Notes: The effects of a short exposure to red, far-red or alternate red/far-red light on the germination of seeds after-ripened for different periods of time were studied in dormant lines of wild oat (Avena fatua L.). Three stages were distinguishable in the after-ripening period in the response of germination to light. Seeds stayed dormant and showed no response to light during stage I. Phytochrome-mediated germination was observed in seeds during stage II. The phytochrome action disappeared during stage III, i.e. seeds fully germinated following treatments of all light qualities. When the seeds were imbibed in polyethylene glycol solutions, dark germination was reduced and phytochrome again had an effect, which suggested the involvement of phytochrome in water uptake of the seed.

Notes: [Auszug] The method used by us for assaying chromato-grams, or small volumes of solution containing gibberellic acid, is as follows. Non-dormant A. fatua L. seeds are allowed to imbibe water for about 15 min. to soften the testa. Embryos are then excised with a sharp scalpel under a dissecting microscope ...

Notes: High performance liquid chromatography with octadecylsilica (Bondapak C18/Poracil B) column packing was used to purify and separate cytokinins in Sorghum leaf extracts. The column size was 56 × 0.21 cm i.d. By gradient elution, using acidified water with increasing amounts of methanol, the major peaks of cytokinin activity, as determined by the callus tissue bioassay. were effectively separated from large amounts of extraneous impurities. These cytokinins were further separated on a microoctadecylsilica column (μBondapak C18, 30 × 0.4 cm i.d.) with a gradient of acidified water-acetonitrile. Zeatin and zeatin riboside gave distinct ultra violet absorption peaks which could be used for quantitative estimation. Biological activity corresponded to the elution of these peaks. These two cytokinins are the major cytokinins in Sorghum leaves.

Notes: Citric, succinic, fumaric, malic, pyruvic and lactic acids induced germination in two genetically pure dormant lines of Avena fatua L. The sensitivity to these acids was low immediately after harvest and increased markedly after a period of dry after-ripening. Because the acids could only overcome dormancy in partly after-ripened caryopses, the mode of their action in these caryopses differed from that of another germination promotor, ethanol, and was similar to that of the germination promoter, sodium nitrate. The mode of action of the organic acids on the partly after-ripened caryopses through lowering pH was indicated by the observation that other non-metabolic weak acids could also break dormancy while neutral pH value salt solutions of some of the tested acids were inactive. The dose-response curves of citric acid for the stimulation of germination and for oxygen uptake were similar, indicating that this organic acid may stimulate germination by promoting oxygen uptake. A time sequence study showed that citric acid stimulated oxygen uptake before the first visible signs of germination. Stimulation of germination and oxygen uptake over a range of pH values showed that those values of pH which stimulated germination also stimulated oxygen uptake, indicating that the ability to stimulate oxygen uptake was not confined to organic acids. The stimulation of both germination and oxygen uptake by citric acid was not inhibited by salicylhydroxamic acid, an inhibitor of alternative respiration, therefore stimulation of both germination and oxygen uptake by citric acid does not require the operation of the alternative pathway of respiration. The function of weak acids as promoters of oxygen uptake is discussed with reference to the breakage of dormancy in partly after-ripened caryopses and the involvement of various respiratory pathways is indicated.

Notes: Adkins, S. W., Symons, S. J. and Simpson, G. M. 1988. The physiological basis of seed dormancy in Avena fatua. VIII. Action of malonic acid - Physiol. Plant, 72: 477–482.A low concentration of malonic acid (50 mM) induced germination in four genetically pure dormant lines of Avena fatua L. Sensitivity to this treatment was poor immediately after harvest but increased markedly during after-ripening, indicating that the mode of action of malonic acid (50 mM) was similar to that of another organic acid, citric acid. Over the concentration range (10–50 mM) where malonic acid promoted germination, oxygen uptake was also stimulated, and this was before the first visible signs of germination. At higher concentrations (100–300 mM) where there was no promotion of germination, malonic acid strongly inhibited oxygen uptake. These results show that malonic acid has a dual effect on oxygen uptake and subsequent germination. Low concentrations (10–50 mM) act by stimulating the Krebs cycle and germination through an acidification reaction like citric acid, and high concentrations (100–300 mM) act by inhibiting germination through enzymatic restraint of the Krebs cycle. The stimulation of both oxygen uptake and germination by three established germination promoters (sodium nitrate, citric acid and ethanol) was inhibited by a high concentration of malonic acid (200 mM) but unaffected by a low concentration (50 mM). These results show that oxygen uptake, and hence the activity of the Krebs cycle, are important processes involved in the dormancy breaking mechanism of these three promotors.

Notes: Summary An investigation was carried out on harmine as a possible anti-parkinson agent in reserpine, chloropromazine or encephalitis induced parkinsonism. Significantly better response to harmine was obtained in the patients with the reserpine-induced symptoms than in the other types. It is proposed that biological antagonism between harmine and reserpine at brain receptors might result in such a differential action. This effect, however, is short and lasts only 3 hours.

Notes: Abstract A rating scale for tardive dyskinesia was developed, consisting of nearly all signs seen by two groups of investigators over a 5-year period. Thirty-four items were included in the scale with a possibility of writing in idiosyncratic signs. The scale was shown to have good reliability and validity in studies carried out by both the New York and Boston groups. It is recommended as a suitable instrument for describing the breadth of tardive dyskinesia syndrome and also for quantifying the disorder. A second scale, “the abbreviated dyskinesia scale”, contains 13 items which are more global than the items in the original scale. It also has been shown to be both reliable and valid. Its use is suited to situations requiring less extensively detailed assessments.

Notes: Abstract The diurnal and weekly variability of tardive dyskinesia (TD) was assessed instrumentally by digital image processing. Weekly assessments were obtained in ten patients over a 6-week period. In six of the ten patients, assessments were obtained four times over a single 12-h period. Results indicate that TD movements measured by instrumental assessment vary from week to week and throughout the day. Factors that appear to have affected the variability were changes in medications and the time of day when the assessments were conducted.

Notes: Abstract The present study was performed to investigate the reliability and validity of a new device for the assessment of oral dyskinesias by means of digital image processing. Twenty schizophrenic patients with tardive dyskinesia (TD) and ten healthy controls were studied. In patients instrumental scores were compared to different clinical rating scale scores. Measurements were repeated after 2 weeks under the same circumstances to assess test-retest stability. Instrumental scores discriminated well between normal subjects and dyskinetic patients and correlated significantly with clinical ratings (r=0.63). The test-retest correlations showed a correspondence not larger than 40%, detecting thus the fluctuation of the TD intensity over time. These results suggest that our device is a reliable and easy to handle technique for the assessment of TD. Furthermore, the ability of the device to analyze the frequency distribution of movements makes it a useful tool for the quantitative and qualitative analysis of TD.
