    Notes: Thymocytes were removed from mice at different times before and after birth, and their phenotype and killing repertoire were analysed after stimulation with human recombinant interleukin 2 (IL-2). Whereas three different tumour targets were killed by all lymphokine-activated killer (LAK) populations tested. EL4 lymphoma cells were only killed by LAK cells derived from thymocytes after, but not before, birth. Cold-target competition tests showed that LAK thymocytes recognized antigenic structures on EL4 cells that arc different from those on other tumour targets. Analysis of EL4 target killing after removal of different subsets from LAK cells revealed that the major part of this killing is exerted by CD8+ cells. Our findings are additional and more direct evidence for partial target specificity in LAK cells.

    Notes: The lactate dehydrogenase isoenzyme pattern has been determined in different murine thymocyte cell populations. Enrichment of thymocytes with more mature cells through total body irradiation results in a smaller percentage of LDH-1, LDH-2, LDH-3 and a greater percentage of LDH-4 and LDH-5. Fractionation of normal thymocytes by velocity sedimentation at unit gravity yields a fraction of cells with a high sedimentation rate. LDH-1 and LDH-2 formed a smaller percentage of the total enzyme activity in these cells. These findings indicate that the LDH isoenzyme distribution is a marker for differentiation of thymocytes.

    Notes: The aim of this study was to examine the cytokine production and cytokine responsiveness of the first T-cell receptor (TcR) positive cells that appear in the murine fetal thymus, namely TcR Vγ3 cells. It is shown that IL-2-cultured fetal TcR Vγ3 thymocytes were capable of producing IL-3, GM-CSF, TNF-α and IFN-γ upon TcR triggering. IL-2, IL-4, IL-5 and IL-6 could not be detected. With regard to cytokine responsiveness, TcR Vγ3 cells proliferated to a high extent when high concentrations of rIL-2 were added. rIL-4or rIL-7 alone, but not rIL-1 alone, were capable of inducing a modest proliferation of TcR Vγ3 thymocytes. When combined with low concentrations of IL-2, a synergistic effect could be observed with IL-1, IL-4 or IL-7. It is shown that the synergistic effect of IL-2 with IL-4 was mainly due to induction of IL-2 receptor expression. The synergistic effect of IL-2 and IL-7 on the proliferation of TcR Vγ3 cells could only be partially inhibited by anti-IL-2 receptor MoAb, and this antibody had no effcet on the IL-2 + IL-1 cultures. These observations can explain the extensive proliferation of TcR Vγ3 thymocytes during fetal life and they indicate that TcR Vγ3 thymocytes have the potential to play a functional role during fetal thymus development.

    Notes: Autologous rosette-forming cells (ARFC) have been considered to be post-thymic precursor cells. Since thymocytes and peripheral T lymphocytes differ considerably in lactate dehydrogenase isoenzyme pattern and in activity of the enzymes of the purine metabolism, we investigated the enzyme profile in the ARFC. The L-lactate:NAD+ oxi-reductase analysis showed an isoenzyme pattern that closely resembled the pattern found in peripheral T lymphocytes and was totally different from the thymocytes. The levels of adenosine deaminase and purine nucleoside phosphorylase were identical to those found in the peripheral T lymphocytes and different from thymocytes. In our hands, the ARFC-enriched suspension contained predominantly OKT4+ and Tμ+ lymphocytes. We propose that ARFC are a heterogeneous population encompassing all known subsets and cannot be considered a separate homogeneous entity.

    Notes: Summary In 16 patients with B-type CLL, the T and B cell compartment was analysed using the monoclonal anti-T-cell antisera OKT1, 3, 4 and 8 and the lactic dehydrogenase enzyme marker. Pertinent findings were: the presence on the CLL cells in 15 out of 16 patients of the antigen determined by the OKT1 antiserum, and, in all patients, a lower total LDH content on a per cell basis combined with a higher percentage-activity in the LDH-3 band as compared to the normal B-cell. Furthermore, the T-cell compartment was also disturbed in these patients, as the ratio of OKT4+ to OKT8+ cells was significantly depressed accompanied by a significant decrease of the LDH-1 percentage-activity in favour of LDH-3 and 4. These findings argue for the B-cell being immature and confirm the recent evidence that the T-cell compartment is changed in B-CLL.

    Notes: Summary The β-glucuronidase staining characteristics of isolated T cell populations and the Tμ and Tγ enriched fractions derived of them were studied. Tγ lymphocytes were obtained from purified T lymphocytes by ox-IgG rosette sedimentation. The rosette-forming cells in the pellet were referred to as Tγ lymphocytes, whereas the lymphocytes in the interface were referred to as Tγ depleted or Tμ lymphocytes. B cells were studied on rosetted mononuclear cells with either mouse erythrocytes or with Staphylococcus Aureus (Cowan I) bacteria, after a preceeding polyvalent anti-human Ig treatment of the cells. While B cells showed mostly no reactivity, Tμ and Tγ cells were respectively characterised by a “dot-like” and “granular” pattern of reactivity. These findings are in agreement with those observed by others after α-naphthyl-acetate esterase or acid phosphatase staining. Within the T lymphocyte fraction, the T non-μ, nonγ lymphocytes seemed to have a granular pattern of reactivity. The same staining pattern was found in non-B, non-T lymphocytes.
