    Notes: The release, dispersal, and ultrastructure of juveniles arising through multiple fission in the benthic foraminiferan Allogromia sp., strain NF (Lee & Pierce, 1963) has been examined by light and electron microscopy. An extensive reticulopodial network participates in the dispersal of fully differentiated young as they emerge from the fragmented parental test. During the earliest stages of release, offspring are of two classes—aroused and unaroused. Unaroused juveniles, which have not extended pseudopods, attach externally to the network and are transported bidirectionally along its surface. Aroused juveniles, which have extended pseudopods and are in protoplasmic continuity with the network, move quickly to the periphery of the network. Within 24 h, juveniles establish a communal “feeding reticulum” in which dispersed individuals are in protoplasmic continuity with neighbors via a common reticulopodial network. At the ultrastructural level, the cell body cytoplasm of unaroused juveniles contains numerous patches of a paracrystalline material, which disappears as their pseudopodia are extended to join the communal feeding reticulum. This paracrystalline material therefore appears to be a temporary reservoir of precursors required for pseudopod construction.

    Notes: Summary Allogromia sp. is a benthic foraminiferan protozoan which extends and withdraws a dynamic network of branching and anastomosing pseudopodia,i.e., reticulopods. Each reticulopod contains an elongate cytoskeleton composed primarily of microtubules (MT). When withdrawal was induced with artificial seawater supplemented with MgCl2, we found a time-dependent decrease in the number of reticulopodial MTs and a concomitant increase in 5-nm-diameter helical filaments. During the initial stages of withdrawal these helical filaments associated laterally to form loose aggregates. Later they formed dense paracrystalline aggregates, which appeared similar to those seen in the cell bodies of untreatedAllogromia juveniles prior to network extension. Similar results were obtained when withdrawal was induced by using seawater supplemented with other salts (NaCl, KCl). Treatment with an isotonic seawater substitute with an altered Na+:K+ ratio induced a momentary withdrawal, after which the organism recovered and reextended a network. During the withdrawal phase of this response, MTs became less abundant and aggregates of helical filaments more conspicuous. Together with earlier observations these findings suggest that helical filaments and paracrystalline material are an alternative or intermediate assembly form of MT proteins.

    Notes: Summary The technical details given in this paper meet the demand in neuroanatomy and neuropathology for methods which combine broad light microscopical surveys with detailed ultrastructural studies of logically selected areas in well perfused brain material, and emerge directly from experiences in Palay's laboratory at the National Institutes of Health in 1956. Using the procedures recommended will give good sections of exceptionally large areas (up to, and above 1 cm × 1 cm) of fully hardened blocks available at all points for electron microscopy. On such large blocks fully correlative, combined light and electron microscopy may be carried out easily. The process is termed ‘hot knife microtomy’. In three different laboratories, primary aldehyde fixation by perfusion and hot knife microtomy have given uniformly excellent data from normal, diseased, and virus-infected brain tissues. These techniques permit full neuroanatomical control and orientation, make comprehensive correlative mapping throughout the CNS feasible, and allow study of the time course of infective processes.
