    Notizen: Summary Autologous transfusions of 111indium-labelled peripheral blood lymphocytes reportedly image the pancreas of patients with Type 1 (insulin-dependent) diabetes at the time of onset. We attempted to apply this technique to the spontaneously diabetic BB/W rat. First, acutely diabetic BB/W rats, diabetes-prone BB/W rats, diabetes-resistant W-line BB/ W rats, and Wistar Furth rats were given autologous transfusions of labelled peripheral blood lymphocytes. Radioactivity recovered from the pancreas was similar in all groups. No correlation was found between the intensity of imaging and the presence or intensity of insulitis. To decrease non-specific intravascular radioactivity, acutely diabetic, diabetes-prone, and W-line rats were perfused 48 h after autologous transfusion of labelled lymphocytes. Again, the intensity of recovered activity was similar in all groups, using both macroautoradiography and numerical counting techniques. A second set of experiments studied diabetes and insulitis induced by passive transfer of concanavalin A-treated splenic lymphocytes from acutely diabetic donors. Activated lymphocytes were labelled with 111Indium and given to groups of diabetes-prone and diabetes-resistant rats. There were no differences in pancreatic localization 72–96 h after injection. Groups of diabetes-prone and diabetes-resistant rats were also given concanavalin A-activated lymphocytes and then challenged 2–10 days later with autologous transfusions of labelled peripheral blood lymphocytes. Again, no differences in organ labelling or imaging were detected. We conclude that the autologous transfusions of 111indium-labelled lymphocytes do not label or image the pancreas of the BB/W rat.

    Notizen: The ferredoxin component (ferredoxinBPH) of biphenyl 2,3-dioxygenase was purified to homogeneity from crude cell extract of Pseudomonas sp strain LB400 using ion exchange, hydrophobic interaction and gel filtration column chromatography. The protein was a monomer with a molecular weight of 15000 and contained 2 gram-atoms each of iron and acid-labile sulfur. Ultraviolet-visible absorbance spectroscopy showed peaks at 325 nm and 460 nm with a broad shoulder around 575 nm. The spectrum was partially bleached in the visible region upon reduction by reductaseBPH with NADPH as the source of electrons. Electron paramagnetic resonance spectrometry showed no signals for the oxidized protein. Upon reduction with sodium dithionite, signals with gx = 1.82, gy = 1.92 and gz = 2.02 were detected. These results indicate that the protein contains a Rieske-type (2Fe-2S) iron-sulfur center. FerredoxinBPH was required for the oxidation of biphenyl by the terminal oxygenase component of the enzyme and is probably involved in the transfer of reducing equivalents from reductaseBPH to the terminal oxygenase during catalysis.

    Notizen: Summary The effects of MSG on PRL cell activity in hamsters was assessed by injecting either MSG (8 mg/g body weight) or saline into male and female hamsters on day 8 of the neonatal period. One-hundred and three days later, the anterior pituitaries were removed for the analysis of PRL synthesis and storage. Serum PRL levels were also determined by radioimmunoassay as an index of in vivo PRL secretion. Blood levels of PRL were decreased by 74% in female and 88% in male hamsters treated with MSG as compared with vehicle controls. In females, glutamate induced a drop in PRL storage (90%) and synthesis (82%) while in male animals glutamate caused a non-significant decrease in PRL storage (48%) and synthesis (31%) as compared with controls. These results indicate that MSG-induced arcuate nucleus lesions inhibit PRL cell activity in the hamster.

    Notizen: Abstract To determine whether the aerobic capacity of tissues required for growth specifically reflects growth rates, we monitored the activities of key enzymes of oxidative, glycolytic and amino acid metabolism in muscle, liver and intestine of Atlantic cod (Gadus morhua) growing at different rates. Fish were maintained individually in small tanks at 10°C and fed on rations that allowed growth rates ranging from-0.6 to 1.6% per day. The correlation between growth rate and muscle enzyme activity was pronounced for the glycolytic enzymes (LDH, PFK and PK). The activities of glycolytic enzymes were more than four times higher for fish having higher growth rates compared to those that did not grow. Mitochondrial enzyme (cytochrome c oxidase, citrate synthase and β-hydroxyacyl-CoA dehydrogenase) activities remained unchanged in fish with positive growth. The liver seems to respond to requirements of growth by an increase in size. In the liver, the activities of the enzymes of amino acid metabolism expressed as units · μg DNA-1 specifically increases with growth rate. In contrast to the two other tissues, the specific activities of mitochondrial enzymes in the intestine increased with growth rate while the relative mass of the intestine remained constant. Intestinal cytochrome c oxidase activity increased from a minimum of about 2 to more than 8 units · g intestine-1. Cytochrome c oxidase activity increased in parallel with the food conversion efficiency. This suggests that the aerobic capacity of the intestine may initially limit the rates of digestion and growth in this species.

Solutions are obtained in primitive variables using a high-order finite element approximation on unstructured grids. Two general-purpose error estimators are developed to analyse finite element solutions and to determine the characteristics of an improved mesh which is adaptively regenerated by the advancing front method. The adaptive methodology is validated on a problem with a known analytical solution. The methodology is then applied to heat transfer predictions for two cases of practical interest. Predictions of the Nusselt number compare well with measurements and constitute an improvement over previous results. © 1997 John Wiley & Sons, Ltd.

    Notizen: It has been established previ- ously that the maintenance of expression of pre-spore-specific genes of Dictyostelium discoideum is prevented by the translational inhibitor cyclohex- imide. The drug had no effect upon the level of transcripts of the other genes examined, prestalk-specific or cell type-nonspecific. However, the interpretation of this result is open to question, because of possible nonspecific effects of cyclo-heximide. We have now characterized the cellular specificity and temporal profiles of mRNA accumu- lation of additional Dictyostelium cDNA clones, and have examined other inhibitors of in vivo protein synthesis. Four structurally and mechanistically distinct translational inhibitors each prevented the reaccumulation of prespore transcripts in cyclic AMP-primed, disaggregated amoebae. These results establish the importance of developmental protein synthesis in the accumulation of prespore gene transcripts. Nuclear run-on transcription assays were used to learn whether protein synthesis is required primarily for mRNA synthesis or transcript stability. A transcriptional time course first demonstrated that the abundance of these cell-specific transcripts during development mirrors their rates of synthesis. Significantly, the protein synthesis requirement of the prespore genes examined also occurs at the level of mRNA transcription, implying the existence of one or more developmentally regulated transcriptional activators.
