape factor), morphological (electron microscopy) and cytochemical (acridine orange fluoro-chromasia and propidium iodide-DNA flow cytometry) features of circulating red blood cells were investigated during two periods of the annual cycle of Rana esculenta: the active phase (July) and the underground hibernating phase (January). The results showed that the hibernating phase is marked by more homogeneity of the red cells, both at nuclear (strongly condensed chromatin) and cytoplasmic level (loss of intact organelles and acridine orange fluorochromasia). The almost complete disappearance of the ‘immature’ erythrocytes from the circulation, during the hibernating phase, should be related to a decrease of haemopoietic activity and to an increase of life span accompanied by uncommon storage in different organs both at vascular and intracellular level.

    Notes: Summary Cytochemical methods were used to determine the distribution of dipeptidylaminopeptidase IV (DAP IV) and II (DAP II) in lymphoid cell populations from patients with lymphoproliferative diseases. Special attention was paid to unusual intracellular distribution patterns which might correlate with the presence of various membrane markers. In healthy patients, about 50% of the circulating lymphocytes were found to be positive to both reactions, the intracellular distribution patterns being variable. The DAP IV reaction was negative in all B-CLL cases. In 2 cases of T-CLL with phenotype E+ OKT 3 + T 4 − T 8 + one was negative and one was weakly positive, while two cases of T-CLL with phenotype E+ OKT 3 + T 4 + T 8 − were both strongly positive. The other non-T lymphoproliferative diseases studied were negative for DAP IV, while one T-ALL and three T-lymphoma cases showed a strong granular or diffuse distribution. The DAP II reaction was strongly positive in all the T lymphoproliferative diseases studied, irrespective of their immunological phenotype. This reaction was also weakly positive in some cases of plasmocytoma and lymphoplasmocytoid lymphoma.

    Notes: Summary The possibility of using propidium iodide, a phenanthridinic fluorochrome specific for double-stranded nucleic acids, for the study of chromatin thermal denaturationin situ has been examined. Smears of lymphocytes and hepatocyte nuclei from 15-day-old rats were fixed in acetic acid-ethanol (1∶3 v/v), treated with RNAse and submitted to different protein extraction procedures, namely, incubation with pepsin, trypsin and sodium chloride. Denaturation experiments were performed in Sörensen buffer at pH 7.4 containing 10% formamide at temperatures between 27 and 95°C. The samples were stained with propidium iodide and mounted in buffer or glycerol. Measurements were performed with a microfluorometer at a wavelength of 546 nm. The results indicate a higher thermostability of lymphocytes as compared to hepatocytes. The denaturation pattern suggests a certain organization complexity of chromatin, better emphasized by the derivative curves which show the presence of at least three fractions with different melting points. After protein extraction, the denaturation curves exhibit a somewhat simplified pattern, with the disappearance of the most stable peak in the derivative curves. The samples mounted in glycerine exhibit a better stability of staining with time, and an increased quantum efficiency of the fluorochrome with regard to those mounted in buffer. These data confirm the importance of protein-DNA interactions in the organization of chromatin and point to some differences, depending on the cell type and on functional activity.
