Notes: : Multiple degradation pathways accounting for the breakdown of the dipeptide sweetener a-L-aspartyl-L-phenylalanine methyl ester (a-APM) in 4 mM phosphate, pH 3.1 and a simulated cola beverage are described. At 20-, 35-, 45- and 55 °C caramel coloring concentrations greater than 700 ppm increased the rate of hydrolysis of the methyl ester forming a-L-aspartyl-L-phenylalanine. At 55 °C and pH 3.1 caramel coloring also enhanced the breakdown of a-APM degradation products. The influence of caramel coloring on the rates of degradation varied considerably for a-APM and each degradation product. A model is proposed based on the expected solution properties of caramel coloring which accounts for the enhanced degradation.

Notes: Ninety-nine classical and 11 atypical bronchopulmonary carcinoids were investigated immunohistochemically with an antibody against secretoneurin, a peptide proteolytically processed from secretogranin II (chromogranin C), as well as antibodies against chromogranin A and B and secretogranin II. Secretoneurin was immunolocalized in 86 tumours (78 classical and eight atypical carcinoids); secretogranin II was found in the same tumours in a similar distribution, whereas chromogranin A was present in all 100 and chromogranin B in 106 tumours investigated. Bronchopulmonary carcinoids are usually not associated with clinically or biochemically distinct syndromes. Although we found bronchial carcinoids with different immunohistochemical chromogranins/secretogranin patterns, no correlation with the biological behaviour of these tumours could be demonstrated.

Notes: Surgical specimens of 65 adrenal and 27 extra-adrenal paragangliomas, the latter comprising 11 carotid body, five jugulotympanic, one aorticopulmonary, eight aorticosympathetic and two visceral autonomic tumours, were examined immunocytochemically for the presence of glial fibrillary acid protein (GFAP) and S-100 protein. Six adrenal and four extra-adrenal (one parasympathetic and three sympathetic) neoplasms pursued a malignant clinical course. S-100 staining of sustentacular (type 2) cells was seen in both adrenal (48/65) and extra-adrenal (23/27) lesions, the 10 malignant tumours being entirely devoid of S-100 protein positive cells. GFAP positivity of type 2 cells was seen in only 16 of the extra-adrenal tumours, all of these lesions belonging to the group of benign parasympathetic paragangliomas. The presence of S-100 positive type 2 cells may thus help to exclude malignancy in individual paraganglioma cases, while GFAP positivity of such cells renders possible the correct typing of benign parasympathetic paragangliomas.

Notes: Creep and cyclic deformation behavior of two lead-free high temperature solder alloys, 95Sn-5Ag and 99Sn-1.0Cu, a high lead alloy 97.SPb-1.SAg-1.0Sn, and an Ag-modified eutectic alloy 62.SSn-36.1Pb-1.4Ag, were studied. Room temperature and high (100°C and 150°C) temperature fatigue tests (with cyclic strain amplitude up to 6.0%) for the four solders were conducted, with the fatigue lives ranging from a few cycles to more than 100,000 cycles. It is shown that among the alloys studied, 62.SSn-36.1Pb-1.4Ag (the modified Sn-Pb eutectic alloy) has the lowest fatigue resistance in term of low cycle fatigue life (strain controlled). The high lead alloy, 97.SPb-1.5Ag-1.0Sn, has the highest strain fatigue resistance in the large strain region (Δ 〉 2.0%). Temperature has a significant effect on alloys 95Sn-5Ag and 99Sn-1.0Cu, but has a negligible effect on the Ag modified Sn-Pb eutectic alloy 62.5Sn-36.1Pb-1.4Ag and 97.5Pb-1.5Ag-1.0Sn. Creep studies show that these alloys generally have a very significant primary creep regime (up to 20%); thus, any realistic constitutive relation has to take such a primary creep phase into consideration. Cyclic deformation of alloy 95Sn-SAg was simulated by using a constitutive relation built upon a 2-cell model, which covers both primary and secondary creep. This model provides a good estimate of the peak stresses (the minimum stress and the maximum stress in each cycle); it agrees with experimental results when the applied cyclic strain is small and/or the applied strain rate is very low.

Notes: Thirteen primary thyroid clear-cell carcinomas and eight thyroid metastases of renal carcinomas are described. A broad variety of features is shown to be responsible for this follicle cell phenotype, some of which also occur in renal carcinomas. In biopsy specimens these two conditions can only be distinguished by immunohistochemistry. Clinical follow-up disclosed that in the papillary and follicular categories of thyroid cancer the biological behaviour of clear-cell tumours exhibits no significant difference to their non clear-cell counterparts. In the case of eight follicle cell carcinomas, various samples were available for study. These revealed some striking variations in histology at different stages, and even during the same stage, of disease. Both reversal of clear cell change and transitions towards other kinds of metaplasia occurred. Morever, maintenance of follicle cell function was documented in several of these lesions by estimations of serum thyroglobulin levels and radioiodine scans. On the basis of these findings the concept of regarding the clear-cell variant as a distinct subtype of thyroid cancer is refuted. Previous statements on the causal mechanism of clear-cell change are discussed and it is suggested that metaplastic transformation accounts for this phenomenon in at least some instances of this heterogeneous condition.

Notes: Summary Leu-M1 antigen is a monocyte/granulocyte-related marker known to be consistently expressed in the Reed-Sternberg cells of patients with Hodgkin's disease. Recently, however, the presence of Leu-M1 has also been noted in tumour cells of a variety of non-haematopoietic neoplasms, most of them adenocarcinomas. The biological significance of this aberrant reaction has not been clarified. We have been able to demonstrate marked epithelial Leu-M1 immunoreactivity (〉15% tumour cells positively stained) in 24 out of 76 (32%) papillary carcinomas of the thyroid gland (PC). This phenomenon was more frequently observed among PCs at an advanced stage of disease (pT4 vs. pT1–3 and M1 vs. M0 p〈0.05). The degree of epithelial Leu-M1 positivity was also shown to be significantly correlated to the clinical course of PC. Irrespective of other morphological and clinical features, death resulting from cancer occurred 17 times more frequently among PCs with marked Leu-M1 positivity (8/24) when compared with tumours with only slight or absent immunoreactivity (1/52) (p〈0.00005). These findings suggest that Leu-M1 immunostaining provides significant prognostic information for patients with papillary carcinoma of the thyroid gland.
