Notes: Summary Streptomyces sp. strain EC10 degraded efficiently the hemicellulose fraction of wheat straw. Three forms of β-xylanases detected in the culture filtrate were purified by precipipation with ammonium sulphate, chromatography on DEAE-Sephadex A-50 and gel filtration on Sephadex G-100. The three purified enzymes (X ia , X ib and X′ ii ) were homogeneous by polyacrylamide gel electrophoresis. The enzymes were typical non-debranching endo-β-xylanases (1,4-β-d-xyla xylanohydrolases; E.C.3.2.1.8) with respective relative molecular weights of 32,000, 22,000 and 21,000 and isoelectric points of 6.8, 8.9 and 5.2. The enzymes were highly specific for xylans and showed optimal activity at pH 7.0–8.0 and 60°C. The preparations were completely free from cellulolytic activity (endoglucanase) and showed high thermal stability. No synergy between the three enzymes was detected for complete xylan hydrolysis of deacetylated arabino- and glucuronoxylans.

Notes: Summary Phanerochaete chrysosporium was proposed for the decolorization and degradation of chlorinated compounds (AOX) present in the E1 stage effluent of the Kraft process. We observed that pellets of the fungus inoculated in the E1 effluent, not supplemented with nutrients, strongly absorb colour and AOX. The data were in agreement with a model describing a multilayer adsorption.

Notes: The Modified Simplex Method (MSM) coupled with contour diagrams is used to determine the rate constants of a kinetic scheme involving three sequential second order processes.Reaction between glutathione and the dithiocarbamate fungicide tetramethylthiuram disulfide produces the oxidized form of glutathione which is then reduced in the presence of NADPH. The concentration of the reducing agent is monitored as a function of time. In the absence of simplifying assumption a closed form solution of the rate equations does not exist. Computed curves of NADPH concentration versus time were generated using the software SIMULCIN.The rate constant values are optimized by the MSM procedure to obtain the best agreement between calculated and experimental data. The significance, validity limits of the estimated rate constants and the relevance of these in vitro data to previous in vivo studies are discussed.
