    Notes: Zusammenfassung Die Untersuchung der pleochroitischen Höfe der Biotite in Orthogneisen, Paragneisen und Bündnerschiefern des südlichen Gotthardmassives ergab durchwegs ein ähnliches Alter, nämlich 〈 60 Millionen Jahre. Es bedeutet dies, dass eventuell vorhandene vortertiäre Höfe in prätriadischen Biotiten der Gneise während der alpinen Metamorphose fast oder ganz durch Rekristallisation zerstört worden sind.

    Notes: Summary A high-resolution laser diffration system suitable for studying the basic mechanical properties of small contractile single cells has been developed. This method was used to establish the mechanical behavior of 95 ventricular cells isolated from adult guinea pig hearts. During contraction, the sarcomere length shortened from 1828±43 nm (mean ±SD) to 1518±99nm. The maximal velocities were 1.98±0.64μm/s for shortening and 1.93±0.54 μm/s for re-lengthening. The twitch duration from 20% shortening to 80% re-lengthening was 622±120 ms.

    Notes: Abstract To measure the mechanical activity of enzymatically isolated mammalian myocytes the principle of laser light diffraction was used. Since the viability of isolated cardiac myocytes showed a marked dependence on the laser power used, an opto-electronic system with improved light sensitivity and low susceptibility to optical noise was developed. The high sensitivity was achieved by a novel approach in the detection of diffraction patterns, that provides a significant reduction of the amount of laser power required. This improvement rendered possible the application of laser diffraction during extended experiments including pharmacological interventions. The static performance of the system, as assessed by means of calibration gratings, showed a resolution in the order of 5 nm for small changes in sarcomere length in the range from 1.2 μm to 2.0 μm. Examples of measurements on resting and contracting cells are presented, and the limitations of the application of the system to biological specimens are discussed.

    Notes: Abstract  We used a fast, fluorescent, potential-sensitive indicator (Di-8-ANEPPS) in combination with laser-scanning confocal microscopy in the line-scan mode (temporal resolution 500 Hz) to independently determine the transmembrane potential in voltage-clamped cells. While a linear relation between command voltage and Di-8-ANEPPS fluorescence was found in unexcitable Sf9 cells, pronounced nonlinearities were observed in cardiac myocytes. Comparison of the fluorescence records and current traces indicated that most of the observed nonlinearities could be attributed to voltage-escape during flow of membrane current. Voltage-escape during large membrane currents may lead to various experimental difficulties during voltage-clamp experiments. The voltage recording technique based on fluorescence was then used to compare the voltage-escape during flow of Na+ and Li+ ions via voltage-dependent (TTX sensitive) Na+ channels in cardiac myocytes. In these experiments, no significant differences in the degree of voltage-escape was found, suggesting that the two currents were similar in amplitude. In addition to the application presented in this paper, confocal microscopic detection of transmembrane potential with fluorescent dyes may be a useful technique for experiments in preparations that are difficult to impale with microelectrodes because of their small size.

    Notes: Abstract We have used ‘caged-ATP’ to investigate the kinetic behavior of KATP channels in ventricular cells from rat heart. In whole cells, loaded with ‘caged-ATP’, an increase of intracellular [ATP] following a UV light flash produced a decrease of KATP channel current that was too slow (τ ≈ 300 ms) to be explained by the expected timecourse of ATP release (τ ≈ 3 ms) and the time-course of channel blockade by ATP (τ ≈ 20 ms). In isolated membrane patches, caged-ATP itself caused partial blockade of KATP channels. Under these conditions, photorelease of ATP caused channel activity to decline further. The results suggest that caged-ATP can bind to the KATP channel but, on binding, decreases the open probability to a lesser extent than does ATP. Additionally, the observations indicate that for photolytically-generated ATP to bind to the channel, caged-ATP must first unbind (slowly) from the channel. We conclude that ‘caged-ATP’ is not fully caged with respect to its allosteric action on the KATP channel.
