    Notes: Cyclooxygenases-1 and -2 are both expressed in neuronal cells in vivo. In the neuroblastoma cell lines NG108 and N2a, however, only cyclooxygenase-1 was detectable. Differentiation of the cells with retinoic acid increased cyclooxygenase-1 mRNA and protein expression within 24 and 48 h, respectively. A further increase was observed when the cells were concomitantly treated with the glucocorticoid dexamethasone (a 2–3-fold increase compared with retinoic acid alone). In the absence of retinoic acid, dexamethasone only slightly up-regulated cyclooxygenase-1 expression. The inhibitor of protein synthesis cycloheximide abrogated the effect of dexamethasone, indicating the involvement of newly synthesised proteins. Retinoic acid increased the transcription of cyclooxygenase-1 mRNA, determined with a luciferase-coupled promoter construct. Dexamethasone only slightly augmented cyclooxygenase-1-promoter activity but increased cyclooxygenase-1 mRNA stability. Other corticosteroids, hydrocortisone and aldosterone, also up-regulated cyclooxygenase-1 whereas neurosteroids or oestrogen were ineffective. Up-regulation was mediated primarily by the glucocorticoid receptor, because the receptor antagonist RU486 strongly reduced the effects of all corticosteroids. This indicated that in NG108 cells, the mineralocorticoid aldosterone may bind to the glucocorticoid receptor. Treatment of NG108 or N2a cells with corticosteroids did not alter the morphological phenotype obtained during differentiation. We thus show that corticosteroids, which down-regulate cyclooxygenase expression in most cell types, up-regulate cyclooxygenase-1 during neuronal differentiation.

    Notes: Abstract:  Long-term occupational exposure to formaldehyde (FA) increases the risk for nasopharyngeal squamous cell carcinoma. As the skin is also in contact with FA by environmental exposure, we tested the genotoxic properties of appropriate low concentrations (〈100 µM) of FA on cultured keratinocytes and fibroblasts of human skin. The initial DNA damage was assessed by comet assay. The induction of DNA protein crosslinks was measured by the ability of FA to reduce DNA migration induced by methyl-methane-sulfonate. Upon 4 h of exposure to FA, significant (P 〈 0.05) crosslink formations were observed in fibroblasts (50 µM FA) and in keratinocytes (25 µM FA). Upon 8 h of exposure to FA (25 µM FA), significant crosslink formations were observed in both the cell types. FA is known to inhibit different DNA repair pathways. Therefore, we studied the effect of FA on UV-induced repair. Human keratinocytes and fibroblasts exposed to 10 µM FA prior to UV irradiation showed disturbed repair kinetics after UVC and UVB, but not after UVA irradiation. Single-strand breaks (SSBs) derived from nucleotide excision repair disappeared 6 h after solely UVC (3 mJ/cm2) or 3 h solely UVB (30 mJ/cm2) exposure in both the cell types. In the presence of FA, SSBs were still present at these time points containing a reference to a delay in DNA resynthesis/ligation. FA at a concentration not inducing micronuclei (12.5 µM) caused significant increase of UVC-induced (4 mJ/cm2) chromosomal damage. Proliferation of keratinocytes and fibroblasts was in parallel to observed DNA damages. In conclusion, our data suggest that environmental exposure to FA may contribute to UV-induced skin carcinogenesis.

    Notes: Zusammenfassung Operationsziel Deckung eines Weichteildefektes am distalen Unterarm und der Hand durch einen fasziokutanen gestielten Lappen. Indikationen Maximal 15 × 5 cm große primäre oder sekundäre Haut- und Weichteildefekte an der Hand oder am distalen Unterarm. Kontraindikationen Fehlende Arteria interossea posterior. Fehlende Anastomose der Arteria interossea posterior zur Arteria interossea anterior. Defekte von geringer Größe (unter 3 × 3 cm), die einen Wundverschluß durch Mobilisation des umgebenden Gewebes erlauben. Operationstechnik Anzeichnen des Lappens. Die Gefäßachse läuft in einer Linie vom Epicondylus lateralis humeri zum distalen Radioulnargelenk, wobei der Unterarm in Neutralstellung rotiert steht und der Ellenbogen 90° gebeugt ist. Die maximale Lappengröße beträgt 17 cm Länge und 7 cm Breite. Erster Hautschnitt ulnar im Bereich des angezeichneten Lappens auf die Muskelfaszie des Musculus extensor carpi ulnaris. Präparation des Septum intermusculare mit den perforierenden Hautästen der Arteria interossea posterior. Radiale Umschneidung der Hautinsel und Präparation des Septum intermusculare von radial. Achten auf die motorischen Äste des Nervus radialis. Ligieren der Arteria interossea posterior distal der motorischen Äste. Inzision der Haut über dem Gefäßverlauf, Lösen des Stieles durch Koagulation von Seitenästen zur Muskulatur und zum Knochen. Lösen des Septum intermusculare von ulnar. Darstellen der Kommunikation mit den Vasa interossea anteriora. Legen und spannungsfreies Einnähen des Lappens in den entsprechenden Defekt. Knickbildungen des Lappenstieles vermeiden. Verschluß des Hebedefektes direkt oder mit Vollhaut oder Spalthaut. Ergebnisse In dreieinhalb Jahren wurden bei zehn Patienten Weichteildefekte an der Hand mit dem dorsalen Interosseuslappen gedeckt. Die mittlere Zeit bis zum Einheilen des Lappens betrug 15,9 Tage, die mittlere Nachuntersuchungszeit 19,1 Monate. Alle Lappen sind eingeheilt. Als Komplikationen traten bei zwei Patienten vorübergehende Lappenstauungen auf.

    Notes: Abstract A model of a highly idealized spherically symmetric object radiating away its mass with constant luminosity is presented. The body starts at t = −∞ with both infinite mass and radius and contracts to a point at t = 0 without forming an event horizon. Its material particles are moving non-geodesically and shearfree while transporting heat to the surface. Unlike in some radiating star models of a similar type, all physically required conditions are satisfied in this model.

    Notes: Abstract Total knee joint transplantation has been performed in animal models and humans. This study investigates the impact of this operation on knee joint function in a dog model. Therefore, replantation was compared to transplantation during a 6-month follow-up period in four dogs in each group. The peak vertical ground reaction force normalized in all legs undergoing replantation and in two of four after transplantation. A third transplant recipient reduced loading from the 4th month due to a local complication, and the fourth succumbed to sepsis 3 months postoperatively. A weight-bearing index (WBI), defined as loading of the grafted divided by loading of both hindlimbs decreased from 0.48 ± 0.08 preoperatively to 0.13 ± 0.10 by 1 month after replantation and from 0.53 ± 0.07 to 0 after transplantation. After 6 months, weight-bearing of all replant recipients was restored, but reduced in two transplant recipients with graft function. Full recovery after replantation, but impaired function after transplantation, was also reflected in the histological results: normal histological pictures of blood vessels, cartilage, bone and soft tissues were found in all replant recipients, but infiltrative vasculopathy indicating chronic rejection was found in the transplanted joints. The results of this animal study confirm that the procedure can lead to satisfactory functional results but also emphasize the need for perfect control of immunosuppression.
