    Notes: Among 54 human monoclonal IgA proteins, 5 inhibited the binding of labeled human monoclonal IgM (IgM Se and IgM Ba) to protein-A-containing staphylococci, whereas 48 did not have this ability. One protein (IpA Ha) showed discordant findings in the IgM Se and IgM Ba inhibition assays. Among the 54 IgA proteins, 49 were of the IgA1 and 5 of the IgA2 subclass All of the 5 IgA2 proteins inhibited the binding of IgM Se and IgM Ba to staphylococci, whereas the 48 proteins that reacted negatively in the two inhibition belonged to the IgA1 subclass. These findings show that the ability to react with staphylococci is a properly of the IgA2 subclass. IgM proteins may also be divided into two groups based on different reactivities with protein A. It is proposed to call the two IgM subclasses IgM1 and IgM2, where the latter is defined by ability to react with protein A.

    Notes: Ricinus agglutinin purified to homogeneity reacts with serum glycoproteins containing terminal nonreducing galactose residues, including about 10% polyclonal IgG. To study the IgG subclass distribution of this reaction, four monoclonal IgG1 proteins, two IgG2, four IgG3, and two IgG4 proteins were tested by affinity chromatography experiments with insolubilized ricinus agglutinin. Only the IgG3 proteins reacted, and the reactive sites were localized to the Fc fragment. Passage of polyclonal human IgG through a ricinus agglutinin–Sepharose column, followed by elution with lactose, permitted the isolation of IgG3 from normal polyclonal IgG.

    Notes: Established chronic gingival lessions were studies in four categories of patients. One groups of healthy subjects was kept plaque-free (CO), while in the second group (CP) moderate accumulations of dental plaque were permitted. The third group (UH) comprised patients with uremia and in hemodialysis. A fourth group (IS) had received renal allografts and was kept on an immunosuppressive regimen. Differential blood counts and serum immunoglobin quantitation from the UH and IS groups gave mean values within normal ranges. Gingival biopsies were obtained from each subject. The connective tissue inflammation (CTI) scores were compared between the groups. Despite abundant local plaque accumulations, the UH group displayed essentially the same CTI scroes as plaque-free controls (CO), while the IS group showed a significantly lower (p 〈 0.05) CTI score than the two in question. The CTI scores of the CP group (controls with plaque) were significantly higher (P 〈 0.01) than those of the UH, IS and CO groups. It is concluded that immunologic homeostasis is important in the periodontal tissue of man.

    Notes: Samples of gingival fluid and plasma were taken from ten patients with varying severity of periodontal disease. Group I comprised seven patients with abundant plaque. Their mean pocket depth at the sampling sites was 5.6 ± 1.27 mm. Group II consisted of three patients with minimal plaque and a mean pocket depth of 1.8 ± 0.24 mm. A comparative analysis by crossed immunoelectrophoresis was carried out. Qualitatively evaluated orosomucoid and the complement factor C3 were degraded in all the gingival fluid samples, in contrast to those of plasma. A quantitative assay of 15 separate proteins in the two biological fluids showed reduced concentrations of alfa1-antichymotrypsin, alfa2-macroglobulin and haptoglobin in most gingival fluids. Orosomucoid, alfa1-antitrypsin and C3 levels were decreased in all the gingival fluid samples. Reference ranges should be stated separately for the two biological fluids. The correlation coefficients between the plasma and the gingival fluid concentrations of each protein were calculated. The co-variation was significant for 13 of the proteins studied. The ratio of the individual protein concentrations in the gingival fluid to that in the plasma was then calculated for each patient. A twoway analysis of variance of the variables patient groups and protein ratios showed that both effects were significant. Gingival fluid is an inflammatory exudate of serumal origin, but its protein profile is clearly modified by the local milieu.

    Notes: The in vitro lymphocyte blastogenic responses of renal allograft recipients treated with immunosuppressive agents were compared with those of systemically healthy subjects with varying degrees of periodontal disease. The healthy subjects were divided into three groups; Group I showed no bleeding gingival units on gentle probing, Group II comprised persons with gingivitis, while Group III consisted of those with periodontitis. The target group (IS) comprised 11 renal allograft recipients maintained on an azathioprine and corticosteroid regimen. Two other such patients (Group CY) treated with cyclophosphamide and corticosteroids were also evaluated. Total and differential peripheral blood leukocyte counts from the IS group gave mean values within normal ranges. The mononuclear cells from peripheral blood were cultured for five days in a medium supplemented with foetal bovine serum. Triplicate cultures were stimulated with phytohemagglutinin and pokeweed mitogen. The responses of the IS patients and the systemically healthy subjects were not significantly different, whereas cells from the patients treated with cyclophosphamide showed impaired proliferative activity. When cultures were stimulated with a sonicate of dental plaque (DP), cells from the Groups II and III showed a statistically significant higher 3H-thymidine uptake than did those from the Groups I and IS (p〈0.05). The difference between the latter two groups was not statistically significant. The weak blastogenic responses of IS cells to DP antigens may be part of the explanation for the mild gingivitis observed in these patients.

    Notes: Ricinus agglutinin purified to homogeneity precipitates certain serum glyco-proteins containing terminal non-reducing galactose residues. Immunoelec-trophoresis of human sera in agarose gel with ricinus agglutinin in the antibody trough gave 2 fusing precipitin lines due to reaction with IgM and haptoglobin. All of 50 monoclonal IgM proteins precipitated with ricinus agglutinin, and the reactive sites were localized to the Fc μ fragment. By affinity chromatography on Sepharose columns containing insolubilized ricinus agglutinin prealbumin, albumin, α1-lipoprotein, α1-antitrypsin, α1-acid glycoprotein, α1-antichymotrypsin, α2HS glycoprotein, Ge-globulin, caerulo-plasmin, β-lipoprotein, β1 C-globulin, and transferrin were not retained, whereas α2-macroglobulin, haptoglobin, IgM, IgA, and IgG were retained and could be eluted with lactose. Fused rocket immunoelectrophoresis revealed varying degrees of microheterogeneity among the latter proteins; almost all of IgM but only a small fraction of polyclonal IgG was retained on the ricinus column.

Die bakteriologische Heilungsrate unmittelbar im Anschluß an die Therapie betrug 82% (18/22) in der Gruppe mit Kombinationstherapie und 72% (13/18) in der Pivmecillinam-Gruppe. Die ursächlichen pathogenen Erreger waren bei 89% der Fälle Enterobakteriazeen. Bis auf einen Stamm waren allein vitro empfindlich gegenüber Mecillinam allein. Es wurden keine Nebenwirkungen festgestellt.
    Notes: Summary Forty hospitalized patients with mostly uncomplicated urinary tract infections received treatment for ten days with either pivmecillinam/pivampicillin or pivmecillinam alone. The bacteriological cure rate immediately after therapy was discontinued was 82% (18/22) in the combination group and 72% (13/18) in the pivmecillinam group. The causative organisms wereEnterobacteriaceae in 89% of the cases. All but one were sensitivein vitro to mecillinam alone. No side-effects were recorded.
