    Notes: Abstract— An assay for 3,4-dihydroxyphenylacetic acid (DOPAC) capable of detecting as little as 1 pmole of DOPAC is described. The basis of the assay is the O-methylation of DOPAC utilizing S-[methyl-3H]adenosyl-l-methionine and a partially purified catechol-O-methyl transferase to form [methyl-3H]homovanillic acid. The [methyl-3H]homovanillic acid is purified with ion exchange resin and either solvent partitions or TLC. Utilizing this assay, the effect of either chlorpromazine or reserpine upon the content of DOPAC both in the caudate nucleus and in the substantia nigra was determined. Both of these drugs increase the level of DOPAC in the caudate nucleus but have minimal effects upon the level of DOPAC in the substantia nigra.

    Notes: —A specific and sensitive, radioisotopic microassay for tryptophan hydroxylase (EC 1.14.36) is described, which is capable of determining enzymatic activity in as little as 5 μg of crude brainstem homogenate. 5-Hydroxytryptophan, the immediate product of hydroxylation of tryptophan is enzymatically converted to N-acetylserotonin. A radioisotopic label is then introduced by the enzymatic methylation of N-acetylserotonin in the presence of [3H]methyl-S-adenosyl-methionine. The [3H]-melatonin thus formed is isolated by extraction and counted.With this assay, the activity in individual hypothalamic nuclei (arcuate nucleus, median eminence, suprachiasmatic nucleus, and medial forebrain bundle) has been measured.

    Notes: Choline acetyltransferase levels have been measured in specific nuclei of the diencephalon. On the whole, the thalamic nuclei contain higher concentrations of this enzyme than do the hypothalamic and preoptic nuclei. Those nuclei which seem to receive the most dense cholinergic innervation contain the highest levels of choline acetyltransferase.

    Notes: Abstract— An enzymatic isotopic assay for the measurement of β-phenylethylamine in brain, with a sensitivity of 100-200 pg, has been developed. With this assay, the endogenous β-phenylethylamine content (1.5 ng/g) in the rat brain has been determined. Phenylalanine administration increases the brain levels of this amine; inhibition of monoamine oxidase causes a 40-fold increase in brain β-phenylethylamine. After a combined treatment with a monoamine oxidase inhibitor and phenylalanine, the β-phenylethylamine content in the brain increases to about 400-fold. This increase can be blocked by the central decarboxylase inhibitor NSD-1055. p-Chlorophenylalanine also increases β-phenylethylamipe concentration in the brain, and this effect is potentiated by a simultaneous administration of phenylalanine.

    Notes: Abstract— 5-Hydroxy-l-tryptophan decarboxylase (EC 4.1.1.28) activity was measured in discrete regions of the rat brain by means of a sensitive micromethod lhat allows the quantitation of enzyme activity in μg amounts of brain tissue.A good correlation was obtained between serotonin levels and 5-hydroxy-l-tryptophan decarboxylase activity in all the brain regions examined. Wide differences in enzyme activity were found in the different brain nuclei, with a 20-fold difference in activity between the most active region (the nucleus raphe dorsalis) and the least active regions (the corpus callosum and the optic tract).

    Notes: Abstract— Biogenic amines and related enzymes were quantitatively measured in the pituitary gland of the rat. The sensitivity of the assays used allows the determinations to be performed in single pituitary lobes. Relatively high values of histamine and serotonin were found in all three lobes, with higher amounts in the posterior and intermediate lobes. Highest catecholamine concentrations were detected in the posterior lobe, and only very low amounts of dopamine were measured in the anterior lobe. Throughout the gland, norepinephrine concentrations were low, about one-tenth that of dopamine. Tryptamine could not be detected. High levels of A and B monoamine oxidase were found in all three pituitary lobes. Tyrosine hydroxylase activity was measured in the posterior and intermediate lobes, but was not detected in the anterior lobe. Tryptophan hydroxylase was present in all three pituitary lobes. A relatively low catechol-O-methyltransferase activity was found in the anterior lobe, and none was detected in the intermediate and posterior lobe. Choline acetyltransferase, dopamine-β-hydroxylase and phenylethanolamine-N-methyltransferase activities could not be detected.

    Notes: Abstract— Superior cervical ganglionectomy results in a complete noradrenergic neuronal denervation of the rat sublingual-submaxillary salivary gland. Dopamine-β-hydroxylase activity in the serous submaxillary gland falls approximately 90% after noradrenergic denervation; but in the mucinproducing sublingual gland dopamine-β-hydroxylase activity is reduced by only 33%. Dopamine-β-hydroxylase immunofluorescence in the submaxillary gland is distributed with noradrenergic neurons and is eliminated by superior cervical ganglionectomy. In the sublingual gland dopamine-β-hydroxylase immunofluorescence is localized within mucinous acini and small ducts, and the disposition and intensity of staining materials is not affected by noradrenergic denervation for up to 30 days. DBH protein in the sublingual gland had little physiologic activity in vivo. Low levels of authentic dopamine-β-hydroxylase activity were detected in saliva. Thus, dopamine-β-hydroxylase protein is present in the sublingual gland in an extraneuronal location and appears to be a secretory product of the gland.

    Notes: —Choline acetyltransferase (ChAc) content of 50 separate rat brain stem nuclei and cerebellum removed by microdissection was determined using a sensitive radiometric assay. The distribution of ChAc activity is uneven, with extremely high levels in the cranial motor nuclei and the nucleus salivatorius. Low ChAc concentrations were observed in the cranial sensory nuclei, the nuclei of the reticular formation, the raphe nuclei and the nuclei of the acoustic system. The lowest ChAc levels were measured in the cerebellum. Comparison of the distribution of ChAc with histochemical localization of acetylcholinesterase revealed generally good agreement, and notable exceptions are discussed.

    Notes: Abstract— The distribution and properties of a nonspecific N-methyltransferase in the rat brain are described. The enzyme N-methylates tryptamine and N-methyltryptamine as well as β-phenylethylamine, phenylethanolamine, tyramine and octopamine. The enzyme exhibits a pH optimum of 7·9 with phosphate buffer and has a Km for tryptamine of 28 μM. There are potent inhibitors to the enzyme that can be removed by dialysis. Enzymatic activity is present in the brains of a number of species including man, rat, mouse, guinea-pig and frog. Its activity is unevenly distributed in the brain with the highest activity in the cerebral cortex and striaturn of the rat and in the subcortical white matter in man. Studies of its subcellular distribution indicate that most of the N-methylation activity is released into the soluble fraction. Enzymatic activity is also present in a number of peripheral tissues of the rat.
