    Notes: The optical absorption spectrum of the hydrated electron was computed at 300 K using the Feynman path integral formulation of quantum statistical mechanics in conjunction with molecular dynamics simulations. In addition, the potential energy of the hydrated electron was studied at 700 K as a function of the liquid density between 0.02 and 1.0 g/cm3 and correlated with the maximum of the absorption spectrum. The procedure to calculate the optical spectrum makes use of the solution of the Schrödinger equation for an ensemble of model potentials which span the region allowed by the fluctuations of the potential well which traps the electron in thermodynamic equilibrium. The results indicate that the absorption band is due to a strongly allowed 1s→2p transition and the breadth of the spectrum is a consequence of the fluctuations in the trap dimensions. Moreover, these calculations imply that the long tail of the absorption spectrum in the UV arises from electrons which are in deep traps. Such traps occur with low but finite probability in the ensemble of solvent configurations which are accessible to the system in thermal equilibrium. Calculations of the average potential energy of the electron as a function of the density reproduced well the measured shift in the absorption spectrum of the excess electron in water.

    Notes: Storage of ripe olives in liquids with high acetic acid content under anaerobic conditions led to glucose accumulation, low ethanol, and high chemical oxygen demand (COD). In contrast, low acid concentrations and anaerobic conditions produced a high level of ethanol and slightly lower COD while aerobic conditions produced low glucose, ethanol and COD. Surface color of ripe olives improved when storage liquids were used in the second washing of the darkening step. Firmness also increased with increasing proportion of storage liquids reuse, although olives from highly acidic media were softer than those stored in low acid media.

    Notes: RFLP analysis of 105 doubled haploid lines from a cross between the barley varieties ‘Magnum’ and ‘Goldmarker’ located the denso dwarfing gene on the long arm of chromosome 3(3H), approximately 8 cM distal to the RFLP locus Xpsrl70. Lines with the denso gene showed a distinctive prostrate juvenile growth habit and tended to have later ear emergence times and lower plant grain weights, ear grain weights and 50 grain weights.

    Notes: Amplified fragment length polymorphism (AFLP) was used to study diversity within and among Spanish olive varieties. A high degree of diversity was found among the varieties present in each growing region. Olive oil production and quality relies on appropriate cultivar selection as well as good orchard management. Production based only on a few superior cultivars would result in improved yield, oil quality, and production management. Amplified fragment length polymorphism were evaluated as a tool to identify the intraspecific and intravarietal diversity of olive. Amplified fragment length polymorphism analysis of 38 accessions belonging to 10 cultivars using six primer combinations produced 106 polymorphic bands. Results were analyzed for similarity among accessions via unweighted pair-group means cluster analysis, resulting in 10 clusters corresponding to named variety designations. Similarity among varieties ranged from 0.60 to 0.72. Diversity within varieties was identified. Similarity within named varieties (accessions with the same varietal name) ranged from 0.75 to 0.96. Differences in several markers were found among 34 accessions. Intravarietal diversity was shown to exist within the Spanish olive varieties grown in the region surrounding Valencia.

    Notes: Concentrations of eight essential minerals were determined in 78 samples of goats'milk from the Canary Islands. The following mean values (ranges) were found: Se (μ/L), 19.98 (9.2-31.2); Fe (mg/L), 0.52 (0.19-1.17); Cu (mg/L), 0.17 (0.08-0.31); Zn (mg/L), 3.31 (1.6-4.6); Na (mg/L), 514 (289-906); K (mg/L), 1585 (1212-2160); Ca (mg/L), 1533 (1118-1962); and Mg (mg/L), 157 (101-210). Comparing with data for human milk and cows' milk from the Canary Islands, the mean concentrations of Se, K and Mg arranged in according the following sequence: goats' milk 〉 cows' milk 〉 human milk. Cows' milk had the highest (P〈0.05) mean concentration for Ca and Zn, while humans had the highest (P 〈 0.05) mean concentration for Cu. The milk of goats milked once per day had significantly higher concentrations of Se and lower Mg and Zn than the milk of goats milked twice per day. While the concentrations of Na and Mg increased significantly from March to August, the concentrations of Se, Fe and Cu in goats' milk decreased significantly from March to June.

    Notes: The correlation between gene expression and DNA methylation is well established. In the present study we examined DNA methylation as a potential regulatory mechanism of class I histocompatibility antigen expression. We selected for study two cell clones from the same mouse tumour (GR9). One of them is H-2 class I-positive (A7) and the other negative (B9). Using five enzymes and three different probes, no rearrangement or deletion was detected in the MHC class I genes of the negative clone compared with the positive one. These results do not provide an explanation for the differences observed in the expression of class I antigens in A7 (Kd, Dd positive) and B9 (Kd, Dd negative). We then studied the DNA methylation status in both clones by Southern blot analysis. Clear differences between the Kd, Dd positive and negative clones were detected. Furthermore, both Kd and Dd were clearly expressed after culturing the H-2 class I negative clone with the demethylating agent 5-azacytidine. These results suggest that the methylation of certain MHC cytoshes is a regulatory mechanism of H-2 class I gene expression.

    Notes: The effects of pouch material (aluminum, plastic), packing pH (3.8,4.3), acidifying compound (lactic, gluconic), and storage temperature were tested on polyphenol content and color of holding brines, surface color, and firmness of ripe olives. Using pouches of plastic films, pH 4.3–4.0 holding solution, and lactic or gluconic acid products with color and texture similar to traditional canned ripe olives were produced. They could be properly stored at room temperature (25°C). As storage temperature rose, color improved but texture was seriously degraded at 50°C.

    Notes: Solutions of caffeic acid and hydroxytyrosol, the 2 orthodiphenols involved in ripe olive darkening, were used as model systems to optimize processing. Lineal, quadratic, and lineal interaction effects of pH and manganese content on oxidation rate of both orthodiphenols were significant. Manganese did not affect activation energy of caffeic acid oxidation but diminished that of hydroxytyrosol. Presence of manganese cations (0.34 mM) and high pH (9.0) also led to the darkest color in the 10 to 30°C temperature range but did not improve the color when added after caffeic acid and hydroxytyrosol oxidation. Results can lead to a faster oxidation period, which currently takes about a week, and darker olives.
