    Notes: An infant girl with chronic eczema, recurrent infections, elevated IgE and impaired neutrophil chemotaxis appeared to belong to the group of patients described by Buckley, Wray & Belmaker (1972). An hereditary influence is suggested by a similar defect found in the patient's mother. Levamisole improved the in vitro defect and the clinical picture.

    Notes: Human neutrophils activated by phorbol myristatc acelatc were cytoioxic lo ox erythrocyles. as determined by the 51Cr release method. Maximal cytolysis was obtained with a phorbol myristute acetate concentration of 5 ng/ml and with an etTcctor to target cell ratio of 1:4. An inlact neutrophil metabolic burst and production of oxygen-dcrived-frec radicals were essential lor the cytotnxic event, since neulrophils from patients with chronic granulo-malous disease failed to exhibit any ox erythrocyte lysis. The target cell destruction was completely prevented by catalase, was unaffected by supcroxidc dismutase. and was reduced approximately to one-third by azide and cyanide. These data suggest that, under our experimental conditions, the ox erythrocyte killing by phorbol myristatc acetate-activated neutrophils mainly depends on myelopcroxidasc and hydrogen peroxide.

    Notes: Summary The colony-stimulating activity (CSA) produced by the blood leukocytes has been studied before and after epinephrine administration in ten normal, 15 splenomegalic, and seven splenectomized subjects through a double layer agar culture system. A significant increase of mean values of the CSA per milliliter produced by blood monocytes has been observed after epinephrine administration in the groups of normal and of splenomegalic subjects. In the group of splenectomized subjects the baseline mean value of CSA per milliliter of blood was higher than those observed in the other groups, but it did not show any increase after epinephrine infusion. The CSA produced by 106 blood leukocytes was similar in all three groups of subjects, and it was not similarly modified by epinephrine administration. Our results seem to indicate that the leukocytes producing CSA are distributed within two rapidly exchangeable blood compartments, the spleen representing an important section of the marginal compartment of blood monocytes.

    Description / Table of Contents: Summary A-35-year-old woman with a long-lasting history of neutropenia and recurrent infections was found to have defective neutrophil chemotaxis, random motility, and in vivo migration. Although the bone marrow granulocyte reserve was normal, the patient failed to release an appropriate amount of granulocytes after injection of etiocholanolone. These features are characteristic of the so-called „Lazy leukocyte syndrome“. The clinical presentation of the five cases of this syndrome so far reported and its pathophysiological aspects are discussed.
    Notes: Zusammenfassung Bei einer 35jährigen Patientin bestanden seit Jahren Neutropenie und Infektanfälligkeit. Die Diagnose eines „Lazy Leukocyte“Syndroms ergab sich aus folgenden Befunden: gestörte Neutrophilen-Chemotaxis; eingeschränkte ungerichtete Motilität und In-vivo-Migration; verminderte Granulozyten-Freisetzung aus dem Knochenmark bei EtiocholanolonReiz trotz normaler Knochenmarks-Zellularität. Klinische Befunde und pathophysiologische Aspekte der bisher mitgeteilten 5 Fälle dieses Syndroms werden besprochen.

    Description / Table of Contents: Summary Blood colony-forming cells (CFU-C) and colony-stimulating activity obtained from feeder layers of peripheral blood leucocytes (leucocyte CSA) have been studied in 69 normal subjects by means of semisolid agar culture system. Groups of normal volunteers were selected according to sex and age (20 to 45 and older than 60 years) and the results compared. The mean number of circulating CFU-C was significantly lower in young women (20–45 years old) than in males over 60 years of age, but no differences were found among the other age and sex groups. Leucocyte CSA did not significantly differ among these groups. In 5 young males the blood CFU-C did not show significant variations at 8 AM and at 4 PM of the same day. When the study was repeated in 18 subjects at longer time intervals, the number of colonies showed a maximum fivefold variation. The amount of plasma and polymorphonuclear granulocytes present in our culture system did not inhibit the colony growth. In most cases, double layer cultures grow a higher number of colonies than single layer, but feeder layers of some normal subject seem to inhibit the colony growth.
    Notes: Zusammenfassung Koloniebildende Zellen (CFU-C) aus dem Blut und Koloniestimulierte Aktivität aus dem „feeder layer“ peripherer Blutleukozyten (Leukozyten-CSA) wurden in einem halbflüssigen Agarkultur-System bei 69 Normalpersonen untersucht. Gesunde Freiwillige wurden nach Geschlecht und Alter (20–45 Jahre und älter als 60 Jahre) in Gruppen unterteilt. Der Anteil der zirkulierenden CFU-C war bei jungen Frauen (20–45 Jahre) signifikant niedriger als in der Gruppe älterer Männer (älter als 60 Jahre), jedoch waren die Unterschiede zwischen den übrigen Gruppen nicht signifikant. Leukozytäre CSA unterschied sich in den verschiedenen Gruppen nicht signifikant. Bei 5 jungen Männern wurden zirkulierende CFU-C am selben Tag morgens um 8.00 und nachmittags um 16.00 Uhr untersucht: Es ergaben sich keine unterschiedlichen Werte. Bei 18 Personen wurden die Untersuchungen nach längeren Zeitintervallen wiederholt: Die Anzahl der gebildeten Kolonien variierte maximal um das Fünffache. Plasma und segmentkernige Granulozyten, wie sie im angewandten Kultursystem üblich waren, inhibierten das Wachstum der Kolonien nicht. In den meisten Fällen ließen sich in doppelschichtigen Kultursystemen höhere Koloniezahlen erreichen als in einschichtigen, jedoch schienen die „feeder layer“ einiger normaler Personen das Koloniewachstum zu inhibieren.

    Notes: Summary The basal and total pyruvate dehydrogenase activities were assayed in circulating lymphocytes from children with juvenile diabetes at diagnosis and after five days of insulin therapy and from control subjects. In untreated diabetic children, basal and total pyruvate dehydrogenase activities were deeply decreased and both showed very similar values; whereas, in control subjects basal activity was about 30% lower than total activity. In diabetic patients treated with insulin (in vivo situation), both basal and total activity levels were equal or even higher than those of the control subjects. The incubation of lymphocytes from diabetic patients with insulin (5 μU/ml) (in vitro situation) stimulates, but less than in vivo, the basal and total pyruvate dehydrogenase activities.

    Notes: Abstract Ten patients with neutrophil dysfunctions and recurrent pyogenic infections, mainly of the skin, middle ear, and respiratory tract, are described. The most frequently affected functions were chemotaxis and bacterial killing. Pharmacologic restoration of functional defects was tried in all cases. Levamisole was given in two cases and ascorbic acid in the other eight cases. During a follow up of at least 18 months, seven patients showed a complete restoration of neutrophil function and a long-lasting clinical remission. One of the two patients with Chronic Granulomatous Disease has been free from infections for 1 year, despite persistent neutrophil dysfunction, while the other did not display consistent clinical improvement. Another patient, who was given ascorbic acid for a short period only due to non compliance, showed neither laboratory nor clinical improvement.
