Abstract: Brain DM-20 proteolipid was previously shown to be structurally different from the myelin major proteo-lipid (MMPL). In an attempt to set up a large-scale purification of DM-20, we studied extraction of brain proteo-lipids with mixtures of methylene chloride containing up to 80% methanol. The CH2Cl2-CH3OH (3:7, vol/vol) mixture is highly selective for the extraction of bovine brain DM-20 compared to MMPL. Purified DM-20 was obtained after chromatography of the extract on methylated Sephadex.
