Notes: To document the effect of hypophysectomy and growth hormone replacement on the ultrastructure of cementogenesis in the developing rat third molar, 12 female Wistar rats were randomly allocated to normal control, hypophysectomized or hypophysectomized plus human growth hormone (for 10 days) treatment groups. The results of this study by electron and light microscopy and morphometry have shown that qualitative and quantitative changes occur in the organelles of cementoblasts forming cellular cementum as a result of hypophysectomy and growth hormone replacement. After hypophysectomy, the changes of less prominent nucleoli and nuclear pores, less prominent Golgi apparatuses and decreased endoplasmic reticulum can be interpreted as diminished cementum matrix biosynthesis – an interpretation that can be confirmed morphometrically by less cellular cementum formation. Growth hormone replacement for 10 days reactivates protein synthesis and cementogenesis as evidenced by ultrastructural changes in cementoblasts and a greater production of cementum.

Notes: summary This study examined the effect of a nightguard vital bleaching procedure on enamel surface morphology and the shear bond strength of a composite resin luting cement to enamel. Extracted human teeth were bleached for 1 week using a vital bleaching product. Control teeth were brushed with a fluoride toothpaste and processed similarly to the bleached teeth, however the bleaching product was substituted with artificial saliva in the night guards. Teeth were stored in artificial saliva for 24 h, 1, 6 or 12 weeks and then examined for any surface changes using light and scanning electron microscopy. The effect of etching surfaces with 37% phosphoric acid was examined at the scanning electron microscope level. The shear bond strength of composite resin luting cement to both buccal and lingual surfaces of bleached and control teeth was determined. Light microscopy investigation suggested the bleaching process resulted in a loss of mineral from enamel which was evident 24 h after bleaching and was sustained following 12 weeks storage in artificial saliva. Scanning electron microscopy showed a definite change in the surface texture of the bleached enamel surface. Acid etching of the bleached enamel surface produced loss of prismatic form and the enamel appeared overetched. The mean shear bond strength between composite resin luting cement and etched enamel tended to be lower for bleached enamel surfaces, however no significant difference in shear bond strength was noted between control and experimental groups. The results of this study suggest that bleaching resulted in changes to the surface and subsurface layers of enamel. Although surface changes were observed in the etched enamel, the shear bond strength of composite resin luting cement to etched bleached enamel appeared to be clinically acceptable

Notes: Abstract. The present study was carried out to determine the effects of citric acid (pH I) on soft tissue healing. Standardised circular wounds were created in the palates of 24 rats using a modified trephine. Test animals had citric acid (pH I) applied to the wounds for 3 min followed by rinsing with saline. In the control animals saline alone was applied for a similar time which was again followed by rinsing. Animals were sacrificed at 0. 1, 4 and 24 h, the palates dissected out and fixed in either neutral formalin, formol-sucrose for histochemical analysis or had epithelial sheets prepared for ATPase staining of Langerhans cells. At O. 1 and 4 h less enzyme activity (non-specific esterase and acid phosphatase) was observed in the epithelium in the edge of the citric acid treated wounds compared to the controls. This was not seen at 24 h. No morphological difference was noted between the citric acid treated wounds and the controls at any of the time intervals. The morphology and distribution of ATPase positive Langerhans cells did not differ between the test and control wounds. These results indicate that the application of citric acid (pH 1) has little effect on the short-term healing of soft tissue wounds in the rat palate.
