    Notes: Summary The ability of three chelating agents, dimercaptopropanesulphonate (DMPS), dimercaptosuccinic acid (DMSA), and diethylenetriaminepentaacetic acid (DTPA), to reduce the retention of platinum in the kidneys was tested in male Sprague-Dawley rats treated 24 h previously with Cisplatin. DMPS and DMSA, when given as four daily doses of 1 mMol/kg, increased the urinary excretion from 10.1%±1.2% to 13.6%±1.3% and 13.5%±2.6%, respectively, but only DMSA caused a small but significant reduction in the kidney platinum content. DTPA was totally ineffective. It is concluded that none of these agents is likely to be useful for the reduction of Cisplatin-induced renal toxicity in the clinical situation.

    Notes: Abstract The influence of an acute dose of lead nitrate or cadmium chloride on the activity of glutathione-S-trans-ferase (GST) was investigated in rats. CdCl2 (10 Μmol/kg) did not influence the enzyme activity in either organ. In contrast Pb(NO3)2 (100 Μmol/kg) caused a significant increase of GST activity in both organs. SDS polyacrylamide gel electrophoresis (SDS-PAGE) showed that in liver the activity enhancement is due mainly to the induction of the isoenzyme GST 7-7, while in the kidneys the activity of all the isoenzymes is increased.

    Notes: Abstract The toxicity of tetrasodium-ethylenediaminetetraacetate (Na-EDTA) was investigated in normal rat kidney cells (NRK-52E, American Type Culture Collection) in culture. Cell death and reduced colony-forming ability were observed at Na-EDTA concentrations 〈100 μM, that is at concentrations lower than that required to chelate all the Ca2+ in the growth medium. No toxicity was observed when cells were exposed to 100 μM Zn- or Fe-EDTA, but the same concentration of Cu-EDTA was as toxic as Na-EDTA. Continuous exposure of the cell cultures to 5 μM Na- or Zn-EDTA for up to 7 weeks yielded no indications of toxicity.

    Description / Table of Contents: Zusammenfassung Die Wirkung von intravenös verabreichtem HgCl2 auf die renale Ausscheidung von alkalischer Phosphatase (AP) und Leucinaminopeptidase (LAP) wurde an Ratten untersucht. Am ersten Tag nach Hg-Injektion erfolgte ein Anstieg der Enzymausscheidung, der von einem Schwellenwert von 0.44 mg Hg/kg ausgehend eine lineare Abhängigkeit von der Hg-Dosis aufwies. Am zweiten Tag wurde ein statistisch signifikanter Effekt bereits nach 0.25 mg Hg/kg beobachtet. Nach Dosen von 0.75 mg Hg/kg oder mehr fand ein Abfall der Enzymaktivität unter die Kontrollwerte statt, der mehr als 4 Tage anhielt. Behandlung mit 2,3-Dimercaptopropansulfonat (DMPS) bewirkte eine Normalisierung der AP-Ausscheidung. Eine Wirkung auf die LAP-Exkretion wurde nur bei baldiger Verabreichung von DMPS beobachtet. Das gleiche gilt für den Effekt von DMPS auf die Hg-induzierte Letalität. Die Nützlichkeit einer LAP-Bestimmung im Urin zur Abschätzung einer Hg-Inkorporation wird diskutiert.
    Notes: Abstract The effect of intravenous injections of HgCl2 on the renal excretion of alkaline phosphatase (AP) and leucine aminopeptidase (LAP) was investigated in rats. On the first day after Hg enzyme excretion showed a linear rise with the Hg dose from a threshold value of 0.44 mg Hg/kg. On the second day a statistically significant effect was seen already after 0.25 mg Hg/kg. After doses of 0.75 mg/kg or more a decrease of enzyme activity below control values occurred which persisted for more than 4 days. Treatment with 2,3-dimercaptopropansulfonate (DMPS) brought about a normalization of AP excretion. An effect on LAP excretion was observed only with early treatment. The same holds for the effect of DMPS on Hg-induced lethality. The usefulness of a measurement of LAP excretion for estimating the exposure to inorganic mercury is discussed.

    Notes: The effects of some new chelating agents on the cadmium burden of CHO cells in culture were investigated. The chelators were sodium-N-(4-methoxybenzyl)-D-glucamine-dithiocarbamate (MeOBG-DTC), sodium-N-benzyl-D-glucaminedithiocarbamate (BG-DTC) and di-isopropylmeso-2,3-dimercapto succinate (DiP-DMSA). The results were compared with the effect of the well known dimercaptopropanol (BAL). The derivates of dithiocarbamate are much less toxic than DiP-DMSA and BAL. All chelators effectively prevent Cd uptake into the cells. Mobilization of intracellular Cd, however, is more effective by the DTC-derivatives than by DiP-DMSA or BAL. Within the cell the major fraction of Cd after 48 hours incubation is found in the nuclei and cytosol and very little in the peroxisomes. The chelating agents remove the metal mostly from nuclei and cytosol. Incubation of the cells with cadmium leads to the induction of a Cd binding protein of an apparent molecular weight of 12500 Da, presumably metallothionein. MeOBG-DTC is more effective in removing the metal from this protein than BG-DTC.

    Notes: The uptake and binding of 59Fe, 67Ga and 239Pu complexed with citrate of transferrin (Tf) and of 125I-labelled Fe-Tf by human lymphoblasts (WI-L2 cells) have been studied. Uptake kinetics of 59Fe-Tf and [125I]-Tf point to internalization by receptor mediated endocytosis. 67Ga binding and uptake is always less. This may be explained by a lower affinity of Ga-complexes for the cell surface. Factors which influence Fe uptake have a similar effect on Ga. 239Pu uptake and binding, however, are different, especially in that Tf does not stimulate 239Pu uptake and may actually decrease it.
