    Notes: Summary The innervated face of electrocytes in the main electric organ of Electrophorus electricus L. was examined by light microscopy, both conventional and with Nomarski contrast, and by transmission and scanning electron microscopy. Acetylcholinesterase cytochemistry was used in the demonstration of the greater density of synapses over the caudal papillae. The various techniques contributed to a better understanding of the distribution and form of papillae and synapses at the posterior face of the electrocyte. Caudal papillae are longer and thinner than those at the rostral face, but it was not possible to recognize a different type sometimes referred to in the literature as small papillae. The contact of nerve endings with the electrocyte seems to be made predominantly on the terminal half of caudal papillae, however a smaller number occur elsewhere on the posterior face. Synaptic terminals frequently appear as round profiles, but may be also elongated, with or without bulges, usually occupying a depression, and separated from the post-synaptic membrane by a space of 60–100 nm, where an expansion may be found.

    Notes: The mature annelid cuticle contains orthogonally oriented collagen in a matrix capped superficially by a dense epicuticle with external corpuscles. The underlying epidermis is a simple columnar epithelium with two major cell types, mucous-secreting cells which secrete through channels in the cuticle to the exterior of the worm, and “supportive” cells which presumably produce and increase the cuticle by secreting into it.The structures of supportive cells, previously interpreted as specialized for establishing interfibrillar collagen order, are revealed by glutaraldehyde fixation as common cellular components without the qualities deemed useful to align collagen. Cell processes which penetrate and sometimes pass completely through the cuticle are not stable, not in geometric order, and lack cilia-like structure. Cilia, unlike the ubiquitous cellular processes, are highly restricted to regions of the epidermis with specialized functions. Cellular control, or other control, of collagen fibrillogenesis remains unestablished.

    Notes: Appearance of collagen fibrils in the cuticle was seen by electron microscopy to be preceded by fonnation of a finely filamentous matrix material. At first, the fine filaments of the matrix are unorganized. However, signs of orthogonal ordering soon appear in the most superficial portion of the cuticle, and subsequently appear more basally and closer to the underlying epidermis. Meanwhile, fibrils of different staining properties and identifiable as collagen begin to be deposited in the superficial portion of the cuticle, the same region which first showed organized fine filaments. Then, like the fine filaments before them, the collagen fibrils polymerize more basally. Collagen appears to polymerize on the preformed skeleton of fine filaments as though the fine filaments caused the collagen to assemble. Neither the polymerization nor ordering of collagen fibrils seems to require direct cellular intervention but occur first in that portion of the cuticle which is furthest away from the underlying epidermis. The fine filaments may be self ordering, extracellular macromolecules which in turn determine the polymerization of collagen fibrils.

    Notes: Summary A polymerase chain reaction (PCR) technique was developed and evaluated for the detection of flaviviruses. A set of sense and antisense oligomeric DNA primers were constructed from nucleotide sequences of the conserved region of the genome of several different flaviviruses. Virus specific complementary DNA (cDNA) was prepared by reverse transcription of total RNA extracted from infected cell cultures. Amplified cDNA was identified by nucleic acid hybridization with specific oligomeric internal probes. Various conditions, such as number of cycles and annealing temperature were examined to optimize the detection of viral RNAs from infected cell cultures. Slot blot hybridization with a radioative probe was used to evaluate the sensitivity of PCR amplification. The PCR amplified RNA sequences of dengue 2 (DEN-2), West Nile (WN), St. Louis encephalitis (SLE) and Kunjin (KUN) virus and detected 0.1 to 1 pg of viral RNA. Japanese encephalitis (JE), Yellow Fever virus (YF), DEN-1, 3, and 4 viruses were not amplified. The more frequent occurrence of mismatches in the 3′ primer binding site may explain the failure to amplify cDNA of these viruses.

    Notes: Summary.  A recently described DNA vaccine for dengue (DEN) type 2 was shown to elicit high levels of neutralizing antibodies in mice. The vaccine candidate consists of the PreM and 92% of the envelope genes of DEN 2 New Guinea C strain. We further evaluated this DNA vaccine candidate by examining the effect of immuno-stimulatory CpG DNA motifs on antibody response and by studying the protective efficacy of the vaccine. The results showed that CpG motifs present in pUC 19 significantly improved the antibody response to a suboptimal dose of 3.1 μg of the DEN DNA vaccine. In a lethal mouse intracerebral challenge model, the vaccine provided a significant level of protection. Sixty percent of the mice immunized with the DEN DNA vaccine plus pUC 19 survived the challenge compared to only 10% in the control group that received vector plus pUC. These studies illustrate that nucleic acid immunization is a viable approach to developing a DEN vaccine and that immuno-stimulatory CpG DNA motifs can be used to lower the minimum dose required to produce an antibody response.

    Notes: Summary The dermal melanophores ofFundulus heteroclitus L. have been investigated by light and electron microscopy with the purpose of revealing the mechanisms controlling pigment migration. As predicted by earlier studies, the nerve endings of a double innervation were found adjacent to and in synaptic relation to the melanophore surface. Not expected were the large number of small pits or invaginations present in the cell surface. These appear identical to the so-called micropinocytotic vesicles found generally in cells of the vascular endothalium and smooth muscle. In chromatophores they are more reasonably interpreted as receptor sites for neurohormones than as uptake and transport mechanisms. Observations made on the kinetics of pigment migration within the processes of these melanophores indicate that the granules move along relatively fixed channels arranged parallel to the long axes of the processes. Examined at fine structure levels, the zones of cytoplasm around these channels are found to be populated by microtubules about 225 Å in diameter aligned parallel to the direction of pigment movement. These long slender elements are present in the processes regardless of whether the melanin is concentrated in the cell center or dispersed. It is reasoned from these and other observations that the microtubules function as cytoskeletal elements which help maintain the extended form of the melanophore arms and at the same time define the channels in which the pigment moves. The possible role of the tubule in generating the motive force for pigment migration is discussed.
