    Notes: An improved technique of electron spin echo envelope modulation (ESEEM) spectroscopy using soft pulses is proposed. It is functionally related to the standard three-pulse stimulated echo experiment, but has an improved bandwidth. The general class of soft-ESEEM experiments is analyzed theoretically by using a product operator formalism. The predicted features are verified experimentally, in particular, the lack of blind spots and the observation of high modulation frequencies.

    Notes: A new and inexpensive X-band resonator structure for pulsed electron-spin resonance and electron-nuclear-double-resonance applications is introduced. The resonator consists of a bridged loop structure and is distinguished by a good reproducibility and reliability. It has a high filling factor and an adjustable Q value and is transparent to radio frequency fields with an upper limit 〉100 MHz. The inner diameter of 5 mm allows convenient sample access of standard 4-mm quartz tubes in the temperature range between 4 and 300 K. Numerical computations of the resonant frequencies, the Q values, and the three-dimensional distributions of the electromagnetic fields are presented. They are based on a discretization method for the solution of Maxwell's equations and include all dielectric elements of the resonator structure. The field homogeneity in the sample area and the influence of the dimensions and the thickness of the metallic layers on field intensity, quality factor, and resonant frequency are determined experimentally. The performance of the resonator is demonstrated by a novel field-swept echo-induced electron-spin-resonance experiment with jumping magnetic field vectors.

    Notes: Design and construction of a broadband electron-nuclear double resonance (ENDOR) spectrometer with polarization-modulated radio frequency fields (PM-ENDOR) is reported. With this type of field configuration, each ENDOR resonance line is labeled by additional parameters so that the interpretation of complicated single-crystal ENDOR spectra may be considerably simplified. Details of the electronics and the X-band microwave cavity suitable to generate polarization-modulated rf fields in the frequency range 2–50 MHz are described. The quality of the PM field over the sample volume is investigated. Performance of the spectrometer is exemplified by applications on transition metal complexes.

    Notes: It is demonstrated by theory and experiment that time-domain broadband electron nuclear double resonance (ENDOR) spectroscopy can be performed by using radio frequency chirp pulses. The chirp ENDOR experiment is shown to have a sensitivity comparable to that of established pulsed ENDOR experiments while featuring at the same time a higher resolution. Two pulsed schemes are proposed for chirp ENDOR that are related to the Davies- and Mims-ENDOR experiments. A simple two-step phase cycle can remove the contributions from unwanted coherence transfer pathways and, unlike in time-domain ENDOR with pulses of fixed radio frequency, no quadrature readout is needed. The basic chirp ENDOR experiment can easily be extended by applying concepts from electron spin echo envelope modulation and nuclear magnetic resonance spectroscopy. A novel hyperfine sublevel correlation (HYSCORE) experiment based on chirp ENDOR is proposed. © 1995 American Institute of Physics.

    Notes: Three-pulse radio-frequency (rf) driven electron spin echo envelope modulation (ESEEM) is introduced as a method to measure hyperfine couplings of electron–nuclear spin systems in liquids. It is distinguished from the corresponding two-pulse experiment by narrower lines. Numerical simulations of rf-driven ESEEM based on Floquet theory are given and compared with average Hamiltonian theory. Artifacts which appear in rf-driven ESEEM spectra are discussed, and approaches for the removal of these artifacts such as time-proportional phase incrementation (TPPI) of the rf phase and an rf phase cycle (0, π) are presented. The validity of the analytical description of the rf-driven ESEEM experiment is shown by comparison with experimental results. © 2001 American Institute of Physics.

    Notes: Investigations were carried out on changes of lactate concentration and pH values at various post-mortem times on different samples of beef and pork muscles (Expts. A and B). The differences in lactate concentrations between the method of Barker and Summerson and the enzymatic assay were considerable, fully confirming previous findings. Besides the lactate and pH values, glucose, fructose, maltose, and ribose were determined after feeding sucrose to some of the animals (Expt. C). Sucrose feeding decreased the pH values and increased the values for lactate in investigated muscles of exhausted animals which had been transported over 340 miles in approximately 24 hr. By this procedure some characteristics of the quality of the meat could he improved. Similar effects were not detected in animals transported only 3.1 miles, although glucose and fructose were present in significantly higher amounts in the sucrose-fed animals than in the control group.

    Notes: Two methods of determining lactic acid in mammalian tissues are compared. Lactic acid was estimated by the procedure of Barker and Summerson and by the enzymatic test with lactic dehydrogenase. The standard error of the mean at the 5% level was 5.8% for the former and 3.1% for the latter. Values for lactic acid found by both methods did not agree in most cases. Differences of over 40% were detected. After chromatographic purification of the extracted lactic acid, the Barker and Summerson values were identical with the enzymatic values. Lactic acid isolated by thin-layer chromatography was identified by its infrared spectrum as the p-phenyl-phenacylester. The enzymatic test should be regarded as the more reliable procedure for determining lactic acid in meat.
