    Notizen: Right-angle wiggling (RAW) electron paramagnetic resonance (EPR) spectroscopy is introduced. In this two-dimensional pulse EPR technique the orientation of the external magnetic field B0 is varied during the pulse sequence. For this purpose an additional sinusoidal field of variable amplitude ΔB0⊥ is applied perpendicular to the orientation of B0. RAW-EPR is an alternative and experimentally much less demanding technique to right-angle spinning for performing anisotropy-resolved EPR experiments [Sierra and Schweiger, Mol. Phys. 95, 973 (1998)]. It can be applied to improve the resolution of EPR spectra of orientationally disordered systems or to separate overlapping single-crystal EPR spectra, and to facilitate an unambiguous interpretation of these spectra. A detailed theoretical description of RAW-EPR is given. Model calculations for systems with axial and orthorhombic symmetry, as well as a number of RAW-EPR experiments on paramagnetic systems with anisotropic interactions are presented. © 2001 American Institute of Physics.

    Notizen: The extremely heat-stable glutamate dehydrogenase (GluDH) from Pyrococcus woesei was crystallized by the hanging-drop vapour-diffusion method. Crystals suitable for X-ray crystallographic investigations were obtained using polyethylene glycol (PEG) 4000 and ammonium acetate as precipitating agents. The crystals obtained diffract to a resolution of 2.8 Å, have a prismatic shape and grow up to 0.5 mm in their maximal dimension. They belong to the triclinic system (space group P1; a = 90.9, b = 92.8, c = 107.1 Å, α = 69.1, β = 80.7, Vγ = 65.0°) with a unit-cell volume of 765052 Å3 which accommodates one GluDH hexamer of 276 kDa. The averaged density of the crystal determined by Ficoll-gradient centrifugation is 1.15 g cm−3, which corresponds to a molecular mass of 255 kDa in the unit cell. A native data set has been collected on an MAR image-plate system using Cu Kα radiation. The completeness of the data set in the range 316–3 Å is 74%, and contains 64% of the data in the outer shell (3.4–2.8 Å), with an average Rmerge value of 9%. Calculation of self-rotation functions revealed the 32 symmetry of the hexamer; 3 non-crystallographic twofold axes were found at a distance of 120° in a plane perpendicular to the non-crystallographic threefold axis.

    Notizen: Summary Growth of an extremely thermophilic archaeon, Pyrococcus woesei, at 90°C in a 2–1 fermentor was significantly enhanced by gassing with N2/CO2 (95%/5%). Both growth and α-amylase activity were also positively influenced by increasing the agitation speed up to 1200 rpm under continuous gassing at 0.2 vvm. However, increasing the agitation speed to 2400 rpm led to decreases in the maximum cell concentration and α-amylase activity. Fed-batch cultivation resulted in increases in the specific growth rate, maximum cell concentration and α-amylase activity. Although the latter two parameters were higher when the broth was supplemented with both starch and concentrated medium, the specific growth rate was relatively smaller. Cultivation in a dialysis reactor gave a cell concentration of 2 × 109 cells/ml, which represents a 2.8-fold increase over that obtained in ordinary batch cultivation. This increase in the cell concentration was accompanied by a 5.2-fold increase in α-amylase activity.

    Notizen: The usual method of calculating functionality is to divide the molecular weight by the equivalent weight. Because of the uncertainty of molecular weight determinations in the range 1000-20,000 a more precise method is needed. Several authors have published work concerning the determination of the extent of reaction at the gel point through the functionalities of the reactants. It occurred to us that this method could be reversed. We chose Stockmayer's treatment, with some changes, to calculate the average functionality of carboxyl-terminated polybutadiene (CTPB): (PAPB)gel = (fE - 1)-1 (gE - 1)- where fE and gE are the weighted average functionalities of all molecules bearing the reactive groups A and B, and PA and PB are the fractions of initially present groups that have reacted. Two systems with an epoxide and glycerol as curing agents were investigated. The influence of dilution was investigated. Nonfunctional polybutadiene did not interfere with the accuracy of the determination of carboxyl functionality. By changing the ratio of epoxide to COOH groups from 0.6 to 1.4 it was shown that the calculated functionality remained constant. Weight-average molecular weights for three CTPB samples were calculated from the functionality and the equivalent weight. They were compared with those obtained from intrinsic-viscosity measurements. The precision of the functionality numbers is believed to be ±1%.

    Notizen: Just how far reactions can go after gelation during the cure of telechelic prepolymers has been a debatable point for some time. Utilizing a recently devised method,7 the curing reactions of some telechelic prepolymers were followed after the gel point. Extents of reaction above 90% occurred only in systems of average functionality close to two, functionality being the average number of reactive groups per molecule. Among systems of higher functionality, maximum extents of reaction of about 70% were most common. The final extent of reaction was only a few per cent above the extent of reaction at the gel point. The maximum extent of reaction varied with the concentration of reactive species and the relationship was a linear one at each functionality of the system. The data were consistent with PA2/r = {0.88/[(h - 1)(j - 1)]} + 0.10 where PA is the fraction of prepolymer reactive groups initially present which have reacted, r is the ratio of the initial number of crosslinking groups to prepolymer reactive groups, and h and j are weighted average functionalities of the two reactants. It is suggested that the limiting factor in defining the final extent of reaction in these systems is the accessibility of reactive groups as determined by solid geometry rather than thermodynamics or reaction kinetics. The final extent of cure is regularly dependent on functionality and one cannot regard functionality and maximum extent of reaction as independent variables.
