    Notes: DL-serine, DL-methionine or DL-serine + DL-methionine in excess inhibited the growth of Tetrahymena pyriformis H. Excess serine was most inhibitory at high concentration of folic acid, whereas the effect of excess methionine or methionine + serine was most pronounced at low levels of folic acid. Inhibition due to excess serine was relieved by raising the level of methionine or by adding pyrimethamine to lower the effective folic acid level, and was intensified by adding Dl.-ethionine or by raising the level of folic acid. Similarly, inhibition due to excess methionine was relieved by supplying more serine or adding DL-ethionine (which reduced the amount of available methionine) and was intensified by adding pyrimethamine. Inhibition by excess methionine + serine was reversed by increasing threonine, provided there was ample guanine present. Low levels of guanine or the presence of 8-azaguanine prevented this reversal. Comparisons are made with the work of others.

    Notes: Abstract Background: The determination of sarcoma grade, histologic type, and differentiation is often pathologist dependent and requires considerable expertise. Methods: Lipid content and composition was analyzed in ex vivo fat, lipoma, and liposarcoma tissue samples using proton-decoupled13C nuclear magnetic resonance (13C-NMR) spectroscopy and correlated with the histologic type and grade of liposarcoma. Results: The well-differentiated liposarcomas were found to have threefold increases in fatty acyl chain content compared with benign lipomas. The fatty acyl chain content of the dedifferentiated and pleomorphic liposarcomas was 1% of that found in lipoma and〈0.2% of that found in well-differentiated liposarcoma. The 2.1- to 2.8-fold increase in the degree of polyunsaturation in the dedifferentiated and pleomorphic liposarcomas compared with well-differentiated liposarcoma could largely be accounted for by the 2.3-fold increase in the percentage of fatty acyl chains of lipid containing linoleic acid. The dedifferentiated and pleomorphic liposarcomas contained both free fatty acids and phospholipids that were not NMR detectable in normal fat, lipoma, and well-differentiated liposarcoma. Conclusion: Ex vivo13C-NMR spectroscopy may be used to distinguish lipoma from well-differentiated, dedifferentiated, and pleomorphic liposarcoma based on changes in lipid and phospholipid metabolite profiles and may serve as adjunct to conventional light microscopy for the determination of liposarcoma histologic type and thus grade.

    Notes: Summary Single and multiple antibiotic-resistant strains were isolated from four insecticidal and two noninsecticidal parent strains ofBacillus sphaericus. Several of the single resistant strains isolated were also cross-resistant to several antibiotics of different modes of action. All of the parent strains were naturally susceptible to 11 and naturally resistant to two out of 22 antimicrobics examined. The majority, 17 out of 23, of antibiotic-resistant strains, that were isolated from the insecticidal parent strains, retained their insecticidal activity while the noninsecticidal strains remained noninsecticidal. Among the isolates were several strains that would be potentially useful as marker strains in the genetic manipulation of the insecticidal strains.

    Notes: Summary Strain SS86-4 was one of 40Bacillus brevis strains shown to be molluscicidal to the schistosomiasis snail vectorBiomphalaria glabrata. When grown in mB4 medium in 2-L fermentors, SS86-4 was molluscicidal only if fructose or phenylalanine was present in the medium. This is reminiscent of secondary fermentation factor effects, in this case an antioxidant effect. In vivo proteases also were capable of reducing molluscicidal activity. The molluscicidal toxin has an LC50 of 1 μg toxin protein ml−1 (approx. 1 p.p.m.) and may be described as a small proteinaceous, heat-stable, oxygen-sensitive entity associated with the particulate portion of the cell wall fraction ofB. brevis that is formed prior to sporulation. Initial information indicates that its HPLC signature shows major peaks at 148.37 and 163.96 s and consists of two bands of approximately 5.3 kDa and 8.7 kDa on PAGE gel.

    Notes: Summary Analysis of aminopeptidase activity in commercially available enzyme panels (SAP-II and SAP-III) indicated qualitative differences among the eight strains ofBacillus sphaericus tested. These differences involved specific substrates utilized, the importance of sampling time, as well as differences between whole cells and cell-free extracts. Further detailed analysis of four of thel-amino acid-β-naphthylamide substrates using polyacrylamide gel electrophoresis (PAGE) demonstrated a series of allelic differences in the multilocus patterns observed. The resulting phenogram constructed from the enzyme and PAGE information showed a marked separation of the noninsecticidal strains from the insecticidal strains (58% similarity). In addition, there was a separation within the insecticidal strains of strain SSII-1, phage type 2 from strain 2297, phage type 4, and of both of these strains from the remaining highly insecticidal members of phage type 3 (strains 1593, 2013-4, 2362, 2500).
