    Notes: Objective:  Enamel matrix derivative (EMD) has been clinically used to promote periodontal tissue regeneration. The purpose of the present study is to clarify EMD affects on osteoblastic cells and bone regeneration.Materials and Methods:  Mouse osteoblastic cells (ST2 cells and KUSA/A1 cells) are used in culture experiments. After cells were treated with EMD, cell growth was evaluated with DNA measurement, 5-bromo-2′-deoxyurydine (BrdU) incorporation assay. Measurement of alkaline phosphatase (ALP) activity and mineralized-nodule (MN) formation, Northern blotting analysis and zymography are also performed. In addition, EMD was applied to a rat skull defect and the defect was radiographically and histologically evaluated 2 weeks after the application.Results:  EMD did not stimulate ST2 cell growth; however, it enhanced KUSA/A1 cell proliferation. Although EMD stimulated ALP activity in both the cells, ALP activity in KUSA/A1 cells was affected to a much greater degree. Corresponding to the increase in ALP activity, MN formation in KUSA/A1 cells was enhanced by EMD. EMD stimulated osteoblastic phenotype expression of KUSA/A1 cells such as type I collagen, osteopontin, transforming growth factor beta 1 and osteocalcin. EMD treatment also stimulated matrix metalloproteinase production in KUSA/A1 cells. Although the effects of EMD on osteoblastic cells depend on cell type, the overall effect of EMD on osteoblastic cells is stimulatory rather than inhibitory. Finally, EMD application to a rat skull defect accelerated new bone formation.Conclusion:  These results indicate that EMD affects osteoblastic cells and has potential as a therapeutic material for bone healing.

    Notes: Abstract Following the usual sleeve or windowresection of the trachea, reconstruction of the trachea can be carried out without difficulty in patients with aggressive thyroid cancer. When the cancer recurs locally in those patients, reoperation often yields a large tracheal defect, for which a commercially available tracheostomy cannula or a silicone-tube does not fit. By using a technique in the maxillofacial prosthetic field, it has become possible to make a new type of prosthesis which is more comfortable and acceptable to the patients with a large tracheal defect. A new type of silicone rubber prosthesis is made individually from the mold of each patient. Several appointments are required for adjustments by the chair side. It must tightly obstruct the tracheostomy stoma so that the patient can easily breathe, speak and take oral fluids. Patients can easily remove and replace this device by themselves and care of the prosthesis is easy. The practical use of this type of prosthesis in three patients is reported herein.

    Notes: Abstract Two fragments that could function as replicational cue elements were isolated from a genomic DNA digest of Aspergillus oryzae on the basis of abnormal behavior in polyacrylamide gel electrophoresis. The vector used in this study contained a scaffold-associated region of the Drosophila melanogaster ftz gene to provide nuclear retention. Neither fragment contained a yeast ARS consensus sequence or an eukaryotic topoisomerase II binding sequence. One of the fragments showed sequence homology with the mitochondrial replication origin of Candida utilis and a portion of mitochondrial DNA of Aspergillus nidulans. This plasmid carrying the cue fragment could also replicate in HeLa and NIH3T3 cells.

    Notes: Abstract Recombinant plasmids carrying a highly curved DNA structure are sometimes unstable in Escherichia coli. In order to know the underlying mechanism, several plasmids carrying one or two highly bent DNA segment(s) from the human adenovirus type 2 (Ad2) enhancer and/or origin region of phage λ replication were systematically constructed and propagated in E. coli. The highly bent DNA segments disturbed the action of DNA topoisomerases: i.e. they were shown to be able to produce an anomalously wide spectrum of linking number topoisomers that tails toward lower supercoiling with a little of the DNA actually positively supercoiled. Furthermore, bent DNA caused multimeric plasmid formation. The linking number topoisomers and multimers seemed to be intermediate topological states of the bent DNA-containing plasmids that would lead to the deletion occurring in them. The nucleotide sequence of a deletion product of a bent DNA-containing plasmid showed that the putative source of the severe topological constraint was entirely eliminated from the plasmid.

    Notes: Summary The poly(BOL) membrane prepared by “casting polymerization” exhibited an excellent fractional solute-rejection behavior in the aqueous solution, having a comparatively sharp boundary region at the molecular weight range of about 200 to 1400, together with an extremely high water permeability. These conspicuous behaviors, as well as a great capacity for water absorption, may result from some delicate arrangement of polar hydrophilic and nonpolar hydrophobic microdomains probably formed along and between the polymer chains.

    Notes: Abstract To examine the roles of histamine and diamine oxidase in the intestine after ischemiareperfusion, we measured histamine content, diamine oxidase activity, and ornithine decarboxylase activity in rat intestinal mucosa 6 hr following various periods of ischemia. In addition, mortality rates of rats after various periods of ischemia were observed. The superior mesenteric artery was occluded for 15, 30, or 60 min. Ornithine decarboxylase activity increased in the 15-, 30-, and 60-min ischemic groups compared to the shamoperated control group. In the prolonged ischemic group (60-min ischemia), both histamine concentration and diamine oxidase activity in the mucosa decreased, contributing to an increase in circulating histamine. In the 60-min ischemic group, the mortality rate of rats was 25%, which was significantly larger than the control groups. Pretreatment with aminoguanidine, which suppressed diamine oxidase activity, increased the mortality rate. These results indicate that histamine released from the intestinal mucosa has a harmful effect on rats, and diamine oxidase activity plays an important role when the small intestine is subjected to prolonged period of ischemia.

    Notes: Abstract To examine the trophic effect of epidermal growth factor on the rat small intestine, we measured diamine oxidase and ornithine decarboxylase activities in intestinal mucosa injured by methotrexate. Methotreaate was infused orally via a gastric tube at a dose of 10 mg/kg per day on 3 successive days (days 1–3). Epidermal growth factor was injected intraperitoneally at a dose of 40 μg/kg per day on 4 successive days following methotrexate infusion (days 4–7). Methotrexate caused a marked decrease in diamine oxidase activity; this decrease returned to a normal level on day 13 in controls. In rats injected with epidermal growth factor, diamine oxidase activity began to recover earlier than in the controls, and returned to a normal level on day 11. Epidermal growth factor enhanced the increase of ornithine decarboxylase activity in mucosa injured by methotrexate. When the increase of ornithine decarboxylase activity was suppressed by α-difluoromethylornithine, epidermal growth factor failed to facilitate the repair of intestinal mucosa. These results indicate that epidermal growth factor enhances intestinal repair following methotrexate infusion, and that this effect is mediated, at least in part, by ornithine decarboxylase. It is proposed that epidermal growth factor can be used clinically as a means to enhance mucosal repair of the intestine after chemotherapy with methotrexate.

    Notes: Abstract: Compared with findings in Western countries, the prevalence of reflux esophagitis in Oriental countries is estimated to be low. In this prospective study, we aimed to examine the proportion of reflux esophagitis in Japanese adults, as evaluated by endoscopy. Endoscopists were prospectively directed to grade esophageal mucosal breaks with esophagitis according to the Los Angeles Classification of Esophagitis in all subjects that underwent endoscopic examination. In total, 6010 subjects underwent endoscopic examination for evaluation of esophagitis grading from December 1996 to February 1998. The subjects included 4394 outpatients who were not receiving medication for gastrointestinal disease and 1616 subjects who visited the hospital for routine physical examinations. The overall proportion of esophagitis was 16.3%. Most of the subjects with esophagitis were classified as having grade A or B (proportion of grades A and B, 9.6% and 4.6%, respectively). The age-related proportion of esophagitis and of severe esophagitis (i.e., grades C and D) increased in females aged over 70 and in males aged over 80. Increased body mass index (partly due to decreased height caused by osteoporosis), and/or hiatal herniation, were related to the proportion of esophagitis in females aged over 70. These data indicated that reflux esophagitis is a common disease in Japan. However, severe esophagitis (grades C and D) is not common.
