    Description / Table of Contents: Summary The treatment of the keratitis herpetica with 5-Iodo-2-deoxyuridine (IUDR) was not without prejudial effects. Therefore the activities of enzymes of energy producing metabolism in experimental keratitis herpetica of rabbits were investigated. The result was, that the activities of 18 enzymes of glycolysis, citric acid cycle and hexosephosphate shunt in the corneas of rabbits with experimental keratitis herpetica were elevated. After treatment of normal rabbit cornea with IUDR a decrease of each of these enzymes was found. The enzyme activities of the normal rabbit cornea compared with the flow rate of the glyclysis proved, that a decrease of the enzyme activities may be cause of disturbances of energy producing metabolism. The levels of some metabolites of energy producing metabolism in the cornea were not alterated by the treatment with IUDR for a limited time. The cause for the decrease of the enzyme activities may be a block of the synthesis of protein by IUDR. The conclusion from analogy of the clinical results of the herpes therapy with IUDR to the experimental results recommends to make little or no use of IUDR in the therapy of severe keratitis dendritica with defects in the bowmans membrane and infiltration of the adjacent parenchym areas.
    Notes: Zusammenfassung Nebenwirkungen bei der Behandlung der Keratitis dendritica mit 5-Jodurazildesoxyribosid (IUDR) waren der Anlaß, die Aktivitäten von Enzymen des Energie liefernden Stoffwechsels bei der experimentellen Herpeskeratitis von Kaninchen zu untersuchen. Dabei stellte sich heraus, daß die Aktivitäten von insgesamt 18 Enzymen der Glykolyse, des Krebs-Cyclus und des Hexose-Phosphat-Shunt bei der Herpeskeratitis von Kaninchen erhöht waren. Nach IUDR-Behandlung von normaler Hornhaut dagegen wurde eine deutliche Abnahme aller untersuchten Enzyme festgestellt. Vergleiche der Enzymaktivitäten in der normalen Kaninchenhornhaut mit den Flußraten der Glykolyse ließen erkennen, daß eine Verminderung der Enzymaktivitäten zu Störungen des Energiestoffwechsels führen kann. Die Gehalte einiger Metabolite des Energie liefernden Stoffwechsels in der Hronhaut zeigten nach kurzfristiger Einwirkung von IUDR keine Veränderungen. Als Ursache für die Abnahme der Enzymaktivitäten wird die Hemmung der Protein synthese durch IUDR angesehen. Der Analogieschluß von den klinischen Ergebnissen der Herpesbehandlung mit IUDR zu den experimentellen Befunden läßt es angeraten erscheinen, bei der schweren Keratitis dendritica mit Defekten in der Bowmanschen Membran und Infiltration angrenzender Parenchymbezirke das IUDR gar nicht oder nur mit Zurückhaltung anzuwenden.
