    Notes: In a patient with contact dermatitis, dexpanthenol was found to be the causative allergen. There was a positive reaction to dexpanthenol on patch testing. Controls did not show any positive reactions to dexpanthenol on patch testing. Additionally, an LTT was performed, Alter pre-incubation with dexpanthenoi-modified microsomes. we observed an increase in lymphocyte proliferation to dexpamhenol, in comparison to dexpamhenol without microsomes. suggesting that microsomes metabolism plays a role in the pathogenesis of dexpanthenol sensitization, because microsomes are known to posses drug metabolizing enzymes such as cytochrome P450.

    Description / Table of Contents: Summary A simple method is proposed for preparing hematologic material for pulse cytophotometry, including 1) treatment for hemolysis of up to 1 ml heparinized blood or aspirated bone marrow in 10 ml of tap water for 20 minutes, 2) storing of the suspension at +6°C after dilution in ethanol (35%), 3) staining of 1 ml of this dilution in 20 ml ethidiumbromide (10 mg/1000 ml saline) for 20 min and measuring at a rate of 500 cells per sec. Disturbing effects by red cells and by clustering of nucleated cells are avoided by this procedure. The registration of DNA-histograms of high accuracy in measurement is possible within 1 hr. The coincidence rate does not exceed 1% of the cells. Resulting from the three years experience in a hematologic laboratory the technic described is very useful especially for the possibilities of pulse cytophotometry in clinical routine and in the control and arrangement of leukemia treatment.
    Notes: Zusammenfassung Aus dreijähriger Erprobung in einem klinisch hämatologischen Labor ergab sich eine einfache Methode der Zellpräparation von Blut und Knochenmark, mit der die Impulszytophotometrie zur Gewinnung von DNS-Histogrammen als Routineverfahren möglichst ausgenutzt wird. Ein Hauptziel der Autoren ist dabei die kurzfristige Kontrolle von zellkinetischen Effekten der Leukämiebehandlung für Wahl und Zeitpunkt der weiteren Therapie. 0,5 bis 1 ml heparinisiertes Venenblut oder Sternalpunktat werden zur Hämolyse für 20 min in 10 ml Leitungswasser suspendiert. Die Probe kann in 35% Äthanol im Kühlschrank aufbewahrt werden. Nach 20 bis 30 min Färbung von 1 ml der Probe in 20 ml Ethidiumbromidlösung (10 mg/l phys. Kochsalzlösung) erfolgt die Messung von 500 Zellen/sec. Einflüsse durch Erythrozyten, Zellverklebungen und eine Koinzidenzrate von mehr als 1% der Zellen werden vermieden. Das DNS-Histogramm 1 h nach Entnahme der Probe vom Patienten ergibt genaue Werte der Anteile von Zellen in Stadien der Proliferation bzw. der S-Phase und (G2+M)-Phase.
