    Notes: Summary The fluorochrome, p-hydrazinoacridine (HA), is used to stain selectively natural macromolecular aldehydes as occur in elastic tissue as well as periodic acid (oxidized) or hydrochloric acid (hydrolyzed) engendered polyaldehydes (e.g., respectively of glycogen, mucosubstances and deoxyribonucleic acid). The reaction is independent of SO2 required in Schiff-type reactions. The mechanism of the staining reaction is presumably via the formation of fluorescent hydrazones from aldehyde condensation with HA. Aldehyde blocking agents, i.e., thiosemicarbazide and sodium borohydride, block the staining reactions with HA. In view of the controversy surrounding the mechanism of Schiff-type reactions and the lack of an adequate explanation for the affinity of miscellaneous fluorochromes for elastic tissue, p-hydrazinoacridine appears to be an ideal reagent, combining a simple staining procedure, excellent histological results and a straightforward mechanism for its selectivity.

    Notes: Summary The adventitial cells surrounding the spermatheca of the reproductive system of Sonorella santaritana (Mollusca: Gastropoda) appear to have an unusual system of vesicles. Electron micrographs of the membranes forming these vesicles show that they have multiple openings to the cell's exterior and that each opening has a “pore complex”. In addition, secondary vesicles appear to be generated by the primary vesicles. Evidence is presented suggesting that these vesicles represent a previously unreported membrane transport system.

    Notes: Summary VIP-containing neurons were localized in lungs from dogs, cats, and human subjects by means of the indirect immunofluorescence technique. Nerve fibers and terminals were observed in the smooth muscle layer and glands of airways, and within the walls of pulmonary and bronchial vessels, especially at the medial-adventitial junction. VIP-positive nerve cell bodies were identified in ganglia located in the walls of bronchi. These findings provide an anatomic basis for the possible modulation of airway and pulmonary vascular function by this neuropeptide.

    Notes: The ultrastructure of the spermatheca of the reproductive tract in the pulmonate snail, Sonorella santaritana, was investigated. This organ has a debris-filled lumen and an outer wall which can be divided into three distinct layers. The cell layer adjacent to the lumen is comprised of two cell types, tall columnar epithelial cells with microvilli and cells lacking microvilli. The next layer also has two cell types, muscle cells and apparent pigment cells. The most distant layer is an adventitia of large glycogen-containing cells. The lumen of the spermatheca contains a core of partially digested sperm and related materials.The luminal contents and the cellular morphology of this organ suggest that the spermathecal functions are both digestive and absorptive. It is proposed that excess sperm and related materials are transported to the spermatheca, digested, and the usable products are reabsorbed.

    Notes: TAG fixation of normal and Ca2+ ionophore-treated rabbit polymorphonuclear leukocytes (PMN) has revealed membrane components not apparent with conventional glutaraldehyde fixation. These included a 30 Å external electron-dense coating on untreated cells. A somewhat thicker coat (40 Å) was observed in ionophore-treated, nondegranulating PMN. In ionophore-treated, degranulating PMN, a 65 Å cell membrane coat was observed. A similar coat was observed on the inner side of the membrane of some azurophil-type granules, but the electron density and thickness were not so pronounced. Cytoplasmic granules were often closely apposed and often protruded outward at the plasma membrane. Extracellular lamellae, sometimes stacked apposed to the plasma membrane, possibly represent remnants of intense granule extrusion. Sequential degranulation of the respective granules was not apparent.
