    Description / Table of Contents: Zusammenfassung An der kauterisierten Kaninchen-Cornea wird die Frage geprüft, ob das Virostaticum 5-Äthyl-2′-desoxyuridin (ÄDU) die Regeneration der geschädigten Hornhaut beeinflußt. Es zeigt sich, daß ÄDU nach subconjunctivaler Injektion und gleichzeitiger lokaler Applikation im Vergleich zu den Kontrollen mit 0,9 % NaCl-Lösung die Regeneration nicht verzögert. In gleicher Weise übt auch das mituntersuchte Thymidin keinen hemmenden Einfluß auf die Regeneration aus. Dagegen hemmt unter den gleichen Versuchsbedingungen 5-Jod-2′-desoxyuridin (IDU) die Regeneration der geschädigten Hornhaut. ÄDU, das nach unseren Versuchen in die DNS von Helazellen sowie von Corneazellen eingebaut wird, hemmt gemäß der vorliegenden Untersuchungen offensichtlich nicht die Zellvermehrung der stromal geschädigten Cornea. Unsere Ergebnisse stützen somit die Auffassung, daß der virostatische Wirkungsmechanismus von ÄDU, im Gegensatz zu dem von IDU, nicht über die Mitosehemmung der Wirtszelle verläuft.
    Notes: Summary We have already shown in experiments in vitro and in vivo that 5-ethyl-2′-deoxyuridine (EDU) is a potential inhibitor of the DNA viruses like herpes, vaccinia and Aujeszky. Following this experimental activity the effectiveness of EDU in the treatment of herpetic and metaherpetic diseases has also been demonstrated. Clinically EDU was applied as eye drops (0.1 to 0.2% solution) and/or injected subconjunctively (0.5% solution). The object of the present investigations was to determine, whether EDU has any deleterious effects on the regeneration of electrically cauterised rabbit cornea. For the clinical application of EDU, these experiments are important in the respect that prior to clinical treatment herpetic ulcers and eruptions of the cornea and of the mucous membranes of the eye are present. The studies have now shown that EDU when injected subconjunctively (0.3cm3, 0.5% twice daily) and applied locally (4 times a day) on 4 consecutive days does not inhibit the corneal regeneration in comparison to the animals accordingly treated with 0.9% NaCl solution. In the same way thymidine also did not exersise inhibitory effect on the corneal regeneration. Contrary to that under our conditions 5-Iodo-2′-deoxyuridine (IDU) potentially inhibited the regeneration when applied as a 0.1% solution. Ethyldeoxyuridine which in our previous studies is incorporated into the DNA of Hela cells and of the rabbit corneal cells, according to these investigations obviously does not inhibit the replication of the stromal cells. These results support our view that the mode of virostatic activity of 5-ethyl-2′-deoxyuridine, in contrast to that of the 5-Iodo-2′-deoxyuridine, is not related to the inhibition of the mitosis of the host cells.
