    Notes: The regulation of nitrogen fixation in Azospirillum brasilense is very complicated, and it responds to exogenous fixed nitrogen or a change of oxygen concentration. This regulation occurs at both transcriptional and posttranslational levels. Unlike regulation seen in Klebsiella pneumoniae, transcription of nifA does not require NTRB/NTRC in A. brasilense and the expression of nifHDK is controlled by posttranslational regulation of NIFA activity. Addition of NH+4 or a shift from microaerobic to anaerobic conditions also causes a rapid loss of nitrogenase activity in A. brasilense. This posttranslational regulation of nitrogenase activity involves the DRAT/DRAG regulatory system, which is similar to that of Rhodospirillum rubrum. Both DRAT and DRAG activities are regulated in vivo, but the mechanisms for their regulation are unknown.

    Notes: Summary The role of the two glnA linked genes, glnL and glnG, in regulation of glnA expression and nitrogen metabolism in Escherichia coli has been studied by analysis of 131 glnL and 164 glnG genetically characterized mutations. A comparison of phenotypes with genetic position was performed for all mutations in glnL and glnG. We determined the ability of mutants to derepress GS, to grow on a variety of nitrogen sources in the absence of glutamine, and to suppress the glutamine requirement caused by a glnF mutation. The results indicate that both glnL and glnG products mediate negative regulation of glnA. The glnG product, but not that of glnL, is required for derepression of glnA. Both glnL and glnG products are required for positive regulation of gene products involved in the utilization of poor nitrogen sources. In each gene, point mutations were found which confer a phenotype dramatically different than that caused by insertion mutations. These point mutations fall into several frequently occurring classes. The phenotypes of these classes suggests that each gene product has bifunctional regulatory properties. Further, each class tends to be located in only a portion of a gene suggesting that the region encoding each function is genetically distinct. The role of glutamine synthetase in the regulation of glnA expression was investigated using two-dimensional polyacrylamide gel electrophoresis on extracts of 38 GlnA- mutants. Results of this analysis argue that glutamine synthetase is not structurally involved in the regulation of glnA expression.

    Notes: Summary Nitrogen fixation activity in the photosynthetic bacterium Rhodospirillum rubrum is controlled by the reversible ADP-ribosylation of the dinitrogenase reductase component of the nitrogenase enzyme complex. This report describes the cloning and characterization of the genes encoding the ADP-ribosyltransferase (draT) and the ADP-ribosylglycohydrolase (draG) involved in this regulation. These genes are shown to be contiguous on the R. rubrum chromosome and highly linked to the nifHDK genes. Sequence analysis revealed the use of TTG as the initiation codon of the draT gene as well as a potential open reading frame immediately downstream of draG. The mono-ADP-ribosylation system in R. rubrum is the first in which both the target protein and modifying enzymes as well as their structural genes have been isolated, making it the model system of choice for analysis of this post-translational regulatory mechanism.

    Notes: The circular dichroism spectra of many natural DNAs and double-stranded synthetic polynucleotides were obtained. The eight first-neighbor contributions to the CD spectra of a DNA have been extracted from these data. Therefore, the CD spectrum for any DNA with known first-neighbor frequencies may be easily calculated. For a natural DNA the CD spectrum may be approximated by assuming the first-neighbor frequencies have the most probable values consistent with the base composition. Under favorable conditions, the measured CD spectrum can be used to determine thirteen of the sixteen first-neighbor frequencies of a DNA to ± 0.02 mole percent. The TG, CA, and TA first-neighbor cannot be unambiguously resolved by our method. The accuracy of the first-neighbor frequency analysis depends on the number of different first-neighbors present in the DNA and the extent to which they differ from the most probable value.The extinction coefficient at 260 nm and the base composition can also be calculated from the CD spectrum.
