    Notizen: Summary The authors isolated numerous microorganisms with the capacity to assimilate large amounts of benzoate from many soil samples. Several of them were selected and subjected to mutation mainly by ultraviolet irradiation. One mutant lacking active muconate-lactonizing enzyme, the parent strain of which was identified as belonging to the genus Arthrobacter, was isolated and found to be capable of producing cis, cis-muconic acid with a quantitative yield of 44.1 g/l over 48 h in a 30 1 jar fermentor by successive feeding of small amounts of benzoate. This mutant, however, was more sensitive to high concentrations of the substrate than the parent strain. As few intermediates and isomers other than cis, cis-muconic acid were accumulated in the large fermentor, a large amount of pure cis, cis-muconic acid was easily obtained from the broth by salting out and recrystallization at a high recovery rate.

    Notizen: Summary Production of anthocyanin was greater in conditioned medium than in LS medium. The highest production was obtained after 2 days culturing, and the value reached 1100 μg/g cell, which was about twice that of LS medium cultured in each cell inoculum. The highest anthocyanin content was obtained with a 1g cell (fresh weight) inoculation using conditioned medium. The percentage of the main anthocyanin, peonidin-3-glucoside, increased in accordance with the decrease in cell inoculum size, contrary to the results obtained for cyanidin-3-glucoside. This tendency was more clearly observed in LS medium than in conditioned medium.

    Notizen: Abstract Fragaria ananassa (strawberry) callus, which produced high amounts of anthocyanin in the dark, was isolated from a cell line not producing anthocyanin. The isolated callus (FAR) was homogeneous and more than 90% of the cells were pigmented. The FAR callus accumulated more than 1000 μg of anthocyanin per g fresh cell in the dark. Four different basal solid media were examined to maintain FAR callus: Though growth rate and anthocyanin concentration were different on each media, total anthocyanin production was about the same at 400 μg anthocyanin/0.1 g fresh cell wt after 22 days. This FAR cell line could therefore be used for the industrial production of anthocyanin.

    Notizen: Abstract Anthocyanin accumulation in strawberry (Fragaria ananassa) cells cultured on a solid medium was monitored using an image-processing system that did not require direct sampling or destruction of the cells. Because of the intercellular heterogeneity of secondary metabolite production in plant cell cultures, the maximum metabolite concentration in individual cells is often more than 10 times higher than that of the average concentration. An image-processing based method enabled the growth and the pigmentation behavior of individual cells to be traced. Changes in the time courses of the anthocyanin content of individual cells differed from each other, although the average anthocyanin contents increased gradually with time in a batch culture. However, these various changing patterns in the anthocyanin content of each cell were independent of the cell cycle. In addition, image analysis revealed that the two cells just after cell division were almost identical to each other both in size and anthocyanin content. The proposed method which uses an image-processing system provides a useful tool for analyzing the secondary metabolism in individual cultured plant cells.

    Notizen: The reaction behavior of Saccharomyces formaosensis imobilized by polyacrylamide gel is presented. Two types of the immobilized yeast are studied, i. e. the immobilized resting yeast and the immobilized growing yeast. For both of the yeast, reaction retes are expressed by the Michaelis-Menten type equation with a linear ethanol inhibition factor. The Michaelis constants aere close each other, but considerably larger that of native S. cerevisiae. Distribution of the growing yeast cell inside the carrier gel is presented. It is found that the cell density is somewhat higher near the surface of the carrier.

    Notizen: The production characteristics for Taxol (paclitaxel) using free and immobilized cells of Taxus cuspidata were investigated in a perfusion culture bioreactor. Although the cell growth was inhibited by higher dilution rates, the specific production rate of Taxol was increased by perfusion compared with that using batch operation. Perfusion cultures using a nylon-mesh cell separator for free suspension cells showed similar production profiles to those obtained using immobilized cells. Continuous Taxol production was successfully obtained at an approximate specific production rate of 0.3 mg/g DCW (dry cell weight) per day for up to 40 days. © 1997 John Wiley & Sons, Inc.
