    Notes: This study examined the role of the diagonal band of Broca (DBB) in drinking behaviour and vasopressin release. Adult male rats were anaesthetized (pentobarbital 50 mg/kg) and received DBB injections of either ibotenic acid (0.5 µl of 5 µg/µl) or vehicle (0.5 µl of phosphate-buffered saline). Although baseline drinking and urine output were not affected, drinking to 30% polyethylene glycol (MW 8000; 1 ml/100 g s.c.) and angiotensin II (0, 1.5 and 3.0 mg/kg s.c.) were significantly increased in ibotenic acid in phosphate-buffered saline (DBBX) rats. Drinking to hypertonic saline (0.9, 4 and 6%; 1 ml/100 g), and water deprivation were not significantly affected. DBBX rats had significantly lower basal heart rates than controls but the cardiovascular responses to infusions of angiotensin II (100 ng/kg/min i.v. for 45 min) were not affected. DBBX rats had significantly higher basal vasopressin, but angiotensin-stimulated vasopressin release was not significantly different. Although the DBB is not involved in basal water intake, it is involved in dipsogenic responses to hypovolemic stimuli and possibly basal autonomic function and basal vasopressin release.

    Notes: A solution-hybridization S1-nuclease protection assay was used to evaluate the expression of messenger RNAs for the activin βA subunit and type II activin receptor in adult rat brain. Results indicate the presence of βA subunit mRNA in both hypothalamus and brainstem, with approximately two-fold higher levels in brainstem. Levels of activin type II receptor mRNA were similar in the hypothalamus of young virgin and 15-day lactating females, and in females in which pups were removed after a 5-day lactation period. Male rats castrated prepubertally (30 days p.n.) had approximately 220% higher (P〈0.05) hypothalamic activin type II receptor mRNA levels than postpubertal, 3-month old age-matched sham controls. Two month treatment of castrate rats with estradiol (200 ng/g, i.p. every 2 days) reduced hypothalamic activin type II receptor mRNA expression to control levels; the same dose of testosterone had no effect. The expression of the hypothalamic activin type II receptor gene may be estrogen-regulated in vivo.

    Notes: Experiments examined release of atrial natriuretic factor (ANF), measured by radioimmunoassay, from acutely prepared explants of rat hypothalamus maintained in vitro by intra-arterial perfusion of artificial cerebrospinal fluid. Perfusates collected from intact preparations contained 6.1 ± 0.6 pg (mean ± SEM) of ANF per 2-min sample. Following a 3-min infusion of noradrenaline (60 μM), ANF release increased significantly (P〈0.05) to 11.4±1.4 pg/sample. Media collected from hypophysectomized preparations showed the same basal ANF release (6.8 ± 0.9 pg/sample) as intact preparations, but demonstrated no significant increase after noradrenaline infusions. Levels of spontaneous ANF release were not appreciably affected by the absence of the paraventricular nuclei and/or the anteroventral third ventricle area.Extracted material from the perfusate by reverse-phase high performance liquid chromatography revealed two main peaks of immunoreactive ANF: a small molecular weight form that coeluted with synthetic ANF (99–126) and with similar biological activity in a radioreceptor assay, and a larger molecular weight form with the same elution profile as the ANF (1–126) prohormone.These observations indicate that the ANF released from perfused rat hypothalamic explants contains distinct contributions from the hypothalamus (sites undetermined) and the neurointermediate lobe.
