    Notes: Abstract Bisphosphonates are being used in disorders associated with accelerated resorption of bone, particularly Paget's disease of bone and the bone disease of malignancy. Their undoubted biological efficacy and relatively low apparent toxicity make them attractive candidates for the management of osteoporosis. The bisphosphonate alendronate has many characteristics which suggest that it is suitable for use in osteoporosis. It is a potent inhibitor of osteoclast-mediated bone resorption with no adverse effect on the mineralization of bone. Earlier studies have shown it to be one of the most active bisphosphonates in Paget's disease and the hypercalcemia of malignancy. In common with other bisphosphonates tested thus far, alendronate appears to inhibit bone loss in a variety of experimental models of osteoporosis. Long-term studies are needed to determine its steady-state effects on bone mass in man. Most data indicate that alendronate is capable at least of decreasing the rate of bone loss, and might even induce increments in bone mass for many years. Since the experimental studies show that the increase in bone mass observed with alendronate is associated with an increase in bone strength, its use is likely to decrease the frequency of fractures. However, direct clinical evidence for this requires the outcome of well-designed long-term prospective studies.

    Notes: Summary Cells obtained from surgical bone specimens of eight patients with Paget’s disease of bone were maintained in culture for up to 8 months and seven passages. The doubling time during the period of maximal cell growth ranged from 4 to 12 days. Evidence consistent with the hypothesis that many of the cells were bone cells included the following: (a) histochemical techniques demonstrated staining of some cells for alkaline phosphatase or acid phosphatase and succinic dehydrogenase; (b) parathyroid extract stimulated increased uptake of3H-thymidine and3H-uridine; (c) parathyroid extract suppressed and salmon calcitonin stimulated uptake of3H-proline; and (d) crystalline calcium deposits were found within cells and extracellularly. Ultrastructural analysis revealed that three of the eight cultures contained cells whose nuclei had inclusions which were almost identical to those found in the osteoclast nuclei of all patients with Paget’s disease. The maintenance of cells derived from pagetic bone in long-term culture should aid in testing the hypothesis that Paget’s disease represents a slow virus infection of bone.

    Description / Table of Contents: Zusammenfassung Es wurden die täglichen Immunglobulinveränderungen bei nierentransplantierten Patienten in den ersten drei Wochen nach der Transplantation untersucht. Entgegen den Befunden anderer Autoren an nicht splenektomierten Patienten, wurde bei unseren durchwegs splenektomierten Patienten, die sehr niedere IgM Spiegeln aufwiesen, nur selten ein IgM Abfall während einer Abstoßungskrise beobachtet. Wir fanden vielmehr, daß in rund der Hälfte der klinisch diagnostizierten Abstoßungskrisen kurzfristige Immunglobulingipfel auftreten. Sie wurden immer 1–6 Tage vor oder gleichzeitig mit der klinischen Diagnose einer Abstoßungskrise festgestellt. Eine statistische Wahrscheinlichkeitsprüfung mit dem generell einseitigen Binomialtest zeigte eine signifikante Beziehung zwischen IgM Gipfeln und zu erwartenden Abstoßungskrisen, aber keinen solchen Zusammenhang für IgA Gipfel. Für die Immunglobuline IgG konnte keine Korrelation zu den klinischen Abstoßungskrisen gefunden werden. In Übereinstimmung mit anderen Autoren fanden wir ebenfalls einen deutlichen Abfall dieser Gammaglobulinfraktion unter der Initialdosierung von I g Methylprednisolon und gleichzeitiger Azathioprin-Medikation. Interessant erwies sich der hohe korrelative Zusammenhang zwischen der IgG und Serumalbuminverminderung nach der Transplantation. Die von uns ebenfalls gefundenen geringfügigen Proteinverluste mit dem Urin bestätigen die Ansichten anderer Arbeitsgruppen, daß für das auffallend starke Absinken der IgG Fraktion und des Serumalbumins in erster Linie ein gesteigerter Katabolismus bzw. eine Hemmung der Proteinsynthese angenommen werden muß.
    Notes: Summary In the first three weeks after kidney transplantation the patients were examined for daily changes in immunoglobulin levels. Only very seldom was a decrease of IgM detected in our patients who had undergone splenectomy. During half of the rejection crisis there were indeed short periods of increase (spikes) in immunoglobulins. These were observed always before or concomitantly with the clinical diagnosis of a rejection reaction. In order to determine the connection between immunoglobulin-spikes and an approaching rejection episode the general one-sided binominal test was used. A significant relation was established between IgM peaks and an approaching rejection crisis. No such relation could be found for the IgA peaks observed. The IgG globulins showed no consistent pattern during a transplantation rejection. A persitent fall of the IgG globulins was noted in the first ten days after renal allograft with the immunosuppressive regimen of an initial gramme of methylprednisolon and high dosage of azothioprine. A good correlation was observed with the simultaneous depression of serum albumin concentrations. As losses of protein in urine are minimal (already confirmed by other authors), it must be assumed that in the first place a high rate of catabolism and a restriction in the protein synthesis is responsible for the decrease in the IgG globulins and the serum albumins.

    Notes: Abstract Field studies of the desert spider Agelenopsis aperta revealed a primarily monogamous mating system. However polygyny, polyandry and polygynandry were superimposed upon the primary system, with 9% of the marked males and 11% of the marked females in a field population mating more than once. In the laboratory males commonly mated multiply with fertile offspring resulting, while females were less likely than males to mate multiply. Monogamy under field conditions was enforced by two factors: (1) high travel costs to males, and (2) a significant decline in female receptivity after the first mating. Heavy males were more likely to be accepted by females both in the field, and in female choice experiments conducted in the laboratory. Finally, male weight determined the outcome of male-male agonistic interactions over females. One possible explanation for female choice in this system which lacks male parental investment is that females may be using male size as an indicator of future success of their offspring.
