    Notes: Awareness of crop biosecurity and phytosanitation has been heightened since 9/11 and the unresolved anthrax releases in October 2001. Crops are highly vulnerable to accidental or deliberate introductions of crop pathogens from outside U.S. borders. Strategic thinking about protection against deliberate or accidental release of a plant pathogen is an urgent priority. Rapid detection will be the key to success. This review summarizes recent progress in the development of rapid real-time PCR protocols and evaluates their effectiveness in a proposed nationwide network of diagnostic laboratories that will facilitate rapid diagnostics and improved communication.

    Notes: The 16S–23S rRNA internal transcribed spacer regions (ITS1) from 14 strains of Pseudomonas syringae and P. fluorescens were sequenced. ITS1 exhibited significant sequence variability among different operons within a single genome. From 1 to 4 types of ITS1 were found in individual genomes of the P. syringae and P. fluorescens strains. A total of eight ITS1 types were identified among strains studied. The ITS1 nucleotide sequences consisted of conserved blocks including, among others, a stem-forming region of box B, tRNAIle and tRNAAla genes and several variable blocks. The differences in the variable regions were mostly due to insertions and/or deletions of nucleotide blocks. The intragenomic heterogeneity of ITS1 was brought about by different combinations of variable blocks, which possibly have resulted from recombination and horizontal transfer.

    Notes: Abstract Bacteria associated with phony disease of peach (PDP) and plum leaf scald (PLS) were consistently isolated from diseased trees but not from healthy trees. Colonies of the bacteria grew slowly on PW agar, reaching 0.2 mm to 0.7 mm in diameter in 2 to 3 weeks. The bacteria did not grow on nutrient agar or other general-purpose media. Cells of the bacteria were 0.3 μm to 0.4 μm in width and 2.6 μm to 20.0 μm in length. The topography of the cell walls revealed numerous ridges and furrows. Cells extracted from diseased plants and those from culture gave a strong fluorescence when stained with immunoglobulin G to cells and purified membranes of the bacteria extracted from peach and plum in earlier studies. Immunoglobulin G to cells of the Pierce's disease bacterium from culture also reacted with the bacteria. No discernible differences were observed between strains associated with PDP and PLS in the United States and PLS in Brazil.

    Notes: Abstract Renografin density gradient centrifugation and carboxymethyl (CM)-and diethylaminoethyl (DEAE)-cellulose ion-exchange chromatography were compared for purification of cells of phony peach, rickettsia-like bacterium (RLB) from plant extracts. Centrifugation on 30–40% linear gradients resulted in four bands. Cells of RLB sedimented into the third band located 4.9–5.4 cm below the meniscus. Phase-contrast microscopy, plant peroxidase activity, and viable bacterial assays showed the resulting cells ofRLB to be relatively free of host contaminants. A typical purification from 120 ml of extract containing 7.2×109 RLB cells resulted in the recovery of 7.0×107 RLB cells. Cells ofRLB were not successfully purified byCM andDEAE chromatography. Unitil a medium is available for cultivation of plantRLB, Renografin density gradient centrifugation should be a useful tool for obtaining cells ofRLB for biochemical and serological investigations.
