    Notes: Abstract Clonal propagation of Virginia Pine (Pinus virginiana Mill.) was achieved by organogenesis on cotyledon explants. The influence of several cytokinins and abscisic acid on adventitious shoot production from cotyledon explants was investigated. Benzyladenine was more effective in shoot induction than three other cytokinins tested. Benzyladenine (22.2 μM) in combination with naphthaleneacetic acid(0.05 μM) in a Gresshoff and Doy (1972) medium was found to increase shoot bud production. Abscisic acid (7.6 μM) in combination with benzyladenine and naphthaleneacetic acid enhanced shoot formation by an additional 65%. Root initiation was achieved with 0.5 strength Gresshoff and Doy media amended with naphthaleneacetic acid (1.3 μM), indole-3-butyric acid (1.2 μM) and benzyladenine (0.4 μM). Over 2400 plantlets from 2 families survived and were transferred to a greenhouse in preparation for field planting. After ten months, the maximum number of surviving plantlets/seed explant from these two sources was 57 for family ALPV-38 and 41 for family ALPV-78, respectively.

    Notes: Summary DNA methylation has been associated with gene activity in differentiating and developing plant tissues. The objective of this study was to determine the involvement of methylation in the expression of a gene transferred into carrot (Daucus carota L.) tissues by particle bombardment. Expression of the Dc8-GUS gene construct in response to treatments with 5-azacytidine (S-azaC) and to in vitro methylation by methylases was investigated by histochemical assay of GUS activity. The 5-azaC treatment increased the frequency of Dc8-driven GUS expression in both calli and somatic embryos. The increase occurred with treatment either to E. coli containing the plasmid insert or to the carrot tissues before bombardment. GUS expression, increased by the 5-azaC treatment, was enhanced by ABA treatment of both calli and somatic embryos and was more prominent in the latter. Increased digestion of the 5-azaC-treated plasmid DNA with EcoRII suggested that demethylation had occurred. In vitro methylation of Dc8-GUS by methylases generally resulted in a lower frequency of GUS expression. SssI methylase completely inhibited GUS expression. The level of GUS expression was correlated with the extent of methylation of the plasmid.

    Notes: Abstract Enhancement of shoot regeneration from loblolly pine (Pinus taeda L.) cotyledon explants was studied by addition of abscisic acid (ABA) to Gresshoff-Doy (GD) shoot induction medium containing benzylaminopurine (BA) and naphthaleneacetic acid (NAA). Addition of ABA (10−7 M) doubled the morphogenic area of cotyledons and increased the fresh weight of cotyledon explants by 40 to 45% after 4 weeks. A 4-week exposure to ABA resulted in a larger morphogenic area per cotyledon than 3, 2, or 1 week(s) respectively. The enhancement by ABA was related to the explant seed source and was not increased by prolonged exposure. Compared to controls, shoot number was enhanced by 31% and 56% with 2 and 4 weeks of ABA (10−7 M) exposure, respectively. Abscisic acid has a role in enhancing shoot morphogenesis in loblolly pine.

    Notes: Summary Embryogenic callus developed in 55% of the mature embryo explants of Norway spruce (Picea abies L.) growing on a LP medium minus the amino acids and sugars (except sucrose). This is the highest reported yield of embryogenic callus from mature embryos of P. abies that has ever been reported. Callus induction from either the middle or the end of the hypocotyl of the embryos began after 2–3 weeks. Three types of calli were recovered: (a) globular, (b) light green-compact, (c) white mucilaginous. Only the white mucilaginous calli were embryogenic. The globular and light green-compact calli never become embryogenic, even after several subcultures. The development of somatic embryos was accomplished on half-strength macro-elements of NSIII medium containing 1 μM α-naphthaleneacetic acid, 1 μM abscisic acid, and 3% sucrose. The addition of 10−7 M buthionine sulfoximine to the medium increased the development of somatic embryos by three fold. These results suggest that there is a great potential for increasing the frequency and development of somatic embryos in P. abies. Careful selection of the genotype and modification of the culture medium is required.

    Notes: Abstract Dry seeds of Leucadendron laureolum (Lam.) Fourc. (Proteaceae) were exposed for different intervals (range: 7 to 84 days) to visible, UV-A and UV-B radiation of different biologically effective dose (range: 0 to 11.43 kJ m-2 d-1). Changes in seed germination, physiology and ultrastructure, and residual UV effects on seedling performance, were examined. Germination was depressed in seeds following short (7-day) exposures to UV radiation. This depression was intensified with increased UV exposure dose, and most pronounced at shorter UV-B wavelengths. Also glutathione reductase (GR) activities increased in seeds exposed to shorter UV-B wavelengths, but these were unaffected by irradiation dose level in the UV-B range. Electrolyte leakage rates from UV-irradiated seeds were unaltered, which indicated that germination depression did not result from intrinsic membrane damage. The reversal of germination depression (UV-induced dormancy) in UV-irradiated seeds by red light pointed to the possible involvement of phytochrome in this photo-response. Germination depression disappeared in seeds after 56-days irradiation, possibly due to photoreceptor damage by excess UV light. At this stage, all UV irradiated seeds, irrespective of treatment wavelength or dose level, exhibited increased electrolyte leakage rates, which indicated membrane perturbation. Also, increased GR activities were observed in irradiated seeds, but these were proportionately smaller in seeds exposed to shorter wavelength UV-B radiation (9.1 to 35.8% increase) than longer wavelength UV-A (73.4% increase) and visible (97.7% increase) radiation. This implied a metabolic limitation for scavenging of free radicals and peroxides in aging seeds exposed to UV-B radiation, which pointed to accelerated seed deterioration. It was indirectly supported by ultrastructural evidence of sub-cellular damage (lipid coagulation and plasmalemma withdrawal from cell walls) in embryonic tissues of seeds after 84 days UV-B exposure, and reflected in decreased leaf numbers, photochemical efficiencies, and foliar chlorophyll a and carotenoid levels in seedlings cultured from these seeds.

    Notes: Abstract Water status of Pinus taeda L. callus supported on Murashige and Skoog (MS) liquid medium was characterized over an 8 week period using thermocouple psychrometry. Medium with 30 gl−1 sucrose was used to produce a high water potential (Ψw) of −0.4 MPa (H), and the same medium was used to create a moderate Ψw of −0.7 MPa (M) by the addition of 10% polyethylene glycol (PEG, w/v, MW=8000). Calli were produced from cotyledon explants on H medium for 2 weeks and then transferred to either M or H medium. Callus absorption of PEG accounted for 40% of the callus dry weight and less than 7% of the callus fresh weight. Callus dry weight (without the PEG fraction) on M medium was 40% of that observed on H medium. Fresh weight on M medium was only 15% of that observed on H medium. The Ψw of both H and M media remained constant throughout the culture period. On H medium, callus Ψw and osmotic potential (Ψs) both increased 0.05 MPa/week with the callus Ψw approaching that of the external medium. On M medium, callus Ψw and Ψs both decreased more than 0.1 MPa/week with the callus Ψw decreasing greatly below that of the external medium. The latter was attributed to a rapidly produced osmotic shock induced upon callus transfer and/or PEG which caused less callus hydration and resulted in reduced growth. Callus turgor potential (Ψp) was estimated to be +0.02 to +0.09 MPa and turgor was maintained as callus Ψw increased or decreased. After 8 weeks, cell volumes from callus on M medium were 50 to 60% less than on H medium, suggesting that reduced cell volumes were related to turgor maintenance.
