    Notes: Abstract. Clathrin, which constitutes coated vesicles, plays important roles in neuronal functions. In the brains of the patients with Alzheimer's disease, distribution of clathrin was immunohistochemically investigated using four monoclonal antibodies against clathrin light chains, LCB.1, LCB.2, X-16 and CON.1, to study the involvement of clathrin in the pathology of Alzheimer's disease. LCB.1, LCB.2, X-16, and CON.1 bind to the aminoterminus of the clathrin light chain b(LCb), to the neuron-specific insert of LCb, to the light chain a(LCa), and to LCa and LCb, respectively. In Alzheimer brains, granular staining of LCB.2 around neurons in the hippocampus was wekaer or patchcily defected in comparison with control brains. Some neurofibrillary tangles and neurons were intensely stained in Alzheimer brains by LCB.2, whereas neurons were weakly stained in control brains. Crowns of some senile plaques in the brains of early onset Alzheimer's disease were positively stained by LCB.2. LCB.1 supported the observations of LCB.2. Reactive astrocytes in Alzheimer brains were intensely stained by X-16. On the other hand, Western blot analysis using LCB.2 and X-16 demonstrated no apparent differences in protein amounts and molecular weights of LCa and LCb between control and Alzheimer brains. These observations demonstrated abnormal distribution of clathrin in Alzheimer brains, implying impairment of axonal transport in this disease.

    Notes: Zusammenfassung 1. Es wurde festgestellt, daß Ospolot [N-(4′Sulfamylphenyl)-butansultam-(1,4)] ein potenter Hemmer der Carboanhydratase sowohl in vitro als auch in vivo ist. Die Hemmung scheint „non-kompetitiv“ zu sein. Die Bindung zwischen dem Pharmakon und dem Enzym ist reversibel. 2. Die zeitlichen Abläufe der Hemmung der Carboanhydratase im Gehirn nach Verabreichung von Enzymhemmern von der Schwankung der Krampfschwelle nach elektrischer Reizung und der Konzentration der Pharmaka im Gehirn gehen parallel. Da die Konzentration der Pharmaka in den Erythrocyten viel früher das Maximum erreicht, scheint der antiepileptische Effekt der Carboanhydratase-Hemmer nicht mit der allgemeinen Acidosis, sondern auf eine direkte Hemmung der Gehirn-Carboanhydratase zurückzuführen zu sein. 3. Die Verschiedenheit der klinischen Indikationsbreite von Acetazolamid und Ospolot scheint davon abzuhängen, wie die Pharmaka im Gehirn zur Verteilung kommen.

    Notes: Summary Linkage and haplotype analysis of eleven early-onset Alzheimer disease (AD) families was performed in relation to D21S210 and microsatellite DNA polymorphisms localized on chromosome 14q24.3. Linkage analysis of eight informative families out of eleven early-onset AD families disclosed the highest LOD score of 3.45 (θ=0.00) at D14S77, while the locus of β/A4 amyloid protein precursor gene was formally excluded within 10 cM from D21S210, given the evidence of recombinations in five families. Transmission disequilibrium study between the patients and controls without dementia indicated significant differences at D14S43 (p=0.0001) and D14S71 (p=0.02). Association study between genotypes linked or related to onset of AD and those of control also revealed a significant difference at D14S43 (p〈0.05), suggesting the existence of linkage disequilibrium. Moreover, the haplotypes at D14S43 linked with the onset of AD indicated a significant relationship with the mean age at onset. These results support that the major locus of earlyonset familial AD is located on 14q24.3, and its close linkage to D14S43 and the existence of allelic heterogeneity were suggested.
