    Notes: Abstract Positron emission tomography (PET) studies using [18F]-l-dopa were carried out in 9 patients with supranuclear palsy and 13 controls. For quantification of PET data a rate constant Ki was calculated for the radiotracer using a graphical method. Corrections for nonspecific activity were performed in both arterial plasma and brain tissue. The purpose of this study was to test the hypothesis that parametric images of the rate constant K mapping can be obtained on a pixel-by-pixel basis using an appropriate mathematical algorithm. Ki values from these parametric images and the graphical approach were compared. Both correlated closely, with y=0.013+0.947*x, r=0.992 and y=−0.052+1.048*x, r=0.965 in patients and controls, respectively. Contrast measurements were also performed and showed a striking increase in contrast on parametric images. K mapping offers several advantages over the graphical approach, since parametric images are time-independent, i. e. one image represents the quantitative result of the study. In addition, parametric images of the rate constant are normalized to arterial plasma radioactivity and corrected for tissue metabolites. Thus, parametric images of Ki in different individuals can be compared directly without further processing in order to assess the nigrostriatal integrity.

    Notes: Summary. We used [18F]6-fluorodopa (FD) positron emission tomography (PET) to examine the severity of nigrostriatal dopaminergic dysfunction in 67 patients with Idiopathic Parkinsonism (IP), 52 with fluctuations and 15 with a stable response to levodopa. FD uptake (Ki) was reduced by 12% in the caudate (p = 0.08) and by 28% in the putamen (p = 0.0004) of patients with fluctuations compared to those with a stable response. However, there was considerable overlap of FD Ki values between the two groups. The fluctuators had a longer symptom duration (11.6 ± 5.7 years) than the patients with a stable response to levodopa (4.3 ± 2.4 years; p 〈 0.0001) and the age of onset of symptoms was earlier in the fluctuators (43.9 ± 8.9 versus 54.1 ± 10.4; p = 0.0004). Similar reductions in FD Ki in the fluctuators persisted following adjustment for these variables (7.5% in the caudate and 26% in the putamen; p = n.s. and 0.007, respectively). When smaller groups (n = 15 each) were matched for duration of symptoms, the reduction in caudate Ki in the fluctuators was only 1.9% (p = n.s.), but there was still a 24% reduction in putamen Ki (p = 0.05). These findings suggest that fluctuators and non-fluctuators may differ in the severity of their nigrostriatal damage and provide modest support for the hypothesis that fluctuations may in part reflect altered "buffering" capacity of dopaminergic nerve terminals. However, the considerable overlap between groups suggests that other factors such as altered postsynaptic mechanisms and/or increased turnover of dopamine may make a substantial contribution to the development of motor fluctuations.

This review focuses on the production of these isotopes, the chemistry of labelling radiopharmaceuticals with these isotopes, automated chemistry and finally future considerations and applications for both11C and18F.

    Notes: Abstract An institute for the training of radiochemist was proposed in 1989 by scientists/educators from Simon Fraser University (SFU) and the TRIUMF Nuclear Research Facility in Canada. The intensive program spans 6 weeks and includes seminars and problem sesssions as well as practical laboratory experience. Topics include health physics, synthesis of short lived radiopharmaceuticals, automation and quality control. The first offering was in May–June 1990. Based on this experience a textbook on Radiochemistry and Radiopharmaceutical Chemistry has been initiated. Parts of the program may be implemented into a credit course to be offered through the Department of Chemistry at SFU.

    Notes: Abstract A new method for the highly efficient separation of48V for radiotracer experiments is presented. It is based on the proton irradiation of a high purity Ti plate which is dissolved in sulfuric acid to form Ti3+ and V4+. Both ions are quantitatively absorbed on AG50W-X8 cation exchange resin and are converted to the respective higher valence state by an on-column reaction with diluted nitric acid and hydrogen peroxide. The eluate containing V5+ is evaporated to dryness in presence of sulfuric acid and treated with ethanol to reduce [48V]-vanadate to [48V]-vanadyl for labeling experiments in pharmaceutical research.
