    Notes: Previous studies from our laboratory suggested that architectural alterations of the cell membranes may have a major significance in the pathogenesis of psoriasis. Consequently, the membrane-bound Mg++ activated ATP-hydrolytic activity was investigated in normal and psoriatic epidermis under the electron microscope.The present study revealed that, in contrast to normal epidermis, only minimal ATP-hydrolytic activity is present on the cell membranes of psoriatic keratinocytes. This finding may reflect either a diminished attachment of substrate on the altered cell surface or an enzyme defect of the cell membrane. In both cases the reduced interaction between ATP and membrane-bound ATP-hydrolysing enzymes represents a functional membrane disorder of the psoriatic keratinocyte, presumably resulting in an insufficient utilization of ATP for active transport mechanisms on the cell surface.

    Notes: Multiple malignant Spiegler tumors were observed on the scalp of a 67-year-old female patient with brachydactyly and racket-nails affecting all fingers of both hands. The occurrence of similar clinical features in the family of the patient is suggestive of a hereditary disorder. The light and electron microscopical findings are described.

    Notes: This is a report of a case of a 12-year-old boy with an asymptomatic papular eruption present since birth, which showed the histological features of a hamartomatous proliferation of cutaneous smooth muscle cells, most likely of the arrectores pilorum muscles. The present paper describes the fine structure of this condition, for which the term “cutaneous smooth muscle hamartoma” is suggested.

    Notes: Summary.— The structure of callus differs in many respects from that of normal human stratum corneum. This finding was confirmed both by scanning and transmission electron microscopy. Callous material consists of 2 types of horny cell as does normal stratum corneum. Both types, however, are thicker (5– 8 μm) and extremely interdigitated as compared with normal horny cells. All intercellular spaces in callus are occupied by a dense cement material; signs of desquamation are therefore absent. After treatment with acid sodium thioglycolate (aNaThG) the dense matrix of callus keratin is reduced, the keratin framework of the horny cells is loosened and the characteristic α-keratin pattern can be observed. Moreover, most of the intercellular cement material is dissolved and some of the desmosomes are degraded. After aNaThG treatment and prolonged fixation in osmium, only the horny cell borders and a fibrillar skeleton remain.According to our findings, callus keratin is constructed similarly to the keratin of finger pad epidermis and of nail. The mode of keratinization in callus resembles that of hyperkeratosis by proliferation and not parakeratosis. Compared to nail keratin and the keratins of hair, however, callus keratin is less resistant to aNaThG. The high aNaThG-resistance of the horny cell border points to its function as a solid, stable container for keratins.

    Notes: Drugs and metastatic malignant tumors may induce severe alterations in cutaneous axons. The most prominent ultrastructural feature of axonal dystrophy of this type was the presence of multiple lamellated osmiophilic bodies (LOB), similar to those occurring in hereditary disorders with increase of brain gangliosides and lipid storage. Chloroquine, ergotamine, ethaverine and chronic abuse of non-narcotic analgesic drugs seem to be responsible for the drug-induced cutaneous neuro-phospholipidosis, whereas the same condition was found in two patients with metastatic bronchogenic carcinoma and malignant melanoma.

    Notes: Alternative activation of macrophages, induced by Th2 cytokines and glucocorticoids, is essential for the proper functioning of anti-inflammatory immune reactions. To this end, alternatively activated macrophages (aaMΦ) express a not yet fully unravelled set of genes including cytokines such as alternative macrophage activation-associated CC-chemokine (AMAC)-1 and pattern recognition molecules such as the scavenger receptor CD163. In order to further characterize the molecular repertoire of aaMΦ, differential gene expression was analyzed by combining subtractive suppression cloning and differential hybridization. We show here that aaMΦ induced by interleukin (IL)-4 overexpress the prototype extracellular matrix (ECM) protein fibronectin on the mRNA and protein level. This overall increase is accompanied by a shift in fibronectin splice variants from an embryonic to a mature pattern. In addition, the expression of another ECM protein, βIG-H3, is also upregulated by IL-4 in aaMΦ. In contrast to IL-4 and in line with its inhibitory effect on wound healing, dexamethasone exerts a strongly suppressive effect on fibronectin and βIG-H3 expression. In conclusion, overexpression of ECM proteins induced by IL-4 in macrophages suggests that aaMΦ may be involved in ECM deposition and tissue remodelling during the healing phase of acute inflammatory reactions and in chronic inflammatory diseases.

    Notes: Abstract: In contrast to extracellular, long chain ceramides which comprise a structural component of the epidermal water barrier, intracellular ceramides originating from sphingomyelin hydrolysis have been shown to inhibit proliferation and to induce apoptosis in different cell populations. To further elucidate the possible role of intracellular ceramides in human epidermis, two new cell-permeable ceramide analogues, N-thioacetylsphingosine (C2-Cer=S) and 4-dodecanoylamino-decan-5-ol (FS-5), were synthesized and tested for their ability to suppress cell growth and to induce apoptosis in immortalized human keratinocytes. It was shown that the well-investigated ceramide analogue N-acetylsphingosine (C2-Cer=O), as well as the new compound C2-Cer=S inhibited proliferation of HaCaT cells with half-inhibitory concentrations (IC50) of 20 μg/ml and 10 μg/ml, respectively, whereas FS-5 has been potent with an IC50〉40 μg/ml. Overall, all three ceramide analogues induced apoptosis in HaCaT cells as assessed by DNA-fragmentation using ELISA technique and in situ nick end labelling, thereby confirming the importance of ceramide signalling in keratinocytes.

    Notes: Contact allergy to oil of turpentine was reported to have become rare. However, the evaluation of standardized data of 45,005 patients tested 1992–1997 in 30 Dermatological Centers associated with the German-Austrian Information Network of Departments of Dermatology (IVDK) showed an increase in positive patch test reactions to turpentine from 0.5% during the years 1992–1995, up to 1.7% in 1996 and 3.1% in 1997. In particular, 17,347 patients tested in 1996–1997 were evaluated in detail by comparing 431 individuals with positive patch test reactions with the rest of the group found negative to turpentine. Using the so-called MOAHLFA index, the following characteristics were shown. Turpentine allergy (a) was found to be significantly less frequent in men and in patients with occupational dermatitis, (b) showed no difference in its association with atopic dermatitis, (c) patients with turpentine allergy had significantly less symptoms of the hands, more symptoms of the legs or in the face and (d) were significantly more often aged over 60 years. Also, patients sensitized to turpentine had increased rates of additional sensitizations. The definite reason for the increase in turpentine sensitization in the population tested here is not clear. Therefore, a detailed exposure analysis is necessary; the new increase in turpentine allergies may be due to popular topical remedies or household chemicals.
