    Notes: Abstract: The effect of taurine on the release of [3H]acetylcholine ([3H]ACh) and [3H]-γ-aminobutyric acid ([3H]GABA) from preloaded locust synaptosomes has been studied. Veratridine (100 μM) and K+ (100 mM) both evoked [3H]ACh release and this was reduced in a concentration-dependent manner by taurine (5, 10, and 20 mM). In contrast to this, veratridine induced no observable release of [3H]GABA, and the response to K+ was slight. In the presence of taurine, however, a concentration-dependent enhancement of [3H]GABA release was observed. Since nipecotic acid (1 mM), an inhibitor of neuronal GABA uptake, also revealed [3H]GABA release induced by veratridine, it is suggested that both this effect and that of taurine are due to prevention of GABA reuptake. These results suggest that taurine may act as a neuromodulator in insects.

    Notes: Abstract: The effect of taurine (2-aminoethanesulphonic acid) on 45Ca2+ accumulation in resting and depolarised synaptosomes obtained from the locust Schistocerca americana gregaria was studied. Taurine reduced 45Ca2+ accumulation in resting synaptosomes, and this effect was more pronounced when synaptosomes were depolarised with either high [K+] or veratridine. Veratridine-induced 45Ca2+ accumulation was not affected by either γ-aminobutyric acid or leucine, but was reduced by both verapamil and tetrodotoxin.

    Notes: Epidermolysis bullosa acquisita (EBA) is an acquired subepidermal bullous disease characterized by IgG autoantibodies directed against type VII collagen, the major component of anchoring fibrils. The classical phenotype of EBA is a non-inflammatory, mechanobullous disease resembling the dystrophic forms of inherited epidermolysis bullosa. Mucous membrane involvement is frequent but usually mild. We report a 1-year-old girl suffering from IgA-EBA, who presented with an initial eruption of disseminated urticarial lesions and tense blisters of the skin but subsequently developed severe oral and ocular lesions reminiscent of cicatricial pemphigoid. Direct immunofluorescence of the skin and buccal mucosa revealed linear IgA and C3 at the basement membrane zone (BMZ). IgA anti-BMZ autoantibodies stained the dermal side of salt-split skin by indirect immunofluorescence and recognized a dermal protein of 290 kDa co-migrating with type VII collagen by immunoblotting. Direct and indirect immunoelectron microscopy revealed IgA deposits overlying the anchoring fibrils. The ocular involvement led to total blindness in spite of intense treatment. This case of childhood IgA-EBA is particularly striking because of the cicatricial pemphigoid phenotype with severe ocular involvement which resulted in blindness. It reinforces the necessity to use modern immunological methods to classify autoimmune bullous diseases in order to allow early and appropriate treatment.

    Notes: Migraine is an episodic pain disorder whose pathophysiology is related to deficiency of serotonin signaling and abnormal function of the P/Q-type calcium channel, CACNA1A. Because the relationship of the CACNA1A channel to serotonin signaling is unknown and potentially of therapeutic interest we have used genetic analysis of the Caenorhabditis elegans ortholog of this calcium channel, UNC-2, to help identify candidate downstream effectors of the human channel. By genetic dissection of the lethargic mutant phenotype of unc-2, we have established an epistasis pathway showing that UNC-2 function antagonizes a transforming growth factor (TGF)-β pathway influencing movement rate. This same UNC-2/TGF-β pathway is required for accumulation of normal serotonin levels and stress-induced modulation of tryptophan hydroxylase (tph) expression in the serotonergic chemosensory ADF neurons, but not the NSM neurons. We also show that transgenic expression of the migraine-associated Ca2+ channel, CACNA1A, in unc-2 animals can functionally substitute for UNC-2 in stress-activated regulation of tph expression. The demonstration that these evolutionarily related channels share a conserved ability to modulate tph expression through their effects on TGF-β signaling provides the first specific example of how CACNA1A function may influence levels of the critical migraine neurotransmitter serotonin.

    Notes: The calorific value of the three-spined stickleback, Gasterosteus aculeatus, varied with its size, sex and the season. Fish of 16 mg dry weight had a value of 1950 cal/g while for fish of 313 mg the value was 470 cal/g. The califoric value of gravid females was 23% above that of males of comparable size. The califoric value of a tropical cichlid, Tilapia nilotica, with a dry weight of 11 g was 4300 cal/g, the calorific values of the tissues ranged from 2500 cal/g for bone to 5190 cal/g for muscle.

    Notes: 1. The plasma half-life and apparent volume of distribution of phenazone (antipyrine) was studied in twenty-six normal adults (20–40 years of age) and twenty-six geriatric patients.2. A wide range of values for plasma half-life was recorded in both groups (young 7·7–19·9 h, elderly 9·5–30·1 h) but the mean plasma half-life for the geriatric group (16·8 h; s.e.m. = 5·9) was significantly longer (P 〈 0·01) than the normal adult group (12·5 h; s.e.m. = 3·2).3. No difference in plasma half-life was noted between sexes in either the geriatric or control groups.4. Doubling the dose of phenazone from 10 to 20 mg/kg made no difference to the plasma half-life in a subgroup of the geriatric patients.5. The volume of distribution of phenazone, representing total body water, was 0·59–0·63 l/kg in all groups except the elderly females, in whom it was 0·48 l/kg. This latter group showed a correspondingly higher plasma concentration extrapolated to time zero.6. The difference in plasma half-life phenazone between normal adult and elderly groups is statistically significant, but the wide range of values in each group makes the biological significance less clear. Alteration of drug distribution with resultant higher plasma or tissue concentrations may contribute to adverse drug effects, particularly in the elderly female.

Plasma and urine steroid and plasma ACTH levels were measured for 2 weeks in eight subjects (six female, two male) with Cushing's disease with each given 200 mg ketoconazole orally four times daily.2. Treatment was associated with major falls in excretion of free cortisol and the cortisol metabolites tetrahydrocortisol, 5α-tetrahydrocortisol and tetrahydrocortisone.3. Plasma cortisol and ACTH levels did not change significantly.4. Excretion of the androgen metabolites, androsterone, 11β-hydroxyandrosterone and aetiocholanolone was also reduced but there was no significant fall in plasma testosterone. Plasma levels of progesterone, 17α-hydroxyprogesterone and 11β-deoxycortisol and excretion of pregnanediol, pregnanetriol and tetrahydrodeoxycortisol rose with treatment.5. Analysis of steroid precursor/product ratios indicated that ketoconazole significantly inhibited 11β-hydroxylase and 17,20-lyase but not 17α-hydroxylase in these patients with Cushing's disease.
